Low phytic acid (LPA) soybean [Glycine max (L.) Merr] genotypes reduce indigestible PA in soybean seeds in order to improve feeding efficiency of mono- and agastric animals, but often exhibit low field emergence, resulting in reduced yield. In this study, four LPA soybean varieties with two different genetic backgrounds were studied to assess their emergence and yield characters under 12 seed treatment combinations including two broad-spectrum, preplant fungicides (i.e., ApronMaxx (mefenoxam: (R,S)-2-[(2,6-dimethylphenyl)-methoxyacetylamino]-propionic acid methyl ester; fludioxonil: 4-(2,2-difluoro-1,3-benzodioxol-4-yl)-1H-pyrrole-3-carbonitrile) and Rancona Summit (ipconazole: 2-[(4-chlorophenyl)methyl]-5-(1-methylethyl)-1-(1H-1,2,4-triazol-1-ylmethyl) cyclopentanol; metalaxyl: N-(methooxyacetyl)-N-(2,6-xylyl)-DL-alaninate)), osmotic priming, and MicroCel-E coating. Two normal-PA (NPA) varieties served as controls. Both irrigated and non-irrigated plots were planted in Blacksburg and Orange, Virginia, USA in 2014 and 2015. Results revealed that three seed treatments (fungicides Rancona Summit and ApronMaxx, as well as Priming + Rancona) significantly improved field emergence by 6.4–11.6% across all genotypes, compared with untreated seeds. Seed priming was negatively associated with emergence across LPA genotypes. Seed treatments did not increase the yield of any genotype. LPA genotypes containing mips or lpa1/lpa2 mutations, produced satisfactory emergence similar to NPA under certain soil and environmental conditions due to the interaction of genotype and environment. Effective seed treatments applied to LPA soybeans along with the successful development of LPA germplasm by soybean breeding programs, will increase use of LPA varieties by commercial soybean growers, ultimately improving animal nutrition while easing environmental impact.

Abstract Phytic acid (myo-inositol-1,2,3,4,5,6-hexakisphosphate), the major storage form of phosphorus in cereals, is considered as an antinutrient in food and feed. During the past few years, various cereals have been subjected to mutation breeding for generating low phytic acid (lpa) crops. Recently, it was demonstrated that reduction of phytic acid in the rice mutant Os-lpa-MH86-1 obtained by gamma irradiation was due to a disruption of OsSULTR3;3, an orthologue of the sulfate transporter family group 3 genes. The application of a GC/MS-based metabolite profiling approach revealed that the reduction of phytic acid was accompanied by changes in concentrations of metabolites from different classes in the Os-lpa-MH86-1 mutant.Lpa mutant lines often exhibit lower grain yield and seed viability compared with their wild-type parents. To improve the agronomic performance of the Os-lpa-MH86-1 mutant, cross-breeding with a commercial cultivar was performed. The resulting progenies were genotyped using molecular markers to identify homozygous wildtype and lpa mutants from generations F4 to F7. The objectives of this study were: (i) to observe the consistent metabolic changes in Os-lpa-MH86-1 lpa mutants by following their composition over several independent field trials; (ii) to investigate the impact of cross-breeding on the phytic acid content and the metabolic phenotype of the homozygous lpa mutant; and (iii) to assess the stability of the mutation-specific metabolite signature in the lpa progenies over several generations. Statistical assessment of the data via multivariate and univariate approaches demonstrated that the lpa trait and the mutation-induced metabolite signature in the lpa progenies were comparable to the progenitor Os-lpa-MH86-1 mutant and consistently expressed over generations. These findings extend the basis for implementing mutation breeding in the generation of lpa rice cultivars.

The legume seed coat is the site of sucrose unloading and the metabolism of imported ureides and synthesis of amino acids for the developing embryo. The cell types directly responsible for these functions in the seed coat are not known. We recently described a convex layer of tissue on the inside surface of the soybean (Glycine max L. Merr.) seed coat that was termed “antipit” because it was in direct opposition to the concave pit on the abaxial surface of the cotyledon. Cone cells of the antipit contained numerous hypertrophied Golgi apparatus and laminated rough endoplasmic reticulum common to actively secreting cells. The initial report by Dzikowski (1936) described the morphology of the pit and antipit in G. max and found these structures in only 68 of the 169 seed accessions examined.

Penelitian tentang pengaruh pemberian Pupuk organik cair (POC) asal limbah tumbuhan terhadap serapan hara N dan P serta produksi tanaman kedelai (Glycine max (L) Merrill) di lahan gambut telah dilaksanakan di kampus II Unisi Fakultas Pertanian Jl. Lintas Propinsi Parit 01, Desa Pulau Palas, Kecamatan Tembilahan Hulu, Kabupaten Indragiri Hilir Propinsi Riau. Dimulai dari bulan Agustus sampai bulan Oktober 2013. Tujuan dari penelitian ini untuk mendapatkan POC asal limbah tumbuhan yang terbaik untuk serapan hara N dan P serta produksi tanaman kedelai di lahan gambut. Penelitian ini menggunakan rancangan acak kelompok (RAK) faktor tunggal dengan 7 perlakuan dan 4 ulangan, 2 tanaman dijadikan sampel. Perlakuan dosis POC limbah tanaman pisang dan POC limbah sayur kol yang diberikan yaitu 0 L/Ha, 200 L/Ha, 400 L/Ha dan 600 L/Ha. Parameter pengamatan yaitu : serapan hara N dan P pada fase awal generatif, tinggi tanaman, jumlah bintil akar, polong hampa, produksi perplot, berat 100 biji dan brangkasan kering. Data pengamatan dianalisis dengan sidik ragam (ANOVA) dan dilanjutkan dengan Uji Lanjut Tukey HSD pada taraf 5%. Berdasarkan penelitian yang telah dilaksanakan dapat disimpulkan bahwa pemberian POC asal limbah tumbuhan tidak berpengaruh nyata terhadap serapan hara N dan P, tinggi tanaman, jumlah bintil akar, polong hampa, brangkasan kering tetapi berpengaruh nyata terhadap produksi perplot dan berat 100 biji.
