The effect of β-alanine on the concentration of taurine and other free amino acids (FAA) in different osmotic conditions in Mytilus galloprovincialis have been investigated. Significant variation of the concentration of FAA as a function of the salinity have been observed.β-alanine incorporation decreases the level of cysteic acid and taurine as well as the level of other FAA.

The hemolymph metabolome of Mytilus galloprovincialis injected with live Vibrio splendidus bacteria was analyzed by 1H-NMR spectrometry. Changes in spectral hemolymph profiles were already detected after mussel acclimation (3 days at 18 or 25 °C). A significant decrease of succinic acid was accompanied by an increase of most free amino acids, mytilitol, and, to a smaller degree, osmolytes. These metabolic changes are consistent with effective osmoregulation, and the restart of aerobic respiration after the functional anaerobiosis occurred during transport. The injection of Vibrio splendidus in mussels acclimated at 18°C caused a significant decrease of several amino acids, sugars, and unassigned chemical species, more pronounced at 24 than at 12 h postinjection. Correlation heatmaps indicated dynamic metabolic adjustments and the relevance of protein turnover in maintaining the homeostasis during the response to stressful stimuli. This study confirms NMR-based metabolomics as a feasible analytical approach complementary to other omics techniques in the investigation of the functional mussel responses to environmental challenges.

Определяли активность цитоплазматических оксидоредуктаз: малатдегидрогеназы (МДГ, 1.1.1.37) и лактатдегидрогеназы (ЛДГ, 1.1.1.27), в тканях Mytilus galloprovincialis Lamarck, 1819 (Bivalvia: Mytilidae), Аnadara kagoshimensis (Tokunaga, 1906) (Bivalvia: Arcidae) и Rapana venosa (Valenciennes, 1846) (Gastropoda: Muricidae). Объектами исследования служили половозрелые моллюски. Длина раковины мидии составляла 50–55 мм, анадары – 25–33 мм, рапаны – 50–80 мм. Активность ферментов измеряли спектрофотометрически (при 340 нм и 25ºC) по скорости окисления НАДН в цитоплазме тканей. На основе анализа собственных и опубликованных литературных данных проведено сравнение активности ферментов и содержания низкомолекулярных антиоксидантов (НМАО) у исследованных моллюсков. Ткани с меньшим индексом МДГ/ЛДГ (гепатопанкреас мидии, нога анадары, гонады рапаны), в которых преобладают аэробные процессы, содержат наибольшее количество каротиноидов, глутатиона и меньше мочевины. Активность ферментов энергетического обмена и резерв НМАО в тканях вселенца анадары значительно больше, чем у черноморской мидии.

AbstractHemocyanins are giant oxygen transport proteins that freely float within the hemolymph of most molluscs. The basic quaternary structure of molluscan hemocyanins is a cylindrical decamer with a diameter of 35 nm which is built of 400 kDa subunits. Previously published results, however, showed that one out of two hemocyanin subunits of Rapana venosa encompasses two polypeptides, one 300 kDa and one 100 kDa polypeptide which aggregate to typical 4 MDa and 8 MDa hemocyanin (di-)decamer molecules. It was shown that the polypeptides are bound most probably by one or more cysteine disulfide bridges but it remained open if these polypeptides were coded by one or two genes. Our here presented results clearly showed that both polypeptides are coded by one gene only and that this phenomenon can also be found in the gastropod Nucella lapillus. Thus, it can be defined as clade-specific for Muricidae, a group of the very diverse Caenogastropoda. In addition, we discovered a further deviation of this hemocyanin subunit within both species, namely a region of 340 mainly hydrophilic amino acids (especially histidines and aspartic acids) which have not been identified in any other molluscan hemocyanin, yet. Our results indicate that, within the quaternary structure, these additional amino acids most probably protrude within the inner part of didecamer cylinders, forming a large extra mass of up to 800 kDa. They presumably influence the structure of the protein and may affect the functionality. Thus, these findings reveal further insights into the evolution and structures of gastropod hemocyanins.
