Effects of larval diet on the life-history traits and phenotypic expression of pyrethroid resistance in the major malaria vector Anopheles gambiae s.s.

The success achieved in reducing malaria transmission by vector control is threatened by insecticide resistance. To strengthen the current vector control programmes, the non-genetic factors underlying the emergence of insecticide resistance in Anopheles vectors and its widespread need to be explored. This study aimed to assess the effects of larval diet on some life-history traits and pyrethroid-insecticide susceptibility of Anopheles gambiae s.s. Three (3) An. gambiae strains, namely Kisumu (insecticide susceptible), AcerKis (homozygous ace-1 R G119S resistant) and KisKdr (homozygous kdr R L1014F resistant) were fed with three different diets (low, medium, and high) of TetraMin ® Baby fish food. Pre-imaginal developmental time, larval mortality, adult emergence rate and female wing length were measured. Mosquito females were exposed to insecticide-treated net (ITN) PermaNet 2.0 and PermaNet 3.0. In the three An. gambiae strains, significant differences in adult emergence rates ( F = 1054.2; df = 2; p <0.01), mosquito wing length ( F = 970.5; df = 2; p <0.01) and adult survival post insecticide exposure ( χ2 = 173; df = 2; p <0.01), were noticed among the three larval diets. Larvae fed with the low food diets took more time to develop, were smaller at emergence and displayed a short lifespan, while the specimens fed with a high regime developed faster and into big adults. Although being fed with a high diet, none of An. gambiae strain harbouring the kdr R and ace-1 R allele survived 24 hours after exposure against PermaNet 3.0. This study showed that variation in the larval diet significantly impacts An. gambiae life-history traits such as larval mortality and developmental time, adult wing length, and female susceptibility to pyrethroid insecticides. Further investigations through field-based studies would allow an in-depth understanding of the implications of these non-genetic parameters on the physiological traits of malaria vectors and consequently improve resistance management.

Comparative efficacy of two pyrethroid-piperonyl butoxide nets (Olyset Plus and PermaNet 3.0) against pyrethroid resistant malaria vectors: a non-inferiority assessment

Background Pyrethroid-PBO nets were conditionally recommended for control of malaria transmitted by mosquitoes with oxidase-based pyrethroid-resistance based on epidemiological evidence of additional protective effect with Olyset Plus compared to a pyrethroid-only net (Olyset Net). Entomological studies can be used to assess the comparative performance of other brands of pyrethroid-PBO ITNs to Olyset Plus. Methods An experimental hut trial was performed in Cové, Benin to compare PermaNet 3.0 (deltamethrin plus PBO on roof panel only) to Olyset Plus (permethrin plus PBO on all panels) against wild pyrethroid-resistant Anopheles gambiae sensu lato (s.l.) following World Health Organization (WHO) guidelines. Both nets were tested unwashed and after 20 standardized washes compared to Olyset Net. Laboratory bioassays were also performed to help explain findings in the experimental huts. Results With unwashed nets, mosquito mortality was higher in huts with PermaNet 3.0 compared to Olyset Plus (41% vs. 28%, P < 0.001). After 20 washes, mortality declined significantly with PermaNet 3.0 (41% unwashed vs. 17% after washing P < 0.001), but not with Olyset Plus (28% unwashed vs. 24% after washing P = 0.433); Olyset Plus induced significantly higher mortality than PermaNet 3.0 and Olyset Net after 20 washes. PermaNet 3.0 showed a higher wash retention of PBO compared to Olyset Plus. A non-inferiority analysis performed with data from unwashed and washed nets together using a margin recommended by the WHO, showed that PermaNet 3.0 was non-inferior to Olyset Plus in terms of mosquito mortality (25% with Olyset Plus vs. 27% with PermaNet 3.0, OR = 1.528, 95%CI = 1.02–2.29) but not in reducing mosquito feeding (25% with Olyset Plus vs. 30% with PermaNet 3.0, OR = 1.192, 95%CI = 0.77–1.84). Both pyrethroid-PBO nets were superior to Olyset Net. Conclusion Olyset Plus outperformed PermaNet 3.0 in terms of its ability to cause greater margins of improved mosquito mortality compared to a standard pyrethroid net, after multiple standardized washes. However, using a margin of non-inferiority defined by the WHO, PermaNet 3.0 was non-inferior to Olyset Plus in inducing mosquito mortality. Considering the low levels of mortality observed and increasing pyrethroid-resistance in West Africa, it is unclear whether either of these nets would demonstrate the same epidemiological impact observed in community trials in East Africa.

