Ulva and Enteromorpha (Ulvales, Ulvophyceae, Chlorophyta) in eastern Australia: Comparison of morphological features and analyses of nuclear rDNA sequence data

Specimens of Ulva Linnaeus and Enteromorpha Link (Ulvales, Ulvophyceae, Chlorophyta) were collected from 12 sites in eastern Australia and identified to species level on the basis of morphological characters described in the major Australian study of Womersley (1984). The species recognised were Ulva australis Areschoug, U. lactuca Linnaeus, U. fasciata Delile, Enteromorpha compressa (Linnaeus) Greville, E. flexuosa (Wulfen ex Roth) J.Agardh and E. intestinalis (Linnaeus) Link. Species placement within the genera Ulva and Enteromorpha is problematic and features of DNA were examined in order to assess their potential use as characters in species determinations. Analyses were conducted on sequence data derived from the internal transcribed spacer region ITS2, and the 5.8S gene. Groupings of the 12 isolates based on DNA analyses do not correlate with the species identified using morphology nor with the two genera. Further work is required before authoritative conclusions can be reached regarding the significance of morphological plasticity in determination of differences within and between Ulva and Enteromorpha species; nevertheless, DNA studies may provide a backbone of characters upon which to base such a study.

Wrightoporia subavellanea sp. nov. is described and illustrated from southern China on the basis of morphological characters and molecular data. It is characterized by an annual growth habit, resupinate basidiocarps with white rhizomorphs, large pores, narrow and strongly dextrinoid skeletal hyphae, broadly ellipsoid to subglobose, thick-walled, finely asperulate and strongly amyloid basidiospores (3.8–4.2 × 2.8–3.2 µm), and presence of gloeoplerous hyphae in trama. A molecular study based the combined ITS (internal transcribed spacer region) and nLSU (the large nuclear ribosomal RNA subunit) dataset supported the position of the new species in Wrightoporia. A key to accepted species of Wrightoporia in China is provided.

Anopheles (Nyssorhynchus) oswaldoi (Peryassú, 1922) comprises a species complex in South America. To fully characterize other taxa within the Oswaldoi Complex, it is essential to fix the identity of the nominotypical member. Given that the no type was designated in the original description, a lectotype is formally designated from the remaining syntypes in the Museu Nacional do Rio de Janeiro. These and other recently collected specimens from the type locality (Espírito Santo, Brazil) and the State of São Paulo, Brazil were used to redescribe the species using morphological characters of the adult female, male and male genitalia, and the fourth-instar larva and pupa. The larva, pupa, and male genitalia are illustrated. Diagnostic morphological characters of the adult female and male genitalia are provided to distinguish An. oswaldoi s.s. from the morphologically similar An. konderi, An. galvaoi, and An. ininii. DNA sequence data from the second nuclear internal transcribed spacer region (ITS2) are included to fix the molecular identity of An. oswaldoi s.s.

Two new rust species, Chrysomyxa diebuensis and C. zhuoniensis, on Picea asperata are recognized by morphological characters and DNA sequence data. A detailed description, illustrations, and discussion concerning morphologically similar and phylogenetically closely related species are provided for each species. From light and scanning electron microscopy observations C. diebuensis is characterized by the nailhead to peltate aeciospores, with separated stilt-like base. C. zhuoniensis differs from other known Chrysomyxa species in the annulate aeciospores with distinct longitudinal smooth cap at ends of spores, as well as with a broken, fissured edge. Analysis based on internal transcribed spacer region (ITS) partial gene sequences reveals that the two species cluster as a highly supported group in the phylogenetic trees. Correlations between the morphological and phylogenetic features are discussed. Illustrations and a detailed description are also provided for the aecia of C. succinea in China for the first time.

AbstractThe lithobiomorph centipede Henicops is widely distributed in Australia and New Zealand, with five described species, as well as two species in New Caledonia and Lord Howe Island. Parsimony, maximum likelihood and Bayesian analyses of ca. 800 aligned bases of sequence data from 16S rRNA and 28S rRNA were conducted on a dataset including multiple individuals of Henicops species from populations sampled from different parts of species' geographic ranges, together with the allied henicopines Lamyctes and Easonobius. Morphological characters are included in parsimony analyses. Molecular and combined datasets unite species from eastern Australia and New Zealand to the exclusion of species from Western Australia, New Caledonia and Lord Howe Island. The molecular data favour these two geographic groupings as clades, whereas inclusion of morphology resolves New Caledonia, Lord Howe Island, southwest Western Australia and Queensland as successive sisters to southeastern Australia and New Zealand. The basal position of the Lord Howe Island species in the phylogeny favours a diversification of Australasian Henicops since the late Miocene unless the Lord Howe species originated in a biota that pre-dates the island. The molecular and combined data resolve the widespread morphospecies H. maculatus as paraphyletic, with its populations contributing to the geographic groupings New South Wales + New Zealand and Tasmania + Victoria.

AbstractThe phylogenetic relationships of the genus Bryoria were examined using ITS, partial glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and mitochondrial SSU rDNA sequence data in addition to 20 chemical and morphological characters. This first comprehensive molecular study to assess Bryoria phylogeny includes representatives from all the traditionally recognized four sections. Combined cladistic analyses of 88 Bryoria specimens representing at least 25 species resulted in highly resolved phylogenies. Based on the results, a new infrageneric classification for the genus is proposed. Five sections are recognized, largely corresponding to the existing classification, with the addition of section Americanae. The study shows that while most species with an erect growth-form are clearly monophyletic, current species status of many pendent taxa can be questioned.

