The present study was intended to evaluate Anti-inflammatory activity ofC. rottleriextracts (Hydroalcoholic, Methanol, Ethyl acetate and Hexane). The Anti-inflammatory activity ofC.rottleriextracts at doses of 125mg/kg, 250mg/kg and 500mg/kg using carrageenan induced rat paw edema model compared with standard drug (Indomethacin). The selected plant extracts significantly inhibited paw edema along with the standard drug Indomethacin. Of all extracts, methanol extract produced significant effect on reduction of increased paw thickness, hydro alcoholic and ethyl acetate extracts produced moderate percentage inhibition and hexane extract produced low level of percentage inhibition in reducing paw edema on carrageenan induced rats. In all extracts, methanol extract at a dose of 500mg/kg showed more percentage inhibition i.e . 53.47±2.19. From the results obtained during the study it is concluded thatC. rottlerihaving the bioactive molecule responsible for Anti-inflammatory activity by individually or by combination of different bio-active compounds present in it. Further is necessary for isolation and characterization of bioactive molecules which are responsible for the selected plant biological activities.

We investigated the anti-inflammatory activity ofSenecio salignus. This medicinal plant is often used in Mexico for the treatment of fever and rheumatism. Chloroform and methanol extracts of the plant were tested on 12-O-tetradecanoylphorbol-13-acetate- (TPA-) induced edema in mice ears. The methanol extract of the plant inhibited edema by36±4.4% compared with the control, while the chloroform extract exhibited an even greater level of inhibition (64.1%). The chloroform extract was then fractionated, and the composition of the active fraction was determined by GC-MS. The anti-inflammatory activity of this fraction was then tested on TPA-induced ear edema in mice, and we found that the active fraction could inhibit edema by 46.9%. The anti-inflammatory effect of the fraction was also tested on carrageenan-induced paw edema in rats at doses of 100 mg/kg; a58.9±2.8% reduction of the edema was observed 4 h after administration of carrageenan, and the effect was maintained for 5 h.

  Objective: This study aimed to investigate the anti-inflammatory activity of methanol extract and fractions of bacteria associated with sponge (Haliclona amboinensis) and to evaluate their effect in reducing NO production and inhibiting cyclooxygenase-1 (COX-1), cyclooxgenase-2 (COX-2) and secretory phospholipase A2 (sPLA2) activity.Methods: All bacterial isolates were cultured and supernatants were collected for the extraction of secondary metabolites using diaion HP-20 to obtain methanol extracts. Evaluation of cytotoxicity property was carried out on macrophage cell lines (RAW264.7) by 3-(4,5-dimethylthiazol- 2-yl) 2,5-diphenyl tetrazoliumbromide assay. Anti-inflammatory screening was done by inducible nitric oxide assay on RAW264.7 cell lines with lipopolysaccharide (LPS) stimulation. Dianion HP-20 was used to remove salt content. A selected methanol extract was subjected to further fractionations by C-18 reverse phase and their anti-inflammatory potential was evaluated by COX-1 and COX-2, and sPLA2 enzymatic assay.Results: Seven methanol extracts showed no cytotoxic property against RAW 264.7 cell line (inhibitory concentration 50% > 30 μg/ml) and selected for anti-inflammatory screening assay. Result showed methanol extract HM 1.2 reduced NO production >80% and it has been selected for phytochemical screening, further fractionations and assay. Phytochemical screening showed alkaloids and terpenoids present in the HM 1.2. The HM 1.2 and its fractions (F1, F2, F1C1, F1C2, F1C3, and F1C4) were proven to inhibit COX-1, COX-2, and sPLA2 activity in the range of 60.516-116.886%, 20.554- 116.457%, and 70.2667-114.8148%, respectively.Conclusions: This study revealed that bacteria associated with H. amboinensis have produced anti-inflammatory activity via reducing NO production and inhibiting COX-1, COX-2, and sPLA2 activity.

Objective: The objective of this study was to determine the anti-inflammatory activity of methanol extract of Niebuhria apetala and its possible mechanism of action. Methods: Methanol extract of Niebuhria apetala leaf (NAL) was assessed for its anti-inflammatory activity by in vitro methods. Using albumin denaturation assay, proteinase inhibitory activity, membrane stabilization, and antilipoxygenase activity at different concentrations, in vitro anti-inflammatory activity was estimated. The standard drug used for this purpose was aspirin. Results: Methanol extract NAL at a concentration range of 100–500 μg/ml significant (p<0.01) protects the heat-induced protein denaturation. At the concentration of 500 mg/ml, NAL showed significant (p<0.01) inhibition of protease inhibitory action. Heat-induced hemolysis of erythrocyte, hypotonicity-induced hemolysis, and lipooxygenase activity were significant (p<0.01) inhibited at the concentration of 500 μg/ml. Conclusion: Finally, the present study indicates that methanol extract of Niebuhria apetala can be a potential source of anti-inflammatory agent.

