Fundamental aspects of DNA replication, such as the anatomy of replication stall sites, how replisomes are influenced by gene transcription and whether the progression of sister replisomes is coordinated are poorly understood. Available techniques do not allow the precise mapping of the positions of individual replisomes on chromatin. We have developed a new method called Replicon-seq that entails the excision of full-length replicons by controlled nuclease cleavage at replication forks. Replicons are sequenced using Nanopore, which provides a single molecule readout of long DNA molecules. Using Replicon-seq, we have investigated replisome movement along chromatin. We found that sister replisomes progress with remarkable consistency from the origin of replication but function autonomously. Replication forks that encounter obstacles pause for a short duration but rapidly resume synthesis. The helicase Rrm3 plays a critical role both in mitigating the effect of protein barriers and facilitating efficient termination. Replicon-seq provides an unprecedented means of defining replisome movement across the genome.

In electronic warfare, source enumeration and direction-of-arrival estimation are important. The source enumeration method based on eigenvalues of covariance matrix from received is one of the most used methods. However, there are some drawbacks such as accuracy less than 100 % at high SNR, poor performance at low SNR and reduction of maximum number of estimating sources. We suggested new method based on eigenvalues gaps, which is named AREG(Accumulated Ratio of Eigenvalues Gaps). Meanwhile, FGML(Fast Gridless Maximum Likelihood) which reconstructs the covariance matrix was suggested by Wu et al., and it improves performance of the existing source enumeration methods without modification of algorithms. In this paper, first, we combine AREG with FGML to improve the performance. Second, we compare the performance of source enumeration and direction-of-arrival estimation methods in Rayleigh fading. Third, we suggest new method named REG(Ratio of Eigenvalues Gaps) to reduce performance degradation in Rayleigh Fading environment of AREG.

A new method to estimate the Doppler and multipath time delay is presented in impulse noise environment. First, the Doppler is estimated by energy cumulation of multipath component based on fractional lower-order-frational correlation transform. Then, an order-reduced signal is reconstructed combining the Doppler with the prior knowledge of the transmitted signal, and the echoes signal is converted to many single-frequency signals. Finally, the multipath time delay is obtained by the fractional lower-order power spectrum method. The method is adapted for low SNR noise, can restrain the affection of the cross-items, and has a high time-delay estimation resolution. Some computer simulations are given in this paper and the results show that the new method is valid.

<p><span lang="EN-US">The rapid intensification rate and short duration are currently two different notions in defining flash droughts. Here we make a thorough evaluation on these two approaches and illustrate which approach can provide more accurate information of flash drought and why. Based on the conception of intensification rate, a new method focusing on soil moisture depletion during the onset-development phase is proposed, and its performance in monitoring flash drought events, their onset time and spatial dynamics are compared with those from the short-duration method. Results show that the proposed rapid-intensification approach is superior to the short-duration approach in capturing the continuous evolution process of drought. Since the short-duration approach ignores the change of soil moisture with time, it can hardly ensure the identified flash drought events all have the rapid evolving characteristic. Meanwhile, the miss monitoring for flash drought onset is also observed for the short-duration approach. The unreasonable selections of hydro-meteorological variables and corresponding thresholds, particularly refers to that of temperature, is the main reason for the poor behavior of the short-duration approach.</span></p>
