Isolation and Analysis of Genistein of Overripe Tempe using Column Chromatography MethodABSTRACTGenistein is one of the aglycone isoflavone compounds in tempe that has various biochemical activities, including anticancer, antitumor, and antioxidants. Commonly used isoflavone extraction methods resulted in isoflavone crude extract. The aim of this study was to isolate the genistein of overripe tempe through determining the appropriate combination of mobile phases in genistein isolation and the determination of genistein content in both crude extract and isolate. The overripe tempe was first extracted, then genistein was isolated from the crude extract using column chromatography method. The determination of mobile phase combination was done by Thin Layer Chromatography while the genistein content was quantitatively determined by using High Performance Liquid Chromatography. The results showed that the appropriate combination of mobile phase for genistein isolation was chloroform : methanol (15 : 1, v/v). Genistein content in the crude extract and isolates were 4737.50 and 31.36 μg/g extract, respectively. The genistein purity in the isolates was 63.80%, while the purity in the isoflavone extract was 31.98%.Keywords: genistein, HPLC, isoflavone, overripe tempe, TLC ABSTRAKGenistein merupakan salah satu senyawa isoflavon aglikon dalam tempe yang memiliki bermacam-macam aktivitas biokimia, diantaranya antikanker, antitumor, dan antioksidan. Metode ekstraksi isoflavon yang umum diterapkan, menghasilkan ekstrak kasar isoflavon yang masih berupa campuran. Tujuan dari penelitian ini adalah untuk mengisolasi genistein dari tempe busuk melalui tahap penentuan kombinasi fase gerak yang tepat dalam isolasi genistein serta penentuan kandungan genistein baik dalam ekstrak kasar maupun isolat. Tempe busuk mula-mula diekstrak, selanjutnya genistein diisolasi dari ekstrak kasar menggunakan metode kromatografi kolom. Penentuan kombinasi fase gerak dilakukan secara Kromatografi Lapis Tipis, sedangkan kandungan genistein secara kuantitatif ditentukan dengan menggunakan Kromatografi Cair Kinerja Tinggi. Hasil penelitian menunjukkan bahwa kombinasi fase gerak yang tepat untuk isolasi genistein adalah kloroform : metanol (15 : 1, v/v). Kandungan genistein dalam ekstrak kasar dan isolat genistein berturut-turut sebesar 4737,50 dan 31,36 μg/g ekstrak. Kemurnian genistein dalam isolat adalah sebesar 63,80%, sedangkan kemurniannya dalam ekstrak isoflavon adalah sebesar 31,98%. Kata Kunci: genistein, HPLC, isoflavon, tempe busuk, KLT

Grapsus sp crab has a greenish black body color and also known as stone crab. Crabs in the genus Grapsus sp have a swift movement, long legs, they do not have swimming legs and have small reddish purple or purple-orange color, claws on the body of this crab indicated the presence of pigments such as carotenoid pigments. Carotenoid pigments are one form of secondary metabolites which consist of carotene and xanthophyll groups. Carotenoid pigments are present in yellow, orange or orange red which are also found in crabs. Separation of carotenoid pigments can be done using the TLC method, High Performance Liquid Chromatography (HPLC) and Column Chromatography (CC). The purpose of this study was to determine the type of carotenoid pigment from male Grapsus sp crab extract using the Column Chromatography separation method. From the results of this study, the carotenoid pigment content in the 1,2 and 3 carapace was 46,85 μg, 39 µg, and 33,14 µg. The carotenoid pigment concentrations in carapace extracts 1,2 and 3 are 25,38 µg/g, 23,4 µg/g and 5,11 µg/g. From the results of the separation using the column chromatography method, the type of carotenoid pigment identified from the carapace extract of Grapsus sp male is β-Carotene, Ekinenon, Astaxantine, Kantaxantine and Astacen.Keywords: Grapsus sp, Carotenoid, Column ChromatographyKepiting Grapsus sp memiliki warna tubuh hitam kehijauan dan dikenal dengan nama kepiting batu. Kepiting dalam genus Grapsus sp memiliki gerakkan yang cekatan, mempunyai kaki yang panjang, tidak memiliki kaki renang dan memiliki capit berukuran kecil yang berwarna ungu kemerahan atau ungu-oranye warna pada tubuh kepiting ini mengindikasikan adanya kandungan pigmen seperti pigmen karotenoid. Pigmen karotenoid merupakan salah satu bentuk metabolit sekunder yang yang terdiri dari golongan karoten dan xantofil. Pigmen karotenoid  hadir dalam warna kuning, oranye, atau merah oranye, yang juga ditemukan pada kepiting. Pemisahan pigmen karotenoid dapat dilakukan dengan menggunakan metode KLT, Kromatografi Cair Kinerja Tinggi (KCKT) dan Kromatografi Kolom (KK). Tujuan dari penelitian ini adalah untuk mengetahui jenis pigmen karotenoid dari ekstrak kepiting Grapsus sp Jantan dengan menggunakan metode pemisahan Kromatografi Kolom. Dari hasil penelitian ini, didapatkan kandungan pigmen karotenoid pada karapas 1,2 dan 3 adalah 46,85 µg 39 µg, dan 33,14 µg. Konsentrasi pigmen karotenoid pada ekstrak karapas 1,2 dan 3 adalah 25,38 µg/g, 23,4 µg/g dan 5,11 µg/g. Hasil pemisahan menggunakan metode pemisahan kromatografi kolom didapatkan ekstrak karapas kepiting Grapsus sp jantan memiliki jenis pigmen β-Karoten, Ekinenon, Astaxantin, Kantaxantin dan Astasen.Kata kunci: Grapsus sp, Karotenoid, Kromatografi Kolom,

A reliable and reproducible reversed-phase high performance liquid chromatography (RP-HPLC) was developed for the quantitative determination of Remipril drug content from marketed bulk tablets. The active ingredient of Remipril separation achieved with C18 column using the methanol water mobile phase in the ratio of 40:60 (v/v). The active ingredient of the drug content quantify with UV detector at 215 nm. The retention time of Remipril is 5.63 min. A good linearity relation (R2=0.999) was obtained between drug concentration and average peak areas. The limit of detection and limit of quantification of the instrument were calculated 0.03 and 0.09 µg/mL, respectively. The accuracy of the method validation was determined 102.72% by recoveries method.