Objective: Sarcostemma brunonianum Wight and Arn is a potential medicinal plant belonging to Asclepiadaceae. Bioactive constituents of the plant support the application of treating various ailments in the traditional system of medicine. The study aims to determine the presence of various phytoconstituents in stem, root, and flowers. Methods: Hot percolation method was carried out to obtain crude extracts using different solvent systems from low polar-high polar solvents ranging from petroleum ether, chloroform, (mid-polar) ethyl acetate, methanol and water. Estimation of total phenols, tannins and In vitro antioxidant, anti-inflammatory activities were evaluated for the determination of potential pharmaceutical drugs. Results: The results revealed the presence of some phytoconstituents such as phenols, tannins, glycosides, gums and mucilages. Ascorbic acid, BSA, Rutin and Gallic acid were used as the reference standard. The total phenolic content was found to be high in stem methanol extract 440.84±69.99 mg/g Gallic acid equivalent, whereas the tannin content was 291.78±4.68 mg/g GAE. The result proves that the S. brunonianum stem methanol extract possesses antioxidant and anti-inflammatory activities when compared to reference standards. In vitro, Nitric oxide scavenging activity of stem showed a maximum % of inhibition in methanol stem extract (24.39µg/ml) and anti-inflammatory activity maximum inhibition was found to be (55.56 %) in stem methanol and flower(53.62 %). The IC50 (concentration required for 50% inhibition) was also calculated for the DPPH radical model. Conclusion: This study results proclaims and justifies the role of folklore medicinal plant S. brunonianum in the treatment of inflammatory-related ailments and can be recommended for an effective drug.

Objective: This current investigation assesses in vivo central and peripheral analgesic effects and anti-inflammatory properties of fractions obtained from Galphimia glauca (GG) stem methanol extract. Methods: The laboratory models such as Swiss albino mice and Wistar albino rats were employed in the studies. The GG stem methanol extract was subjected to fractionation with solvents such as hexane, chloroform, ethyl acetate, and methanol. Orally, the dose range of 100, 200, and 400 mg/kg was given for 1 day for evaluating analgesic (hotplate test, tail clip test, writhing test, and formalin test) and weekdays for assessing anti-inflammatory activity (carrageenan and cotton pellet test methods), respectively. The experimental studies were further conducted for determining the involvement of central and peripheral receptor actions in the analgesic activity of the extract by prechallenging it with naloxone and acetic acid, respectively. The in vivo anti-inflammatory studies were conducted using carrageenan-induced rat paw edema model and cotton pellet granuloma test. Results: The LD50 of the extract was found to be >2000 mg/kg b.w. The methanol fraction of 400 mg/kg dose exhibited significant (p≤0.001) and dose-dependent analgesic and anti-inflammatory activity. It also exhibited central and peripheral analgesic actions when treated with naloxone and acetic acid, respectively. Conclusion: The results revealed that the stem methanol fraction has more potential in terms of analgesic and anti-inflammatory properties.

Objective: In the current research, to determine the stem extract of Meliosma simplicifolia (L.) for total phenol, tannin, total flavonoid, antioxidant activity, anti-inflammatory and identify the phytoconstituents utilizing GC-MS and FT-IR. Methods: The ability of the plant extract to act as hydrogen/electrons donor or scavenger of radicals was determined by in vitro antioxidant assays using 2,2-diphenyl-2-picryl-hydrazyl free radical (DPPH) scavenging, reducing power assay, superoxide radical (O2•) scavenging activity. The anti-inflammatory activity was evaluated using carrageenan and formalin-induced paw edema models using Wistar albino rats. The GC-MS and FT-IR analysis of the methanolic stem extract of M. simplicifolia was revealed the presence of phytochemicals. Results: Quantitative studies of estimated phenol, flavonoid and tannin, as for the methanol extract of stem showed the highest content of phenolic compounds (39.83±3.62GAE mg/100). Antioxidant activities were concluded the estimation M. simplicifolia stem for as followed the studies. In stem the methanol extract showed the highest DPPH scavenging activity (124.3µg/ml). The anti-inflammatory activity has shown in high doses of methanolic extract 250 mg/kg of significant value (p<0.05) inhibition of paw edema, on 6th hour, respectively. The FT-IR analysis has confirmed their characteristic peak values and functional groups. Conclusion: M. simplicifolia has an effective of anti-inflammatory activity and constitutes a potential source for the development of new treatments.
