Electron crystallography of membrane phospholipids

Membrane phospholipids are notoriously difficult to crystallize to sample sizes suitable for collection of single crystal X-ray intensity data, accounting for the relatively few crystal structures which have been reported to date. At the 24th Annual EMSA meeting, Parsons and Nyburg suggested that the enhanced scattering of electrons by matter be exploited to obtain single crystal electron diffraction patterns from the more readily available microcrystalline preparations, an example of which was shown in a subsequent paper. For almost 20 years ve have investigated the utility of such intensity data for ab initio crystal structure analysis.

The crystal structure of Na2S2O6·2 H2O has been refined by means of single crystal X-ray intensity data. The structure of the isotypic Li2S2O6·2 H20 was also determined by single crystal studies. Two dimensional differential FOURIER synthesis revealed the approximate positions of the hydrogen atoms. From 1H-NMR investigations, the crystal structure, and information gained by IR spectroscopy the complete atomic arrangement in these two substances was obtained. The influence of the dynamical behaviour of the water molecules is taken into account in determining the hydrogen positions.

Selenourea reacts with dialkyl acetylenedicarboxylates under solvent-free conditions to form 1:1 adducts, which undergo a cyclization reaction to produce alkyl Z-2-(2-amino-4-oxo-1,3-selenazol- 5(4H)-ylidene)acetates, in good yields. The stereochemistry of the ethyl Z-2-(2-amino-4-oxo-1,3- selenazol-5(4H)-ylidene)acetate was established by X-ray single crystal structure analysis. The reaction is completely stereoselective.

In the presence of Na2CO3, the conjugate addition between α-halogeno hydrazones and nitroso compounds proceeded readily, thus delivering multifunctionalized nitrones in the reasonable chemical yields with excellent strereoselectivities. The chemical structure and stereochemical configuration of title chemical entities were unambiguously identified by an X-ray single crystal structure analysis.