Can pyrethroid-piperonyl butoxide (PBO) nets reduce the efficacy of indoor residual spraying with pirimiphos-methyl against pyrethroid-resistant malaria vectors?

As the uptake of pyrethroid-PBO ITNs increases, their combination with IRS insecticides could become an operational reality in many malaria-endemic communities. Pirimiphos-methyl is a pro-insecticide requiring activation by mosquito cytochrome P450 enzymes to induce toxicity while PBO blocks activation of these enzymes in pyrethroid-resistant vector mosquitoes. PBO may thus antagonise the toxicity of pirimiphos-methyl IRS when combined with pyrethroid-PBO ITNs. The impact of combining two major brands of pyrethroid-PBO ITNs (Olyset Plus, PermaNet 3.0) with pirimiphos-methyl IRS (Actellic 300CS) was evaluated against pyrethroid-resistant Anopheles gambiae sl in two parallel experimental hut trials in southern Benin in comparison to bendiocarb IRS and each intervention alone. The wild vector population was resistant to pyrethroids but susceptible to pirimiphos-methyl and bendiocarb. PBO pre-exposure partially restored deltamethrin toxicity but not permethrin. Mosquito mortality in experimental huts was significantly improved in the combinations of bendiocarb IRS with Olyset Plus (33%) and PermaNet 3.0 (38%) compared to bendiocarb IRS alone (14 to 16%, p<0.001), demonstrating an additive effect. Conversely, mortality was significantly reduced in the combinations of pirimiphos-methyl IRS with Olyset Plus (59%) and PermaNet 3.0 (55%) compared to pirimiphos-methyl IRS alone (77 to 78%, p<0.001), demonstrating an antagonistic effect. Combining pirimiphos-methyl IRS with the pyrethroid-PBO ITNs provided significantly improved mosquito mortality (55 to 59%) compared to the pyrethroid-PBO ITNs alone (22 to 26%) and improved blood-feeding inhibition relative to the IRS alone. This study provided evidence of an antagonistic effect when pyrethroid-PBO ITNs were combined with pirimiphos-methyl IRS in the same household resulting in lower levels of vector mosquito mortality compared to the IRS alone. Pirimiphos-methyl IRS also showed potential to significantly enhance malaria control when deployed to complement pyrethroid-PBO ITNs in an area where PBO fails to fully restore susceptibility to pyrethroids.

An Experimental Hut Evaluation of PBO-Based and Pyrethroid-Only Nets against the Malaria Vector Anopheles funestus Reveals a Loss of Bed Nets Efficacy Associated with GSTe2 Metabolic Resistance

Growing insecticide resistance in malaria vectors is threatening the effectiveness of insecticide-based interventions, including Long Lasting Insecticidal Nets (LLINs). However, the impact of metabolic resistance on the effectiveness of these tools remains poorly characterized. Using experimental hut trials and genotyping of a glutathione S-transferase resistance marker (L119F-GSTe2), we established that GST-mediated resistance is reducing the efficacy of LLINs against Anopheles funestus. Hut trials performed in Cameroon revealed that Piperonyl butoxide (PBO)-based nets induced a significantly higher mortality against pyrethroid resistant An. funestus than pyrethroid-only nets. Blood feeding rate and deterrence were significantly higher in all LLINs than control. Genotyping the L119F-GSTe2 mutation revealed that, for permethrin-based nets, 119F-GSTe2 resistant mosquitoes have a greater ability to blood feed than susceptible while the opposite effect is observed for deltamethrin-based nets. For Olyset Plus, a significant association with exophily was observed in resistant mosquitoes (OR = 11.7; p < 0.01). Furthermore, GSTe2-resistant mosquitoes (cone assays) significantly survived with PermaNet 2.0 (OR = 2.1; p < 0.01) and PermaNet 3.0 (side) (OR = 30.1; p < 0.001) but not for Olyset Plus. This study shows that the efficacy of PBO-based nets (e.g., blood feeding inhibition) against pyrethroid resistant malaria vectors could be impacted by other mechanisms including GST-mediated metabolic resistance not affected by the synergistic action of PBO. Mosaic LLINs incorporating a GST inhibitor (diethyl maleate) could help improve their efficacy in areas of GST-mediated resistance.

