Vaccines have saved millions of lives and reduced the severity of many infections, but today the reduction in vaccine coverage has been reflected in the resurgence of epidemics of mumps, whooping cough, measles and chickenpox. This has happened because many people do not recognize the effectiveness of vaccination and fear the side effects, in other words, the main concern is the safety of the vaccines. As a consequence, the greater responsibility of the individual and respect for his will can lead authorities to less vigorously promote the “duty of vaccination”, which is also a social duty. Unfortunately, however, the attitude of individuals is guided by their beliefs about health, which are often supported by an erroneous perception of risk arising from false news.

Doxorubicin (Dox) is a wide-spectrum drug to treat different kinds of cancers. However, in clinical practice, Dox usually showed untargeted distributions to the other organs, which can cause serious side effects, such as cardiotoxity. Herein, the formulation of Dox into nanoparticles is critical to enhance its distribution to tumors. Herein, we used polysaccharide, hyaluronic acid, to stabilize the Dox to form nano-precipitations (PD NPs) for the therapy of osteosarcoma. The PD NPs showed enhanced drug accumulation to tumor cells and realized better anticancer effects than free drugs.

Mental disorders are one of the health disorders that contribute to high rates of disability and mortality worldwide. The current therapeutic modalities used to treat mental disorders are medical and psychological approaches, but it becomes problematic in some conditions, such as drug-resistant mental disorders. Radio Electric Asymmetric Conveyer (REAC) technology can be used as an alternative to overcome this problem. This technology uses radio waves which are guaranteed to be non-invasive and do not cause side effects. This technology enables neuromodulation effects by maximizing cell polarity and optimizing endogenous bioelectric activity. Of course, the REAC's mechanism as a neuromodulator and being a non-invasive technology is safe to use. It allows REAC to be used as an adjuvant therapy to reduce symptoms of several mental disorders such as depression, anxiety, bipolar disorder, phobias, and stress.

Introduction: The corona virus disease-19 pandemic is an ongoing global health crisis and the greatest challenge we have been facing right now in the most peculiar ways. It has caused huge loss of lives and has severely affected the global economy and financial markets. Vaccines are a new critical tool to fight this voracious battle. They have been released in several parts of the world. Although the safety and efficacy of these vaccines have been discussed, we know very little about the post-vaccination experience outside clinical trial situations. Adequate information about the effects of the vaccine can aware the public remove misconceptions and increase vaccine acceptability. Aims: To know the percentage of Covishield vaccine coverage and its adverse effects among the staffs of Nepalgunj Medical College. Methods: A descriptive cross-sectional study was conducted among the vaccinated 91 out of 116 staff members of Nepalgunj Medical College (NGMC), Baijanath Rural Municipality-1, Banke. Results: Out of 91 persons, 15.4%(14) developed headache, 20%(18) developed fever, and 8.7%(17) developed body aches. Of those who developed post-vaccination side effects 47%(16) of the persons were from age group 36-55years,17%(1) of them between age group 55-65 years, and none above 65 years developed fever. 20%(13) of total male and 35%(5) of total female developed fever within 96 hours after receiving first dose of Covishield AstraZeneca vaccine. Conclusion: Most of the study subjects reported milder side effects which lasted for less than 5 days. No casualties were reported. Local pain and swelling at the injection site, headache and fever were the most common side effects. The side effects were more common in younger individuals and women.

