During seed germination, desiccation tolerance is lost in the radicle with progressing radicle protrusion and seedling establishment. This process is accompanied by comprehensive changes in the metabolome and proteome. Germination of Arabidopsis seeds was investigated over 72 h with special focus on the heat-stable proteome including late embryogenesis abundant (LEA) proteins together with changes in primary metabolites. Six metabolites in dry seeds known to be important for seed longevity decreased during germination and seedling establishment, while all other metabolites increased simultaneously with activation of growth and development. Thermo-stable proteins were associated with a multitude of biological processes. In the heat-stable proteome, a relatively similar proportion of fully ordered and fully intrinsically disordered proteins (IDP) was discovered. Highly disordered proteins were found to be associated with functional categories development, protein, RNA and stress. As expected, the majority of LEA proteins decreased during germination and seedling establishment. However, four germination-specific dehydrins were identified, not present in dry seeds. A network analysis of proteins, metabolites and amino acids generated during the course of germination revealed a highly connected LEA protein network.

Seeds from 55 populations of native Alberta red-osier dogwood (Cornus stolonifera Michx.) were collected over 2 years (1984 and 1985) to determine the range of variability for seed germination and the effect of ecoregion and moisture conditions in the collection sites on this trait. Each year the unscarified seeds were stored at 3 °C for 30 days, stratified at 3 °C for an additional 30 days and then incubated on an 8 h light: 16 h dark cycle at a temperature of 25:10 °C (light:dark) for germination. A wide range of germination percentages was observed among populations collected in any 1 year. However, the variability was not due to the ecoregion or moisture condition of the collection site. Germination percentage of populations collected in 1984 and 1985 formed five and seven groups, respectively. A study involving eight sites that were common to 1984, 1985, and 1986 revealed a significant effect of population on seed germination. This population effect was not obscured by the combined effects of year, precipitation, temperature, and seed size. The year effect was significant only when population was used as a covariate. Precipitation during seed formation significantly influenced germination performance even after adjustments were made for population and year. Seed germination in red-osier dogwood appears to be influenced by the population from which the seeds are collected and by yearly environmental fluctuations. Therefore, germination studies in this species must include seeds from diverse populations collected over several years. Specific knowledge about the dormancy, germination, and emergence patterns of weed species aids the development of integrated management strategies. Laboratory studies were conducted to determine the effect of several environmental factors on seed germination and seedling emergence of Cyperus difformis. Germination of freshly harvested seeds was inhibited by darkness; however, when seeds were subsequently transferred to complete light they germinated readily. Our results showed that 2 wk of cold stratification overcome the light requirement for germination. Seeds of C. difformis were able to germinate over a broad range of temperatures (25/15, 30/20, 35/25, and 40/30 ºC day/night). The response of germination rate to temperature was described as a non-linear function. Based on model outputs, the base, the optimum and the ceiling temperatures were estimated as 14.81, 37.72 and 45 ºC, respectively. A temperature of 120 ºC for a 5 min was required to inhibit 50% of maximum germination. The osmotic potential and salinity required for 50% inhibition of maximum germination were -0.47 MPa and 135.57 mM, respectively. High percentage of seed germination (89%) was observed at pH=6 and decreased to 12% at alkaline medium (pH 9) pH. Seeds sown on the soil surface gave the greatest percentage of seedling emergence, and no seedlings emerged from seeds buried in soil at depths of 1 cm.

Important factors affecting seed priming have not been extensively reported in muskmelon (Cucumis melo L.) studies. The optimization of the seed priming technique becomes very important at the commercial scale. Little information has been reported on seedling development of muskmelon subsequent to seed priming. Seeds of muskmelon were primed in darkness at 25°C in different solutions and three osmotic potentials. Seeds were also primed with and without aeration during different periods. In relation to osmotic solutions, an osmotic potential around -1.30 MPa is most adequate for muskmelon priming. Salt solutions gave better germination rate but were deleterious for seed germination, especially at higher osmotic potentials. Aeration of the soaking salt solution gave faster germination at 17°C, and because of the early germination, these treatments probably presented a better seedling development. Deleterious effect on total seed germination was observed for long soaking periods with aeration. Fungal growth increased on seeds primed in aerated solutions. Seeds from priming treatments had a better germination rate and seedling development under 17 and 25°C.
