Polyamines are small positively charged molecules in plants and play important functions in many biological processes under various environmental stresses. One of the most confounding problems relating to polyamines (PAs) in stresses is the lack of understanding of the mechanisms underlying their function(s). Furthermore, a limited number of studies have addressed this issue at the sub-cellular level, especially in tree plants under drought stress. Therefore, in this research, by simulating natural drought stress with polyethylene glycol (PEG) osmotic stress, the relationship between the levels of conjugated polyamines and the activity of H+-ATPase in the plasma membrane was elucidated with the roots of two plum (Prunus salicina L.) cultivars, which were different in drought tolerance, as experimental materials. Furthermore, free PA levels and the activities of S-adenosylmethionine decarboxylase (SAMDC) and transglutaminase (TGase), which were closely associated with the levels of free and conjugated PAs, were also detected. Results showed that under osmotic stress, the increases of the levels of non-covalently conjugated (non-CC) spermidine (Spd) and spermine (Spm), covalently conjugated (CC) putrescine (Put) and Spd in the plasma membrane of drought-tolerant Ganli No. 5 were more significant than those of drought-sensitive Suli No. 3, indicating that these conjugated PAs might be involved in the tolerance of plum seedlings to stress. Furthermore, the conjugated PAs were closely correlated with plum seedling growth, water retention capacity, plasma membrane damage degree, and hydrogen (H+)-ATPase activity in the plasma membrane. To get more complementary pieces of evidence, we subjected plum seedlings to combined treatments of PEG and exogenous PA (Spd and Spm), and an inhibitor of SAMDC [methylglyoxal-bis (guanylhydrazone), (MGBG)] or TGase (o-phenanthroline). These results collectively suggested that non-CC Spd and Spm, CC Put and Spd in plasma membrane might function in enhancing the tolerance of plum seedlings to osmotic stress by stabilizing membrane structure and therefore elevating H+-ATPase activity.

Drought is one of the most detrimental environmental stressors to plants with the potential to decrease crop yields and affect agricultural sustainability. Native bacteria with beneficial traits enhance plant growth and help avoid and reverse the effects of drought in plants to a greater extent. In the present study, we aimed to ( i ) isolate drought-tolerant Bacillus isolates from the rhizosphere soil of wheat crop grown at different locations in Jaisalmer district, Rajasthan state and (ii) further evaluate their ability to enhance plant growth and induce drought tolerance in wheat ( Var. HD-2967) grown under drought stress conditions. Of more than 100 isolates, two putative Bacillus isolates capable of tolerating 30 % polyethylene glycol-6000 (PEG-6000) [equivalent to -9.80 MPa (Megapascal)] were identified as Bacillus altitudinis DT-89 and Bacillus paramycoides DT-113. These isolates exhibited different plant growth promoting (PGP) attributes such as phosphate solubilization, and production of siderophore, exopolysaccharide, ammonia, indole acetic acid and cytokinin at low osmotic stress of 10% PEG-6000 but shown variable response at higher osmotic stress particularly at 30% PEG-6000. However, they did not show any antifungal activity and one isolate was negative for phosphate solubilization. Of two strains, B. altitudinis DT-89 function more prominently with respect to plant growth promotion and drought tolerance to plant in the early stage but protective traits of B. paramycoides DT-113 was more prominent after 75 days as evident by increased EPS (164%), root dry weight (144.44%), chlorophyll content (90.26%), SOD (389%) and proline (99.3%). The results support both the strains as a potential candidate to alleviate drought stress and enhance plant growth in the drought regions.

Extremophiles are the most ancient microbes on the Earth and also a center of attraction for the scientific community for research because of their ability to adapt to extreme habitats. Compatible solutes are among those factors which enable these microorganisms to thrive in such extreme habitats. Under osmotic stress, the majority of extremophiles accumulate specific organic solutes such as amino acids, sugars, polyols, and their derivatives. In addition, proteins in extremophiles are found to be evolved by changing their amino acid composition to alter the hydrophobicity of its core and surface charge to maintain activity. This chapter encompasses a comprehensive study about the role of various compatible solutes in the endurance of microorganisms under extremophilic conditions, synthesis of compatible solutes, nature of extremophilic proteins, and their applications. Furthermore, an attempt has been made to cover various strategies adopted by the scientific community while pursuing research on compatible solutes.

Abstract Vitrification, is an ultra-rapid, manual cooling process that produces glass-like (ice crystal free) solidification. Water is prevented from forming intercellular and intracellular ice crystals during cooling as a result of oocyte dehydration and the use of highly concentrated cryoprotectant. Though oocytes can be cryopreserved without ice crystal formation through vitrification, it is still not clear whether the process of vitrification causes any negative impact (temperature change/chilling effect, osmotic stress, cryoprotectant toxicity, and/or phase transitions) on oocyte quality that translate to diminished embryo developmental potential or subsequent clinical outcomes. In this review, we attempt to assess the technique’s potential effects and the consequence of these effects on outcomes.

Potassium (K) is the most abundant cation in plants, playing an important role in osmoregulation. Little is known about the effect of genotypic variation in the tolerance to osmotic stress under different K treatments in barley. In this study, we measured the interactive effects of osmotic stress and K supply on growth and stress responses of two barley cultivars (Hordeum vulgare L.) and monitored reactive oxygen species (ROS) along with enzymatic antioxidant activity and their respective gene expression level. The selected cultivars (cv. Milford and cv. Sahin-91Sahin-91) were exposed to osmotic stress (−0.7 MPa) induced by polyethylene glycol 6000 (PEG) under low (0.04 mM) and adequate (0.8 mM) K levels in the nutrient solution. Leaf samples were collected and analyzed for levels of K, ROS, kinetic activity of antioxidants enzymes and expression levels of respective genes during the stress period. The results showed that optimal K supply under osmotic stress significantly decreases ROS production and adjusts antioxidant activity, leading to the reduction of oxidative stress in the studied plants. The cultivar Milford had a lower ROS level and a better tolerance to stress compared to the cultivar Sahin-91. We conclude that optimized K supply is of great importance in mitigating ROS-related damage induced by osmotic stress, specifically in drought-sensitive barley cultivars.
