Correcting cell density measurements for tissue hydration changes in scanning electron microscopy — application to the rabbit corneal endothelium

The aim of this study was to examine the endothelial surface morphology and perform a morphometric analysis of the corneal endothelial cells of ostrich (Struthio camelus) using scanning electron microscopy. Polygonality, mean cell area, cell density and coefficient of variation of mean cell area were analyzed. The normal corneal endothelium consisted of polygonal cells of uniform size and shape with few interdigitations of the cell borders. Microvilli appeared as protusions on the cellular surface. The average cell area was 269±18µm² and the endothelial cell density was 3717±240cells mm-2. The coefficient of variation of the cell area was 0.06, and the percentage of hexagonal cells was 75%. The parameters evaluated did not differ significantly between the right and the left eye from the same ostrich. The results of this study showed that the ostrich corneal endothelial cells appear quite similar to those of the other vertebrates.

The objective of this study was evaluate the maintenance of the corneal endothelium of horses in cold EUSOL-C® preservation medium over different periods (seven and 14 days) using scanning electron microscopy. A total of 20 pairs of eyes from horses were analysed. The corneas were divided into four groups of 10 corneas each (G1, G2, G3 and G4): G1 - the samples were kept in the preservation medium for seven days; G3 - the samples were kept in the preservation medium for for 14 days; G2 and G4 were formed by the control corneal buttons of G1 and G3, respectively. The average cell loss observed in G1 was 7.62%, in G2 it was 7.04%, in G3 9.12% and in G4 7.16%. No statistically significant differences were observed between the four groups. It was concluded that the Eusol-C® hypothermic preservation medium provided satisfactory preservation of the corneal endothelium in equine species for up to 14 days.

This article describes the preparation of gradient porosity thermoset polymers. The technique used is based on polymerizing a solution of cross-linkable dicyclopentadiene and 2-propanol. The forming polymer being insoluble in 2-propanol, phase separation occurs. Subsequent drying of the 2-propanol gives porosities up to 80%. An apparatus was built to produce a gradient in 2-propanol concentration in a flask, resulting in polymerized gradient porosity rods. The resulting materials have been characterized by scanning electron microscopy (SEM) and density measurements. A mathematical model which allows prediction of the gradient produced is also presented.

Keratoconjunctivitis sicca was produced experimentally in 16 beagles by bilateral surgical removal of the lacrimal and nictitans glands; four dogs were not treated, and 12 received tear-replacement therapy on post-operative days 7 through 28. Keratoconjunctivitis sicca was verified by reduction in Schirmer tear test values by post-operative day 6, and there was no response on day 28 to tear-replacement therapy. Corneas of both normal and tear-deficient dogs had polygonal squamous epithelial cells of light and dark electron density by scanning electron microscopy. Light cells had more microvilli and microplicae than dark cells. Conjunctivae were similar to corneas, except for numerous goblet cells on the surface. Corneal dark-cell density and goblet cell density were not different between groups. Goblet cells most often occurred singly in normal dogs, while they were in clusters in tear-deficient dogs. A hypothesis that petrolatum/mineral oil ointment should provide more effective artificial tear replacement than hydroxymethylcellulose drops for tear-deficient dogs could not be confirmed by objective analysis of corneal dark-cell density or conjunctival goblet cell density.

The influence of polymer precursor additions on liquid phase sintering of SiC:Al2O3:Y2O3 has been evaluated. Two polymer precursors were used: polymethylhydrogensiloxane and D4Vi (1,3,5,7-tetramethyl-1,3,5,7-tetraviniylcyclotetrasiloxane). The ceramic phase had the following composition in wt%: 91.6 SiC, 4.2 Al2O3 and 4.2 Y2O3. The composites were prepared using the following ceramic phase to polymer ratios in wt%: 82.7: 17.3; 74.0: 26.0 and 71.5: 28.5. Density measurements were carried out using a helium picnometry and the Archimedes method. The crystalline phases were identified by X-ray diffraction analysis and the microstructures were observed by optical and scanning electron microscopy.
