The role of integrins, the cell-surface glycoproteins involved in various cellular functions, is well documented. However, information about their role and expression profile during pregnancy is still scant. In the present study, the expression of the integrin subunits β3, α6 and α5, along with their ligands vitronectin, osteopontin, laminin and fibronectin, was investigated in mouse uterus during different stages of pregnancy, namely 6.5, 8.5 and 13.5 days post coitus (d.p.c.) by immunohistochemical localisation. Integrins β3, α6 and α5 and the extracellular matrix molecules vitronectin and osteopontin exhibited dynamic spatiotemporal changes in their expression pattern in gestational endometrium, whereas fibronectin and laminin demonstrated more-or-less ubiquitous expression. The inter-implantation sites showed localisation of most of these molecules predominantly in the luminal epithelium, whereas their expression was negligible in the stroma. The present study explores the possible role and relevance of the spatiotemporal expression of integrins and their ligands in endometrial/decidual function and the maintenance of pregnancy.

We examine roles of signaling by secreted ligands of the TGF-beta family during Drosophila oogenesis. One family member, the DPP ligand encoded by the decapentaplegic (dpp) gene, is required for patterning of anterior eggshell structures. This requirement presumably reflects the expression pattern of dpp in an anterior subset of somatic follicle cells: the centripetally migrating and the nurse cell-associated follicle cells. Similar requirements are also revealed by mutations in the saxophone (sax)-encoded receptor, consistent with the idea that DPP signaling is, at least in part, mediated by the SAX receptor. A loss of germline sax function results in a block in oogenesis associated with egg chamber degeneration and a failure of the transfer of nurse cell contents to the oocyte, indicating that TGF-beta signaling is required for these events. Some phenotypes of sax mutations during oogenesis suggest that SAX responds to at least one other TGF-beta ligand as well in the posterior follicle cells.

Embryo implantation and decidualization are key steps for successful reproduction. Although numerous factors have been identified to be involved in embryo implantation and decidualization, the mechanisms underlying these processes are still unclear. Based on our preliminary data, Prss56, a trypsin-like serine protease, is strongly expressed at implantation site in mouse uterus. However, the expression, regulation and function of Prss56 during early pregnancy are still unknown. In mouse uterus, Prss56 is strongly expressed in the subluminal stromal cells at implantation site on day 5 of pregnancy compared to inter-implantation site. Under delayed implantation, Prss56 expression is undetected. After delayed implantation is activated by estrogen, Prss56 is obviously induced at implantation site. Under artificial decidualization, Prss56 signal is seen at the primary decidual zone at the initial stage of artificial decidualization. When stromal cells are induced for in vitro decidualization, Prss56 expression is significantly elevated. Dtprp expression under in vitro decidualization is suppressed by Prss56 siRNA. In cultured stromal cells, HB-EGF markedly stimulates Prss56 expression through EGFR/ERK pathway. Based on promoter analysis, we also showed that Egr2 is involved in Prss56 regulation by HB-EGF. Collectively, Prss56 expression at implantation site is modulated by HB-EGF/EGFR/ERK signaling pathway and involved in mouse decidualization.
