Abstract Backguound: To screen the signaling axis of epigenetic modification in serum exosomes of ovarian cancer patients based on sequencing technology and raw signal analysis, in depth study of the potential mechanism of action of ovarian cancer, prediction of potential therapeutic targets and survival prognosis analysis of potential targets.Methods: Serum exosomes from three ovarian cancer patients were selected as the experimental group, and serum exosomes from three uterine fibroid patients as the control group, and whole transcriptome of serum exosomes was performed to obtain differentially expressed lncRNA and mRNA in ovarian cancer,The miRcode database and miRNA target gene prediction website were used to predict the target genes, Cytoscape software was used to draw a ceRNA network model of epigenetic modification of ovarian cancer serum exosomes, and the R language was used for GO and KEGG enrichment analysis of the target genes. Finally, the TCGA website was used to download clinical and expression data related to ovarian cancer, and the common potential target genes obtained in the previous period were analyzed for survival。Results: A total of 117 differentially expressed lncRNAs as well as 513 differentially expressed mRNAs (P < 0.05, |log2 FC|≥ 1.0) were obtained by combining sequencing data and raw signal analysis, and 841 predicted target genes were reciprocally mapped by combining mircode database and miRNA target gene prediction website, resulting in 11 potential target genes related to ovarian cancer (FGFR3, BMPR1B, TRIM29, FBN2, PAPPA, CCDC58, IGSF3, FBXO10, GPAM, HOXA10, LHFPL4), and survival prognosis analysis of the above 11 target genes revealed that the survival curve was statistically significant (P < 0.05) for HOXA10 only genes, but not for the other genes, and through enrichment analysis, we found that the above target genes were mainly involved in biological processes such as regulation of transmembrane receptor protein kinase activity, structural molecule activity with elasticity, transforming growth factor - activated receptor activity, and GABA receptor binding, and were mainly enriched in signaling pathways regulating stem cell pluripotency, bladder cancer, glycerolipid metabolism, central carbon metabolism of cancer, tyrosine stimulation to EGFR in signaling pathways such as resistance to enzyme inhibitors.Conclusions: The serum exosomal DIO3OS-hsa-miR-27a-3p-HOXA10 epigenetic modification signaling axis affects ovarian cancer development and disease survival prognosis by targeting transcriptional dysregulation pathways in cancer.

Abstract Background: Ovarian cancer is one of the common malignant tumors in gynecology. Although the treatment strategy for ovarian cancer has been greatly improved in recent years, due to the metastasis, recurrence and drug resistance, the 5-year overall survival rate of patients is still less than 47%. However, at present, there is no specific markers for clinical application. The purpose of this study is to verify the expression and clinical significance of KIF23 in ovarian cancer and identify potential targets for the clinical treatment of ovarian cancer. Methods: The expression of KIF23 in ovarian cancer tissues and its relationship between survival prognosis and clinical pathological parameters were analyzed in Oncomine, GEO, and TCGA databases. KIF23 expression was analyzed by Kaplan-Meier plotter database and its relationship with chemo-resistance was studied. The molecular mechanism involved in KIF23 was analyzed from the perspective of gene mutation, copy number variation and other genomics. Finally, immunohistochemistry experiment was used to verify the expression of KIF2, and its relationship between the clinical pathological parameters and prognosis of ovarian cancer patients was analyzed by single factor and multivariate Cox regression models. Results: Bioinformatic and experimental results have demonstrated that KIF23 is highly expressed in ovarian cancer, and its high expression is positively correlated with poor prognosis. Overexpression of KIF23 can cause chemotherapy resistance in ovarian cancer and affect the overall survival of patients. Genomics analysis showed that KIF23 expression was associated with mutations such as FLG2 and TTN, and it was significantly enriched in tumor signaling pathways such as DNA replication and cell cycle. Conclusions: KIF23 can not only be used as a biomarker of poor prognosis in patients with various stages of ovarian cancer, but also be used as a molecular targeted drug and an independent prognostic biomarker for the treatment of ovarian cancer patients.

