COVID-19 presentations range from mild to moderate through severe disease but also manifest with persistent illness or viral recrudescence. We hypothesized that the spectrum of COVID-19 disease manifestations was a consequence of SARS-CoV-2-mediated delay in the pathogen-associated molecular pattern (PAMP) response, including dampened type I interferon signaling, thereby shifting the balance of the immune response to be dominated by damage-associated molecular pattern (DAMP) signaling. To test the hypothesis, we constructed a parsimonious mechanistic mathematical model. After calibration of the model for initial viral load and then by varying a few key parameters, we show that the core model generates four distinct viral load, immune response and associated disease trajectories termed “patient archetypes”, whose temporal dynamics are reflected in clinical data from hospitalized COVID-19 patients. The model also accounts for responses to corticosteroid therapy and predicts that vaccine-induced neutralizing antibodies and cellular memory will be protective, including from severe COVID-19 disease. This generalizable modeling framework could be used to analyze protective and pathogenic immune responses to diverse viral infections.

To solve the mobile robot path planning in uncertain environments, a new path planning algorithm is presented on the basis of the biological immune network. The environment surrounding the robot is taken as the antigen, and the behavior strategy of robot is taken as the antibody. The selection model of antibody concentration is defined based on the Jernes idiotypic immune network hypothesis, and the mobile robot path planning is realized through the selection of the antibody concentration. The simulation of path planning for mobile robot in multi-obstacle environments shows that the robot can find a safe path in complicated environments, which verifies the better adaptivity of proposed planning model. The simulation in dynamic environments shows that the robot can safely avoid all dynamic obstacles, which verifies the better flexility of new algorithm.

This chapter shows that some of the basic aspects of the natural immune system discussed in the previous chapter can be used to propose a novel artificial immune network model with the main goals of clustering and filtering crude data sets described by high-dimensional samples. Our aim is not to reproduce with confidence any immune phenomenon, but demonstrate that immune concepts can be used as inspiration to develop novel computational tools for data analysis. As important results of our model, the network evolved will be capable of reducing redundancy and describing data structure, including their spatial distribution and cluster interrelations. Clustering is useful in several exploratory pattern analyses, grouping, decision-making and machine-learning tasks, including data mining, knowledge discovery, document retrieval, image segmentation and automatic pattern classification. The data clustering approach was implemented in association with hierarchical clustering and graphtheoretical techniques, and the network performance is illustrated using several benchmark problems. The computational complexity of the algorithm and a detailed sensitivity analysis of the user-defined parameters are presented. A trade-off among the proposed model for data analysis, connectionist models (artificial neural networks) and evolutionary algorithms is also discussed.
