The Arndt–Eistert reaction offers a convenient method for the synthesis of the higher members of the aliphatic carboxylic acid series. Nonadecanoic acid, eicosanoic acid, and heneicosanoic acid have been prepared successively from stearic acid in good yields. An efficient method of purification of the synthetic products is described. The ultraviolet absorption maxima for some diazoketones derived from the higher members of the aliphatic carboxylic acid series are recorded.

The long-read Nanopore sequencing has been recently applied for assembly of complex genomes and analysis of linear genome organization. The most critical factor for successful long-read sequencing is extraction of high molecular weight (HMW) DNA of sufficient purity and quantity. The challenges associated with input DNA quality are further amplified when working with extremely small insects with hard exoskeletons. Here, we optimized the isolation of HMW DNA from the model beetle Tribolium and tested for use in Nanopore sequencing. We succeeded in overcoming all the difficulties in HMW handling and library preparation that were encountered when using published protocols and commercial kits. Isolation of nuclei and subsequent purification of DNA on an anion-exchange chromatography column resulted in genomic HMW DNA that was efficiently relaxed, of optimal quality and in sufficient quantity for Nanopore MinION sequencing. DNA shearing increased average N50 read values up to 26 kb and allowed us to use a single flow cell in multiple library loads for a total output of more than 13 Gb. Although our focus was on T. castaneum and closely related species, we expect that this protocol, with appropriate modifications, could be extended to other insects, particularly beetles.

A simple, mild and efficient method to prepare HSi- or HOSi-telechelic, high-molecular-weight polydimethylsiloxane polymers (to 41,600 g·mol−1) using the one-shot hydrolysis of MHMH is reported; titration of the water allowed for higher molecular weights (to 153,900 g·mol−1). The “living” character of the chain extension processes was demonstrated by adding a small portion of MHMH and B(C6F5)3 (BCF) to a first formed polymer, which led to a ~2-fold, second growth in molecular weight. The heterogeneous reaction reached completion in less than 30 min, much less in some cases, regardless of whether it was performed neat or 50 wt% in dry toluene; homogeneous reactions in toluene were much slower. The process does not involve traditional redistribution, as judged by the low quantities (<3%) of D4 produced. However, it is not possible to avoid Chojnowski metathesis from MHDDMH giving D3, which occurs competitively with chain extension.