Background: CA-125 level increases in 50% of patients with stage I, 90% of patients with stage II, 92% of patients with stage III and 94% of patients with stage IV ovarian cancer. CA-125 level were not a diagnostic tool to detect ovarian cancer, however it was useful to monitor the progressive of disease and as a prognostic marker.Objectives: The aim of this study is to prove whether CA-125 level before surgery in ovarian cancer patients at Dr. Sardjito Hospital as well as a factor that correlates to the survival prognosis of those patients.Method: This research used cohort retrospective study at Dr. Sardjito Hospital Yogyakarta.Result and Discussion: As much as 71 ovarian cancer patients which had been included in this research with inclusion and exclusion criteria. Subjects were divided into two groups. One group was for patients with low CA-125 level (≤35 U/ml) as much as 18 subjects and another group was for patients with high CA-125 level (>35 U/ml) as much as 53 subjects. The result of a bivariate analysis with an independent survival analysis (Cox’s Regression) was the stage of disease (p=0.005, HR 4.827, CI 95% 1.623 – 14.355) and residual tumour (p=0.029, HR 2.605, CI 95% 1.101 – 6.161) were a survival prognosis factor. Multivariate analysis with a survival analysis (Cox’s Regression) shows CA-125 level (p=0.031, HR 4.131, CI 95% 1.143 – 14.933) and menarche (p=0.003, HR 4.989, CI 95% 1.736 – 14.342) were significantly related with survival prognosis in EOC (Epithelial Ovarian Cancer) patients at Dr. Sardjito Hospital.Conclusion: CA-125 level affects the survival rate of epithelial ovarian cancer patients in Dr. Sardjito Hospital. Besides the level of CA-125, there are other factors that affect the survival rate of epithelial ovarian cancer patients which is the stage of cancer, residual operation and age of menarche.Keywords: CA-125 level, EOC, prognosis, survival.

Circulating tumor cells (CTCs) are essential for hematogenous metastasis. In 2003, it was found out that such cells were present in the blood of patients diagnosed with ovarian cancer (OC). It is known that inflammation plays an important role in tumor progression. There are CSCs with a large number of components in the blood, e.g. cytokines that can modulate the metastatic potential of a tumor cell. The aim of the study is to assess the blood serum cytokine status in ovarian cancer patients with different levels of circulating tumor cells. Materials and Methods. Untreated primary patients (n=24) with histologically or cytologically verified ovarian cancer, stage II–IV according to FIGO classification, were the trial subjects. Flow cytometry was used to detect the number of circulating tumor cells in the blood from the patients; the authors also determined IL-6, IL-17A, IL-1β, TGF-α, IL-4, VEGF, TNF-α, HGF, IL-18, IL-10, IL-8 levels. The results were processed using the Statistica Windows software package. Results. The authors determined that TNF-α, HGF, IL-10, IL-18 cytokine level in the blood serum from OC patients significantly increased, and IL-8 level decreased with CTC increase. Conclusion. The obtained results suggest a correlation of CTC level with TNF-α, HGF, IL-10, IL-18, IL-8 cytokine serum level in patients diagnosed with advanced ovarian cancer. At the same time, a sharp and significant increase in TNF-α level accompanied with CTC increase may indicate a change in the phenotypes of TNF-producing cells in OC. Keywords: ovarian cancer, cytokines, circulating tumor cells. Циркулирующие опухолевые клетки (ЦОК) представляют собой основу гематогенного метастазирования. В 2003 г. было продемонстрировано их существование в крови больных раком яичников (РЯ). Известно, что воспаление играет важную роль в прогрессировании опухолей. В крови ЦОК встречаются с большим количеством компонентов, в т.