The ability of Aspergillus niger strain to reduce organic and mineral pollution as well as the toxicity of two tannery wastewaters, the unhairing effluent (UE) and the final effluent (FE), taken from a local Tunisian tannery and under nonsterile conditions, was studied. Raw effluents show alkaline pH ≥11; thus experiments were carried out at initial pH values and at pH adjusted to 6. Characterization of effluents also revealed high salt levels (EC > 17 mS/cm) and high organic matter content (25 g/L for the UE and 7.2 g/L for the FE) but a low biodegradability since BOD5 did not exceed 2.5 and 1.25 g/L for the UE and the FE, respectively. The results of the biological treatment showed that A. niger was able not only to grow at high pH and salinity values, but also to reduce organic and mineral pollutant load. After treatment, the COD reduction for the UE reached 90% and 70% at pH=6 and at initial pH (12.13), respectively. For the FE, the decrease of COD values reached 75% at pH=6 and 64% at initial pH (11.64). Monitoring of mineral pollution levels showed a reduction in chromium (Cr) concentrations reaching 70% for the FE. This was reflected by an increase of the biomass of A. niger from 9.25 g/L (control) to 9.84 g/L for the FE. To confirm the efficiency of the biological treatment using A. niger, phytotoxicity (tomato seeds) and microtoxicity (Escherichia coli and Bacillus subtilis) tests were carried out. Results of this monitoring showed an important decrease in the toxicity levels for both effluents.

We treat black liquor using Aspergillus niger to extract lignin and to improve the utilization value of pulping black liquor. Results show that: The original black liquor directly to the biological treatment by Aspergillus niger, biological bacteria cannot grow, when reaction liquid pH ≤ 9; Inorganic salt concentration ≤7% (mass ratio); The black liquor load ratio is 20% (mass ratio), the condition of Aspergillus niger growth is good, the ability to produce acid is strong. After treatment of Aspergillus niger, liquid pH decreased to 3, Aspergillus niger changes settling performance of lignin, and it’s good for extraction and purification of lignin. Lignin methoxyl content reduced 2.81%, alcoholic hydroxyl content reduced 0.31%, and carboxyl content increased 0.15%.

Debarking effluents of the forest industry are severely toxic waste streams due to their high tannin content. The objectives of this study were to evaluate the treatment and detoxification of debarking wastewater with tannin tolerant fungi. For this purpose, Aspergillus niger was cultivated on aqueous bark extracts and the toxicity was assayed utilizing methanogenic bacteria as the test organism. Sterilized aqueous extracts of spruce bark were diluted to 5.2 g COD L−1 (containing 2.7 g tannin COD L−1) in either citrate or tartrate buffer and inoculated with A. niger spores. During four day aerobic fermentations with citrate buffer, the elimination of unfiltered and soluble COD was 28 and 63%, respectively. The apparent yield of biomass averaged 34% of the extract COD; however, 12% of the extract COD was tannins adsorbed on the fungal biomass. The decrease in the tannin concentratio was 50%. According to gel chromatography results, the original oligomeric tannins that resisted biodegradation were those of the highest MW. The toxicity was consequently only partially reduced by the fermentation. During the tartrate buffered fermentations, the pH rose due to the metabolism of the organic acid in the buffer. The rise in pH beyond 6 promoted autoxidative reactions that caused extensive polymerization of the tannins which resisted biodegradation. This resulted not only in an enhanced elimination of the tannins but also in a complete detoxification of the extracts. Similar levels of detoxification could be obtained in sterile un-inoculated extracts by autoxidation alone (raising the pH and aerating the extracts). Likewise, extracts fermented in citrate buffer could be completely detoxified by a subsequent autoxidation treatment. The exclusion peak of the gel chromatograms (MW > 30,000 g mole−1) obtained from the highly autoxidized samples accounted for 53% of the UV absorbance, whereas no exclusion peak was present in the unoxidized extracts. For the biological treatment and detoxification of debarking wastewaters, we recommend the cultivation of tannin tolerant fungi followed by short autoxidation treatments that polymerize the oligomeric tannin fraction resisting biodegradation to non-toxic high molecular weight polymers.