Abstract Background Psychosis is a complex mental illness divided by positive symptoms, negative symptoms, and cognitive decline. Clinically available medicines are associated with some serious side effects which limit their use. Treatment with flavonoids has been associated with delayed onset and development, decreased risk, or increased improvement of various neuropsychiatric disorders including psychosis with negligible side effects. Therefore, the present study was aimed to investigate the protective effects of hesperidin (flavonoid) alone or its combination with coenzyme Q10 against ketamine-induced psychotic symptoms in mice. Results Ketamine (50 mg/kg, i.p.) was given for 21 days to induce psychosis in Laca mice of either sex. Locomotor activity and stereotypic behaviors, immobility duration (forced swim test), and increased transfer latency (elevated plus maze) were performed to test the effect of hesperidin (50 mg/kg, 100 mg/kg, 200 mg/kg, p.o.) and coenzyme Q10 (20 mg/kg, 40 mg/kg, p.o.) and combination of hesperidin + coenzyme Q10 followed by biochemical and mitochondrial complexes assays. For 21 days, ketamine (50 mg/kg, i.p.) administration significantly produced increased locomotor activity and stereotypic behaviors (positive symptoms), increased immobility duration (negative symptoms) and cognitive deficits (increases transfer latency) weakens oxidative defense and mitochondrial function. Further, 21 days’ administration of hesperidin and coenzyme Q10 significantly reversed the ketamine-induced psychotic behavioral changes and biochemical alterations and mitochondrial dysfunction in the discrete areas (prefrontal cortex and hippocampus) of mice brains. The potential effect of these drugs was comparable to olanzapine treatment. Moreover, the combination of hesperidin with coenzyme Q10 and or a combination of hesperidin + coenzyme Q10 + olanzapine treatment did not produce a significant effect compared to their per se effect in ketamine-treated animals. Conclusions The study revealed that hesperidin alone or in combination with coenzyme Q10 could reduce psychotic symptoms and improve mitochondrial functions and antioxidant systems in mice, suggesting neuroprotective effects against psychosis.

The crisis of Covid-19 has shaken the world healthcare systems. The intensive care resources to manage the medical conditions associated with Covid-19 are consistently found to be inadequate with exploration and implementation of newer treatment avenues for an early recovery. Presently, the use of Tocilizumab (TCZ) in severe to critical affection of Covid-19 is being practiced as an off-label therapy. A narrative review of present knowledge regarding TCZ pharmacology, indications of its use, and potential side effects with clinical implications for an orthopedic surgeon is presented. The article discusses the clinicopathological factors required to be monitored during the perioperative management of an orthopedic patient who may have received TCZ for Covid-19 related illness. The implications of its usage should alert the orthopaedic surgeons for future management of their arthritic surgical patients. The commonly associated side effects and complications in the post-operative phase following an arthroplasty or any orthopaedic surgery are an area of concern and considerable uncertainty. In the post-Covid-19 recovery phase, when surgeons need to plan a surgical intervention then a thorough evaluation of their Covid-19 medical management history may be warranted. Practical guidelines for the management of arthritic surgical patients have been postulated. With an unregulated increased usage of TCZ during Covid-19 management, an orthopaedic surgeon should worry and needs to be aware of the possible consequences in the perioperative period for the post-surgery management. Future research to gain more insights will confirm the implied concerns.

Abstract Background Infraorbital hollows can give a fatigued or aged appearance which can be treated by volumizing the segmented transition from the tear trough to the cheek with hyaluronic acid filler. Due to thin skin and the complex anatomy of the infraorbital area, both short- and long-term side effects from this treatment are very common. While some patients are clear surgical candidates vs. filler candidates, in real-world practice, many, if not most patients are on a continuum where either procedure is appropriate, and the treatment decision is individualized based on each person’s risk vs. benefit profile. Objectives Common aesthetic side effects from hyaluronic acid filler treatment in the infraorbital area will be reviewed, including their etiology, prevention, detection, and treatment. Method The authors’ experience from injecting the infraorbital areas of more than 800 patients in private clinical practice and observations from both short and long-term follow ups over eight years is leveraged to provide detailed guidance. Results Recommendations on injection techniques, patient selection, and patient education are presented along with algorithms for the prevention and management of bruising, short- and long-term swelling, bumps, and blue discoloration (which is usually secondary to swelling from the filler rather than just the filler alone placed or migrating too superficially). Conclusion For nearly all patients, complete dissolution of filler with hyaluronidase is not required to address the issue, and the guidelines provided here will assist clinicians in management of side effects to increase patient satisfaction with their treatment and aesthetic outcome.