ч. с цитокинами, которые способны модулировать метастатический потенциал опухолевой клетки. Целью данного исследования была оценка цитокинового статуса сыворотки крови больных раком яичников с различным уровнем циркулирующих опухолевых клеток. Материалы и методы. Объектом исследования явились первичные больные с верифицированным (гистологически либо цитологически) раком яичников II–IV степеней по FIGO (n=24) до лечения. В крови пациенток определяли число циркулирующих опухолевых клеток методом проточной цитофлюориметрии и уровни IL-6, IL-17A, IL-1β, TGF-α, IL-4, VEGF, TNF-α, HGF, IL-18, IL-10, IL-8. Обработку результатов проводили с использованием пакета программ Statistica Windows. Результаты. Было установлено, что уровень цитокинов TNF-α, HGF, IL-10, IL-18 в сыворотке крови больных РЯ статистически значимо повышался, а уровень IL-8 снижался с увеличением количества ЦОК. Выводы. Полученные результаты позволяют предполагать корреляцию уровня ЦОК с сывороточным уровнем цитокинов TNF-α, HGF, IL-10, IL-18, IL-8 у больных распространенным РЯ. При этом резкое и значимое возрастание уровня TNF-α при увеличении числа ЦОК может свидетельствовать о смене фенотипов TNF-продуцирующих клеток при РЯ. Background: The term "ovarian cancer" includes several different types of cancer that  arise from cells of the ovary, most commonly, tumors arise from the epithelium or lining cells of the ovary.  Ovarian cancer risk is positively associated with higher consumption of dietary cholesterol and eggs, and inversely associated with a higher intake of vegetables. High consumption of fats may increase circulating estrogen levels, thus increasing the possibility of cell damage and proliferation that is responsible for cancerous growth. Material & Methods: The present study was conducted at Geetanjali Medical College and Hospital, Udaipur (Rajasthan). Total  100 cases (females) attending the obstetrics and gynecology department for some gynecological and other problem  were selected for this study between the age of 40-60 years, who were attending cancer centre at GEETANJALI MEDICAL COLLEGE AND  HOSPITAL, Udaipur (Rajasthan).                GROUP I: - It consisted of healthy females control subjects (n=50) .By routine examination and tests, we ensured that all the subjects were healthy and there were no signs and symptoms or history of ovarian tumor and diseases GROUP II: - It consisted of ovarian cancer females subjects (n=50) with a history of ovarian tumor. Results:   Higher level of cholesterol, LDL, VLDL and low level of HDL are found in ovarian cancer patients. Conclusion: The present study we highlights the importance and role of serum lipid profile in diagnosis, prognosis and recurrence of the disease. The study shows that serum level of cholesterol, LDL, VLDL was elevated in  patients of ovarian cancer while low level of HDL are found in ovarian cancer patients. 2006-ban jelent meg az Európai Rákkutatási és Kezelési Szervezet korábbi megállapításokat korszerűsítő ajánlása, amelyben a rákos betegekben kialakuló anémia kezelésére adott vértranszfúziót, illetve az ezt követő erythropoetin-kezeléseket értékelik. Az ajánlásban a vértranszfúziót a 9 g% hemoglobinszintre csökkenő anémia esetén javasolják. Eddig a kemoterápia következtében kialakult anémia vértranszfúziós kezelésére egyértelműen meghatározott hemoglobin-határértékszint Magyarországon nem szerepelt. Cél: A szerzők az osztályukon 2005-ben kezelt petefészekrákos betegeknek adott vértranszfúziók gyakoriságát vizsgálták. Nemzetközi, illetve hazai egyértelmű ajánlás hiányában a vizsgálati időszakban a vértranszfúziókat – a beteg klinikai állapotát is figyelembe véve – a 10 g%-ot elérő anémia esetén alkalmazták. Anyag és módszer: Az Országos Onkológiai Intézet Nőgyógyászati Osztályán 190 hám eredetű, petefészekrákos betegben történt kemoterápia. Ha a hemoglobinszint 10 g% alá csökkent, választott vörösvértest-transzfúziót végeztek, majd a betegek többségében (51/64 = 79,6%), erythropoetin-kezelés történt. Eredmény: A 190 közül 64 betegnél (64/190 = 34%) történt vérátömlesztés a kemoterápia kapcsán kialakult anémia miatt, s az utóbbiaknak csaknem felében (34/64 = 53%) 1-nél több alkalommal végeztek vértranszfúziót. A betegek 86%-ában a vértranszfúzióra G2-súlyosságú anémia miatt került sor. Az ismételten szükséges vértranszfúziókat a leggyakrabban a carboplatin-gemcitabin- (16/16) kezelések után alkalmazták. Következtetés: A petefészekrákokban adott kemoterápiák a betegek harmadában okoznak 10 g%-nál súlyosabb fokú anémiát. A vérszegénység kezelésében a vértranszfúzión kívül gondolni kell az erythropoetin-készítmények alkalmazására.
