Background: The giant gourami sago strain (Osphronemus goramy Lacepède) has been approved in 2018 as a candidate for freshwater aquaculture in Indonesia. However, information on the species’ reproduction is minimal. This study analyzed the reproductive characteristics of the gourami sago strain broodfish to provide basic knowledge for a future hatchery development strategy. Methods: A total of 10 female and 10 male mature gourami sago strain broodfish were measured for body weight and length, and were evaluated for their reproductive characteristics.  Breeding fish were spawned naturally in a 2×1×0.6 m concrete pond with a male-female sex ratio of 1:1. Egg weight and diameter were measured in 25 eggs per female using, respectively, ACIS AD- 600i scales with 0.01 g accuracy, and a microscope (Labo model L-711) using Canon Digital Camera Software 3 . Semen was collected using plastic syringes in 3 mL aliquots, then placed in an insulated ice-cooled container, and analyzed within two hours of collection. Results: Average weights of female and male broodfish before spawning were 2180±159.78 g and 3060±134.99 g, respectively. The relative fecundity and egg diameter were 1029±36 eggs kg-1 and 2.42±0.05 mm, respectively. The hatching rate and embryo survival to an eyed-egg stage were respectively 76.40±2.27% and 94.76±0.42%.  Sperm characteristics showed that volume was 0.60±0.12 ml kg-1 and percentage of motile sperm was 70.04±2.27%. Female broodfish weight after spawning was strongly correlated with the weight before spawned (r2 = 0.999) and absolute fecundity was also strongly correlated with female broodfish weight before spawning (r2= 0.921). Sperm concentration was moderately correlated with sperm motility (r2 = 0.556) and duration of sperm motility (r2 = 0.502). Conclusions: The gourami sago strain broodfish has suitable reproductive characteristics for the development of hatcheries. Successful natural spawning should be followed by larval weaning and feeding technology to increase growth and survival.

Bleaching profoundly impacts coral reproduction, often for years after an event. However, detailed reproductive characteristics of coral after bleaching have not been broadly described, especially as they relate to cryopreservation. Therefore, in the present study we measured several reproductive characteristics in coral in Kaneohe Bay, Hawaii, for two species, namely Fungia scutaria and Montipora capitata, during the bleaching period of 2014 and 2015. We examined spawning periods, egg morphometry, sperm concentration, fresh and cryopreserved sperm motility exposed to different concentrations of dimethyl sulfoxide, time of first cleavage, larval survival with fresh and cryopreserved spermatozoa, infection success and settlement success. Many of these reproductive parameters were reduced in 2015, especially sperm motility. Once the reduced-motility spermatozoa from 2015 post-bleach were cryopreserved, there was a steep decline in post-thaw viability and this would prevent any substantive further use of these samples in reproduction for conservation benefit. Worldwide, as bleaching events become more frequent, the ability to bank and conserve coral ex situ may be significantly reduced. Thus, it is imperative that while genetic diversity is still high in these populations, intensive efforts are made to bank coral species during non-bleaching periods.

Energy for sperm motility is provided by both glycolytic and respiratory pathways. Mitochondria are involved in the latter pathway and conserve energy of substrate oxidation by coupling to phosphorylation. During spermatogenesis, the mitochondria undergo extensive transformation which in many species leads to the formation of a nebemkem. The nebemkem subsequently forms into a helix around the axial filament complex in the middle piece of spermatozoa.Immature spermatozoa of axolotls contain numerous small spherical mitochondria which are randomly distributed throughout the cytoplasm (Fig. 1). As maturation progresses, the mitochondria appear to migrate to the middle piece region where they become tightly packed to form a crystalline-like sheath. The cytoplasm in this region is no longer abundant (Fig. 2) and the plasma membrane is now closely apposed to the outside of the mitochondrial layer.

