Dissociating action inhibition, conflict monitoring and sensory mismatch into independent components of event related potentials in GO/NOGO task

Abstract Background Impaired error/conflict monitoring as reflected in the event-related potentials (ERPs) has consistently reported in patients with schizophrenia. However, whether this impairment exist from the early phase of psychosis such as first-episode psychosis (FEP) is not yet been clearly reported. To investigate the presence of error/conflict monitoring deficit in early psychosis, we examined the error-related negativity (ERN), error-related positivity (Pe), and correct-response negativity (CRN) during the Go/Nogo task in the patients with FEP. Methods 25 patients with and 25 age, sex matched healthy controls (HCs) were participated in electroencephalographic recording during the Go/Nogo task. Trials with error response was analyzed to define ERN at Fz electrode site and Pe at Pz electrode site. Trials with correct response was used for CRN analysis at Fz electrode site. Independent samples t-test was used to compare the amplitudes of ERP components between FEP and HC groups. Pearson’s correlation analysis was performed to reveal the relationship of altered ERP component with symptomatic severity in patients with schizophrenia. Results FEP patients showed significantly smaller ERN amplitude at Fz electrode site compared to HCs (t=-3.294, p=0.002). However, there was no difference of CRN (t=0.017, p=0.986) and Pe (t=1.806, p=0.077) amplitudes between FEP and HC groups. There was no significant correlation of symptomatic severity and ERN amplitude at Fz electrode site in FEP patients. Discussion These findings suggest that impairments in error/conflict monitoring as reflected by ERN amplitude exist from the early course of psychotic disorder. Future study with larger sample size and subjects at earlier phase such as clinical high risk for psychosis would be needed to confirm the findings of current study.

Event-related potentials of performance monitoring, including N2 (conflict monitoring), error-related negativity and error positivity (ERN and Pe; error monitoring), and P3 (inhibition) have been studied. However, conflict monitoring lacks a behavioural measure, and the functional significance of ERN, Pe, and P3 are debated. To address these issues, a behavioural measure of conflict monitoring was tested by subtracting the reaction time (RT) of a simple from a choice RT task to isolate conflict monitoring; the functions of error monitoring and inhibition were examined. The RT difference correlated with the N2 area (longer conflict monitoring related to a larger N2). ERN and Pe areas were negatively and positively correlated with errors, respectively. P3 magnitude and onset were correlated with an inhibition index. The new behavioural measure provides an accessible way to study conflict monitoring. Theories of conflict monitoring for ERN, error awareness for Pe, and inhibition for P3 were replicated and extended.

Abstract In the last decade, many studies have tried to define the neural correlates of attention deficit hyperactivity disorder (ADHD). The main aim of this study is the comparison of the ERPs independent components in the four QEEG subtypes in a group of ADHD adults as a basis for defining the corresponding endophenotypes among ADHD population. Sixty-seven adults diagnosed as ADHD according to the DSM-IV criteria and 50 age-matched control subjects participated in the study. The brain activity of the subjects was recorded by 19 channel quantitative electroencephalography (QEEG) system in two neuropsychological tasks (visual and emotional continuous performance tests). The ICA method was applied for separation of the independent ERPs components. The components were associated with distinct psychological operations, such as engagement operations (P3bP component), comparison (vcomTL and vcom TR), motor inhibition (P3supF) and monitoring (P4monCC) operations. The ERPs results point out that there is disturbance in executive functioning in investigated ADHD group obtained by the significantly lower amplitude and longer latency for the engagement (P3bP), motor inhibition (P3supF) and monitoring (P4monCC) components. Particularly, the QEEG subtype IV was with the most significant ERPs differences comparing to the other subtypes. In particular, the most prominent difference in the ERPs independent components for the QEEG subtype IV in comparison to other three subtypes, rise many questions and becomes the subject for future research. This study aims to advance and facilitate the use of neurophysiological procedures (QEEG and ERPs) in clinical practice as objective measures of ADHD for better assessment, subtyping and treatment of ADHD.

Aging is accompanied by frontal lobe and non-dominant hemisphere recruitment that supports executive functioning, such as inhibitory control, which is crucial to all cognitive functions. However, the spatio-temporal sequence of processing underlying successful inhibition and how it changes with age is understudied. Thus, we capitalized on the temporal precision of event-related potentials (ERPs) to assess the functional lateralization of N200 (conflict monitoring) and P300 (inhibitory performance evaluation) in young and healthy older adults during comparably performed successful stop-signal inhibition. We additionally used temporal principal components analysis (PCA) to further interrogate the continuous spatio-temporal dynamics underlying N200 and P300 activation for each group. Young adults demonstrated left hemisphere-dominant N200, while older adults demonstrated overall larger amplitudes and right hemisphere dominance. N200 activation was explained by a single PCA factor in both age groups, but with a more anterior scalp distribution in older adults. The P300 amplitudes were larger in the right hemisphere in young, but bilateral in old, with old larger than young in the left hemisphere. P300 was also explained by a single factor in young adults but by two factors in older adults, including distinct parieto-occipital and anterior activation. These findings highlight the differential functional asymmetries of conflict monitoring (N200) and inhibitory evaluation and adaptation (P300) processes and further illuminate unique age-related spatio-temporal recruitment patterns. Older adults demonstrated lateralized recruitment during conflict processing and bilateral recruitment during evaluation and adaptation, with anterior recruitment common to both processes. These fine-grained analyses are critically important for more precise understanding of age-related compensatory activation.

P300 was characterized by an early parieto-occipital peak in both groups, followed by a parietal slow wave only in older adults. A temporally similar but topographically different final wave followed in both groups that showed anterior recruitment in older adults. These findings illuminate differential age-related spatio-temporal recruitment patterns for conflict monitoring and response inhibition that are critically important for understanding age-related compensatory activation.

Deception studies emphasize the important role of event-related potentials (ERPs) to uncover deceptive behavior based on underlying neuro-cognitive processes. The role of conflict monitoring as indicated by the frontal N2 component during truthful and deceptive responses was investigated in an adapted Concealed Information Test (CIT). Previously memorized pictures of faces should either be indicated as truthfully trustworthy, truthfully untrustworthy or trustworthy while concealing the actual untrustworthiness (untrustworthy-probe). Mean, baseline-to-peak and peak-to-peak amplitudes were calculated to examine the robustness of ERP findings across varying quantification techniques. Data of 30 participants (15 female; age: M = 23.73 years, SD = 4.09) revealed longer response times and lower correct rates for deceptive compared to truthful trustworthy responses. The frontal N2 amplitude was more negative for untrustworthy-probe and truthful untrustworthy compared to truthful trustworthy stimuli when measured as mean or baseline-to-peak amplitude. Results suggest that deception evokes conflict monitoring and ERP quantifications are differentially sensitive to a-priori hypotheses.
