AbstractCurrently, in vitro embryo production (IVP) is successfully commercially applied in cattle. However, the high sensitivity of embryos to cryopreservation in comparison to in vivo (IVD) embryos slows the dissemination of this biotechnology. Reduced cryotolerance is frequently associated with lipid accumulation in the cytoplasm mainly due to in vitro culture conditions. The objective of this study was to evaluate the lipid composition of biopsied and sexed embryos, produced either in vivo or in vitro from the same Holstein heifers before and after a slow freezing protocol. Lipid extracts were analysed by liquid chromatography-high resolution mass spectrometry, which enabled the detection of 496 features. Our results highlighted a lipid enrichment of IVP embryos in triglycerides and oxidised glycerophospholipids and a reduced abundance in glycerophospholipids. The slow freezing process affected the lipid profiles of IVP and IVD embryos similarly. Lysophosphatidylcholine content was reduced when embryos were frozen/thawed. In conclusion, the embryonic lipid profile is impacted by IVP and slow freezing protocols but not by sex. Lysophosphatidylcholine seemed highly sensitive to cryopreservation and might contribute to explain the lower quality of frozen embryos. Further studies are required to improve embryo freezability by modulating the lipidome.

Abstract Summary Here we introduce Lipid Mini-On, an open-source tool that performs lipid enrichment analyses and visualizations of lipidomics data. Lipid Mini-On uses a text-mining process to bin individual lipid names into multiple lipid ontology groups based on the classification (e.g. LipidMaps) and other characteristics, such as chain length. Lipid Mini-On provides users with the capability to conduct enrichment analysis of the lipid ontology terms using a Shiny app with options of five statistical approaches. Lipid classes can be added to customize the user’s database and remain updated as new lipid classes are discovered. Visualization of results is available for all classification options (e.g. lipid subclass and individual fatty acid chains). Results are also visualized through an editable network of relationships between the individual lipids and their associated lipid ontology terms. The utility of the tool is demonstrated using biological (e.g. human lung endothelial cells) and environmental (e.g. peat soil) samples. This study was conducted to determine the optimum feed selection for the spiny lebbeid shrimp, Lebbeus groenlandicus, from zoea to megalopa larva. To the shrimp zoeae, untreated hatching Artemia meta-nauplii, or either of two kinds of enriched Artemia, were supplied as feed daily, in a concentration of 3 individuals/ml. Throughout the experimental trials, 17.6-18.1 days elapsed until reaching the megalopa larval stage. In the starving trials, no mortality was observed; the mortality values observed in the experimental trials, between 4.8 and 9.5% were considered low and not significantly different (). Neither fatty nor amino acids varied significantly in the zoea I and II stages in either the feeding or starving trials. Highly unsaturated fatty acids and amino acids were significantly lower in the megalopa stage in the starving group, compared with the feeding groups (). Therefore, feeding was proven not to have any effect in the zoea I and II larval stages. However, for the megalopa larval stage, a feed containing supplementary amino acids and fatty acids evoked a positive effect.