ABSTRAK                                                                        Penelitian bertujuan untuk mengetahui efektivitas nira aren sebagai pengencer alternatif dalam proses pembekuan (kriopreservasi) semen kambing boer.Kriopreservasi semen kambing boer menggunakan pengencer tris-gliserol-kuning telur (P1 73-7-20%), nira aren-gliseol-kuning telur(masing-masing P2 74-6-20%, P3 73-7-20%, dan P4 72-8-20%) dan andromed (P5 tanpa mengandung kuning telur dan gliserol). Parameter evaluasi meliputi motilitas, viabilitas, dan membrane plasma utuh setelah pengenceran, ekuilibrasi dan thawing.  Evaluasi motilitas pasca thawing menunjukkan P5 52% berbeda nyata (P<0.05) dengan P1 42%, selanjutnya P5 dan P1 berbeda sangat nyata (P<0.05) dengan P2 8%, P3 6% dan P4 12%.  Viabilitas pasca thawing menunjukkan P5 65,4% tidak berbeda nyata (P>0,05) dengan P1 61,8%, akan tetapi P5 dan P1 berbeda sangat nyata (P<0.05) dengan P2 26,2%, P3 29,8%, dan P4 34%.  Membran plasma utuh (MPU) pasca thawing menunjukkan P5 66,2% tidak berbeda nyata (P>0,05) dengan P1 65,4%, akan tetapi keduanya berbeda sangat nyata (P<0.05) dengan P2 39%, P3 38%, dan P4 36,2%.  Disimpulkan kriopreservasi semen kambing boer dengan pengencer nira aren dan gliserol pada konsentrasi berbeda belum dapat dipergunakan sebagai sumber bibit berdasarkan standar nasional Indonesia.Kata Kunci : Kambing boer, semen, nira arenABSTRACTThe experiment was conducted to determine the effect of sugar palm juice as alternative extender for cryopreservation process of boer semen.Tris-glycerol-egg yolk (P1 73-7-20%), Sugar palm juice-glyserol-egg yolk (P2 74-6-20%, P373-7-20%, dan P4 72-8-20%), and andromed (P5) used as a extender  in the cryopreservation process of boer semen.  Sperm motility (%), live sperm (%) and sperm membrane integrity (%) were recorded after diluted, equilibration and freeze-thawing.  Result of post thawing motility showed that P5 52% was significantly different (P <0.05) with P1 42%, then P5 and P1 were significantly different (P <0.05) with P2 8%, P3 6% and P4 12%. Viability after thawing showed P5 65.4% was not significantly different (P> 0.05) with P1 61.8%, but P5 and P1 significantly different (P <0.05) with P2 26.2%, P3 29.8 %, and P4 34%. To obtain further perspective on reproduction and particularly gamete function among so-called primitive mammals presently grouped in the Order Insectivora, we have examined the African hedgehog, Atelerix albiventris, in light of unusual features reported in shrews and moles. Atelerix proves to share many but not all of the characteristics seen in these other insectivores. The penis of Atelerix has a 'snail-like' form, but lacks the surface spines common in insectivores and a number of other mammals. Hedgehog spermatozoa display an eccentric insertion of the tail on the sperm head, and they manifest the barbs on the perforatorium that, in shrews, probably effect the initial binding of the sperm head to the zona pellucida. As a possible correlate, the structural matrix of the hedgehog acrosome comprises only two main components, as judged by immunoblotting, rather than the complex of peptides seen in the matrix of some higher mammals. The Fallopian tube of Atelerix is relatively simple; it displays only minor differences in width and in the arborized epithelium between the isthmus and ampulla, and shows no evidence of the unusual sperm crypts that characterize the isthmus or ampulla, depending on the species, in shrews and moles. In common with other insectivores, Atelerix appears to be an induced ovulator, as judged by the ovulation of some 6-8 eggs by about 23 h after injection of hCG. The dense cumulus oophorus appeared to have little matrix, in keeping with the modest dimensions of the tubal ampulla and, while it was not quite as discrete as that of soricids, it did show the same insensitivity to 0.5% (w/v) ovine or bovine hyaluronidase.