Investigation of the influence of a glutathione S-transferase metabolic resistance to pyrethroids/DDT on mating competitiveness in males of the African malaria vector, Anopheles funestus

Background: Metabolic resistance is a serious challenge to current insecticide-based interventions. The extent to which it affects natural populations of mosquitoes including their reproduction ability remains uncharacterised. Here, we investigated the potential impact of the glutathione S-transferase L119F-GSTe2 resistance on the mating competitiveness of male Anopheles funestus, in Cameroon. Methods: Swarms and indoor resting collections took place in March, 2018 in Tibati, Cameroon. WHO tube and cone assays were performed on F1 mosquitoes from indoor collected females to assess the susceptibility profile of malaria vectors. Mosquitoes mated and unmated males collected in the swarms were genotyped for the L119F metabolic marker to assess its association with mating male competitiveness. Results: Susceptibility and synergist assays, showed that this population was multiple resistant to pyrethroids, DDT and carbamates, likely driven by metabolic resistance mechanisms. Cone assays revealed a reduced efficacy of standard pyrethroid-nets (Olyset and PermaNet 2.0) with low mortality (<25%) whereas synergist PBO-Nets (Olyset Plus and PermaNet 3.0) retained greater efficacy with higher mortality (>80%). The L119F-GSTe2 mutation, conferring pyrethroid/DDT resistance, was detected in this An. funestus population at a frequency of 28.8%. In addition, a total of 15 mating swarms were identified and 21 An. funestus couples were isolated from those swarms. A comparative genotyping of the L119F-GSTe2 mutation between mated and unmated males revealed that heterozygote males 119L/F-RS were less able to mate than homozygote susceptible (OR=7.2, P<0.0001). Surprisingly, heterozygote mosquitoes were also less able to mate than homozygote resistant (OR=4.2, P=0.010) suggesting the presence of a heterozygote disadvantage effect. Overall, mosquitoes bearing the L119-S susceptible allele were significantly more able to mate than those with 119F-R resistant allele (OR=2.1, P=0.03). Conclusion: This study provides preliminary evidences that metabolic resistance potentially exerts a fitness cost on mating competiveness in resistant mosquitoes.

Efficacy of a Mosaic Long-Lasting Insecticide Net, PermaNet 3.0, against Wild Populations of Culex quinquefasciatus in Experimental Huts in Togo

PermaNet 3.0 was evaluated against Culex quinquefasciatus in experimental huts in Lomé. Endpoints were deterrence, exophily, blood feeding inhibition, and mortality. Insecticide susceptibility of Culex quinquefasciatus was assessed with permethrin (1%), DDT (4%), bendiocarb (0.1%), deltamethrin (0.5%, 0.05%), carbosulfan (0.4%), and chlorpyrifos methyl (0.4%). Total of 1,223 Cx. quinquefasciatus females were collected. PermaNet 3.0 unwashed deterred 16.84% Culex mosquitoes. After 20 washes, it deterred 5.79% mosquitoes compared to 6.84% deterrence by unwashed PermaNet 2.0. PermaNet 3.0 induced mosquitoes to exit huts 50.48% and inhibited blood feeding 70.97% in unwashed state. After 20 washes, the net induced 42.91% mosquitoes to exit and inhibited 67.06% mosquitoes from blood feeding. PermaNet 3.0 gave 76% personal protection at zero wash and 69% protection after 20 washes. The net retained 7.1% insecticidal effect in the unwashed state and after 20 washes 6.5%. Cx. quinquefasciatus was resistant to tested insecticides (6%–50% mortality). PermaNet 3.0 is a good control tool against mosquitoes. However, Cx. quinquefasciatus is less affected by PermaNet 3.0. The evaluation depicts the success of vector control innovations using pyrethroids and nonpyrethroids in combination on nets. Additional studies with Culex species are recommended to know whether the difference in blood feeding is interspecific (difference in vector behaviour) and not due to intertrial variability.