Background: Fungal infection of the nose and paranasal sinuses is an uncommon condition which is now being increasingly recognized. The clinical presentation is not specific with various symptoms such as nasal obstruction, purulent nasal discharge, facial pain, and chronic cough. Only unilaterality may alert the clinician. Purpose: To find the morphological characteristics of the fungus in patients with paranasal sinus fungus ball. Methods: A retrospective study of 13 paranasal sinus fungus balls cases which underwent endoscopic sinus surgery at Department of Otorhinolaryngology Head and Neck Surgery St. Vincentius A Paulo Hospital Surabaya from March, 2012 until December, 2013. Age, sex, histopathology and fungal cultur were analysed. Histopathologic sections of all the patients were stained with hematoxylin and eosin (H&E), and Gomori methenamine silver (GMS). The specimens were then cultured on Sabouraud dextrose agar plates and incubated at 30°C for 1 month. At the end of the incubation period, the samples were evaluated microscopically to detect fungi and identify their species. Results: The age reported of the 13 patients, was ranging from 36 to 63 years old. There was a significant female predominance, 10 female patients (76.92%) and 3 male patients (23.08%). Histopathological examination showed that most causative agents were Aspergillus species 92.31% (12/13). Culture test was positive for 69.23% (9/13). Aspergillus niger (61.54%, 8/13) is the most frequent fungus reported to cause fungus balls. Conclusion: Pattern of histopathologic on HE and GMS is very helpful and sensitive to identify fungi. The most common isolated mould in our study was Aspergillus niger.Keywords: fungus ball, histopathology and culture, Aspergillus nigerABSTRAK Latar Belakang: Infeksi jamur di hidung dan sinus paranasal merupakan kondisi yang jarang terjadi, namun kini lebih sering ditemukan. Gejala klinisnya tidak spesifik dapat berupa obstruksi hidung, sekret dari hidung, nyeri wajah, dan batuk kronis. Bila terjadi unilateral, patut diwaspadai oleh para klinisi. Tujuan: Untuk mengetahui karakteristik morfologi fungus yang didapati pada pasien sinusitis jamur yang kami teliti. Metode: Dilakukan penelitian retrospektif pada 13 pasien sinusitis jamur yang menjalani bedah sinus endoskopi di Departemen Otorinolaringologi-Kepala Leher Rumah Sakit St. Vincentius A Paulo Surabaya dari bulan Maret 2012 sampai dengan Desember 2013. Dilakukan analisis usia, jenis kelamin, histopatologi dan kultur jamur. Pewarnaan preparat histopatologi menggunakan Hematoxylin dan eosin (H&E) dan Gomori Methenamine Silver (GMS). Kemudian spesimen diletakkan pada piring agar Sabouraud dextrose, dan dilakukan inkubasi pada suhu 30°C selama satu bulan. Pada akhir masa inkubasi, sampel dievaluasi dengan mikroskop untuk mendeteksi jamur dan spesiesnya. Hasil: Didapati usia 13 penderita berkisar dari 36-63 tahun. Wanita lebih dominan sebanyak 10 penderita (76,92 %) dan 3 penderita laki-laki (23,08%). Hasil pemeriksaan histopatologi menunjukkan spesies Aspergillus sebagai penyebab utama (92,31%) pada 12 penderita (12/13).Tes kultur positif pada 69,23% (9/13). Jamur yang paling sering menyebabkan bola jamur pada sinus adalah Aspergillus niger (61,54%, 8/13). Kesimpulan: Pewarnaan preparat histopatologi menggunakan Hematoxylin dan eosin (H&E) dan Gomori Methenamine Silver (GMS) sangat berguna dan sensitif dalam mendeteksi adanya jamur. Jenis jamur yang paling banyak ditemukan pada penelitian kami adalah Aspergillus niger.
