Three groups of seedlings of snap bean cultivars Contender and Conquest were planted in Coto clay, Fraternidad silty clay, and San Antón silt loam; inoculated with 10,000 eggs and second stage juveniles of Meloidogyne incognita and placed in growth chambers at 15, 20 and 25° C for 45 days. Multinucleate giant cells with thick cell walls and dense cytoplasm surrounded by proliferation of small cells, breakdown of cell walls, and malformation of vascular bundles were observed. No giant cells were observed in bacterial nodules, even though several of them were invaded by nematodes. Cytoplasmic content of the giant cells decreased as temperature increased. Nematode numbers within roots increased as the temperature increased. There was a tendency for the formation of oval or rectangular giant cells around the vascular bundles and round giant cells in the cortex. Giant cells with the largest number of nuclei were observed at 20° C, and fewer at 25° C. Giant cell was largest at 20 and 25° C, especially in cv Contender. Necrosis or mechanic injury due to migration of juveniles was not observed. The nematodes appeared to develop best in Fraternidad soil.

Meloidogyne incognita is a root knot nematode (RKN) species which is among the most notoriously unmanageable crop pests with a wide host range. It inhabits plants and induces unique feeding site structures within host roots, known as giant cells (GCs). The cell walls of the GCs undergo the process of both thickening and loosening to allow expansion and finally support nutrient uptake by the nematode. In this study, a comparative in situ analysis of cell wall polysaccharides in the GCs of wild-type Col-0 and the microtubule-defective fra2 katanin mutant, both infected with M. incognita has been carried out. The fra2 mutant had an increased infection rate. Moreover, fra2 roots exhibited a differential pectin and hemicellulose distribution when compared to Col-0 probably mirroring the fra2 root developmental defects. Features of fra2 GC walls include the presence of high-esterified pectic homogalacturonan and pectic arabinan, possibly to compensate for the reduced levels of callose, which was omnipresent in GCs of Col-0. Katanin severing of microtubules seems important in plant defense against M. incognita, with the nematode, however, to be nonchalant about this “katanin deficiency” and eventually induce the necessary GC cell wall modifications to establish a feeding site.

A 55-year-old man with mental retardation and calcaneal tendon thickening was referred for a suspected genetic disease. His serum cholestanol was elevated and genetic analysis of his blood cells for CYP27A1 revealed a homozygous missense mutation. We diagnosed him with cerebrotendinous xanthomatosis (CTX). Chest radiography revealed diffuse micronodular and reticular opacities. Histological findings obtained from the transbronchial lung biopsy revealed foamy macrophages and multinucleate giant cells with marked lipid crystal clefts. Although there are few reports of pulmonary lesions in CTX, we concluded from the radiological and histopathological findings that the pulmonary lesions were indeed caused by the CTX. The patient was treated with chenodeoxycholic acid. His neurological findings and calcaneal tendon thickening were unchanged; however, his serum cholestanol and radiological abnormalities of the chest decreased.

Ten 7-day-old gnotobiotic Beagle puppies were inoculated intraperitoneally with virulent canine distemper virus (R252-CDV). The dogs were killed and perfused with paraformaldehyde/glutaraldehyde from eight to 36 days after inoculation. The developing teeth of the mandibles were examined by light microscopy, and the teeth from three dogs were examined by electron microscopy. Necrosis of individual cells in the stratum intermedium of the developing tooth was the first change, detectable at day 9 post-inoculation. At day 16 post-inoculation, there was disorganization of the ameloblasts. In the stratum intermedium, multinucleate giant cells and large eosinophilic cytoplasmic viral inclusions were prominent. Ultrastructurally, these inclusions consisted of clusters of tubular aggregates typical of canine distemper virus nucleocapsids. At 28 to 36 days post-inoculation, the changes were seen in the reduced enamel epithelium. Multinucleate cells were seen, but no inclusions. Some necrotic cells were seen. In these teeth, ameloblastic cells of the root were morphologically normal. Our results suggest that distemper virus affects developing teeth by direct infection of the enamel organ.

Aqueous leaf extracts were utilized to assess the nematicidal or nematostatic property on second stage juvenile of Meloidogyne incognita. The juvenile were incubate at various concentration of leaf extract viz., 250, 500, 1000 and 2000 ppm. Corrected mortality using Abbot’s formula was recorded after 12, 24 and 48 hours respectively. Correlation coefficient (Pearson) was checked to explain the association between percentages mortality of juvenile with extract concentrations. Linear regression was used to denote concentration and rank dependent outcome of four aqueous plant leaves extracts on the second stage juvenile (J2) mortality. All leaf extracts were found to be nematicidal or nematostatic in property. Maximum juvenile mortality rate was recorded in Xanthium strumarium throughout the incubation period as followed by Acalypha indica, Argemone mexicana and Colocasia gigantean. Concentration depended effect of X. strumarium and C. gigantean proved maximum and minimum level when analyzed by values of regression and correlation. Aqueous leaves extracts of these aforementioned weeds give us an idea about nematicidal properties and therefore may be used as biopesticide in future

Masses removed from the superficial fascia of the jugular groove of a 12-year-old Arabian mare and from the femoral canal of a 7 1/2-year-old female cat appeared to be counterparts of the human malignant giant cell tumor of soft parts, even recapitulating the clinical behavior of the respective subgroups. Histologically, both neoplasms contained the characteristic features of the malignant giant cell tumor of soft parts: large multinucleate giant cells, pleomorphic mononuclear giant cells, histiocytes, fibroblasts and fibrocytes that were sometimes sarcomatous, and foci of hemorrhage and necrosis. Ultrastructural characteristics of the giant cells suggest mesenchymal origin with osteoblastic or chondroblastic differentiation.