ABSTRACT The genus Bakawan includes species of haminoeid snails associated with mangrove habitats and mud flats in the Indo-West Pacific. Here, we revise the diversity and systematics of the genus Bakawan based on our recent molecular phylogeny (Oskars & Malaquias, 2019) and detailed analysis of morphological characters. We examined a range of morphological characters (the shells, external morphology of the animal, jaw, radula, gizzard plate and male reproductive system) using light and scanning electron microscopy. We also carried out a species delimitation analysis (we used the automatic barcode gap discovery method) based on cytochrome c oxidase subunit I DNA sequence data. Four distinct species were recognized: Bakawan rotundata (A. Adams, 1850), which is the type species of the genus and ranges from the eastern Indian Ocean to the western Pacific; B. fusca (A. Adams, 1850), currently known only from the Philippines; and two species new to science, B. puti n. sp., known only from the Philippines, and B. hedleyi n. sp., restricted to tropical eastern Australia.

Biscutella L. ser. Biscutella (= Biscutella ser. Lyratae Malin.) comprises mostly annual or short-lived perennial plants occurring in the Mediterranean basin and the Middle East, which exhibit some diagnostic floral features. Taxa in the series have considerable morphological plasticity, which is not well correlated with clear geographic or ecologic patterns. Traditional taxonomic accounts have focused on a number of vegetative and floral characters that have proved to be highly variable, a fact that contributed to taxonomic inflation mostly in northern Africa. A detailed study and re-evaluation of morphological characters, together with recent phylogenetic data based on concatenation of two plastid and one nuclear region sequence data, yielded the basis for a taxonomic reappraisal of the series. In this respect, a new comprehensive integrative taxonomic arrangement for Biscutella ser. Biscutella is presented in which 10 taxa are accepted, namely seven species and three additional varieties. The name B. eriocarpa DC. is reinterpreted and suggested to include the highest morphological variation found in northern Morocco. Its treatment here accepts two varieties, one of which is described as new (B. eriocarpa var. riphaea A. Vicente, M. Á. Alonso & M. B. Crespo). In addition, the circumscriptions of several species, such as B. boetica Boiss. & Reut., B. didyma L., B. lyrata L., and B. maritima Ten., are revisited. Nomenclatural types, synonymy, brief descriptions, cytogenetic data, conservation status, distribution maps, and identification keys are included for the accepted taxa, with seven lectotypes and one epitype being designated here.

The genus Celtis L. (Cannabaceae) comprises around 73 species distributed in temperate and tropical regions of both hemispheres. Most South American species belong to the subgenus Mertensia Planch., and they are characterized by showing high morphological plasticity. A recent taxonomic revision of this group was carried out with an excessively reductionist criterion regarding the number of accepted species and that does not clearly reflect the morphological differences among them. In order to contribute to the correct delimitation of the specific entities, a morpho-descriptive approach is presented for ten taxa of the genus Celtis (C. brasiliensis, C. chichape, C. clausseniana, C. fluminensis, C. pallida var. pallida, C pallida var. discolor, C. serratissima, C. spinosa, C. spinosissima and C. tala) growing in South American Southern Cone. The morphology was analyzed through the use of a stereomicroscope and/or the tpsDig program for digital images of specimens. As a result, a key is provided for the determination of the aforementioned species and a detailed morphological description of the vegetative (habit, bark, branches, spines, brachyblasts, indumentum and leaves) and reproductive structures (inflorescences, flowers, fruits and endocarp -pyrene-). In addition, a specimen collection guide is proposed with a list of the main morphological characters that should be observed in the field, to facilitate future identifications.

A phylogenetic analysis of Acacia subg. Phyllodineae sect. Botrycephalae, endemic to eastern Australia, is presented based on a combined dataset of ITS and ETS sequences of nrDNA. A smaller set of species was sequenced also for the cpDNA trnK region. A limited number of morphological characters was also combined with the ITS+ETS dataset for most taxa. Thirty-eight of 41 Botrycephalae species were sequenced, together with a sample of ten uninerved phyllodinous species (sect. Phyllodineae). Although these DNA regions showed limited sequence divergence, bootstrap supported nodes of the consensus ITS+ETS tree indicate that Botrycephalae as currently defined is polyphyletic. Eight bipinnate species fell outside the main clade of Botrycephalae species while seven phyllodinous species were nested within it, near the base. The few derived but homoplasious morphological characters that were discovered included: presence of appressed unicellular hairs, presence of jugary and interjugary glands, number of pinnae > 7 and the funicle half–fully encircling the seed. Section Botrycephalae requires redefinition.

The morphology and morphogenesis of the stylonychine hypotrich Rigidohymena candens (Kahl, 1932) Berger, 2011, isolated from garden soil in Qingdao, China, were investigated using live observation and protargol impregnation methods. The Qingdao isolate possesses all diagnostic morphological characters of R. candens. The main events during binary fission are as follows: (i) the proter retains the parental adoral zone of membranelles entirely, whereas the old undulating membranes dedifferentiate into an anlage that gives rise to the leftmost frontal cirrus and the new undulating membranes of the proter; (ii) five streaks of fronto-ventral-transverse cirral anlagen are segmented in the pattern 3 : 3 : 3 : 4 : 4 from left to right, which form two frontal, four frontoventral, one buccal, five ventral and five transverse cirri, respectively; (iii) dorsal morphogenesis is in the typical Oxytricha pattern; (iv) three caudal cirri are formed, one at the posterior end of each of dorsal kineties 1, 2 and 4; and (v) the postoral ventral cirrus V/3 is not involved in primordia formation. The morphological and morphogenetic observations and phylogenetic analyses based on the small-subunit rDNA sequence data support the validity of Rigidohymena Berger, 2011 and its systematic position in the subfamily Stylonychinae.
