More than three decades of research efforts have yielded powerful methodologies based on transition metal template-directed syntheses for the assembly of a huge number of interlocked systems, molecular knots, machines and synthesizers. Such template techniques have been applied in the preparation of mechanically linked electron donor–acceptor artificial photosynthetic models. Consequently, synthetic challenging photoactive rotaxanes and catenanes have been reported, in which the chromophores are not covalently linked but are still associated with undergoing sequential energy (EnT) and electron transfer (ET) processes upon photoexcitation. Many interlocked photosynthetic models produce highly energetic, but still long-living charge separated states (CSS). The present work describes in a historical perspective some key advances in the field of photoactive interlocked systems assembled by transition metal template techniques, which illustrate the usefulness of rotaxanes and catenanes as molecular scaffolds to organize electron donor–acceptor groups. The effects of molecular dynamics, molecular topology, as well as the role of the transition metal ion used as template species, on the thermodynamic and kinetic parameters of the photoinduced energy and electron transfer processes in the interlocked systems are also discussed.

The retina and, in particular, retinal pigment epithelial cells are unusual for being encumbered by exposure to visible light, while being oxygen-rich, and also amassing photoreactive molecules. These fluorophores (bisretinoids) are generated as a byproduct of the activity of vitamin A aldehyde—the chromophore necessary for vision. Bisretinoids form in photoreceptor cells due to random reactions of two molecules of vitamin A aldehyde with phosphatidylethanolamine; bisretinoids are subsequently transferred to retinal pigment epithelial (RPE) cells, where they accumulate in the lysosomal compartment with age. Bisretinoids can generate reactive oxygen species by both energy and electron transfer, and they become photo-oxidized and photolyzed in the process. While these fluorescent molecules are accrued by RPE cells of all healthy eyes, they are also implicated in retinal disease.

Multiscale models combining quantum mechanical and classical descriptions are a very popular strategy to simulate properties and processes of complex systems. Many alternative formulations have been developed, and they are now available in all of the most widely used quantum chemistry packages. Their application to the study of light-driven processes, however, is more recent, and some methodological and numerical problems have yet to be solved. This is especially the case for the polarizable formulation of these models, the recent advances in which we review here. Specifically, we identify and describe the most important specificities that the polarizable formulation introduces into both the simulation of excited-state dynamics and the modeling of excitation energy and electron transfer processes.
