Dollar spot is one of the most destructive diseases in turfgrass. The causal agents belong to the genus Clarireedia, which are known for causing necrotic, sunken spots in turfgrass that coalesce into large damaged areas. In low tolerance settings like turfgrass, it is of vital importance to rapidly detect and identify the pathogens. There are a few methods available to identify the genus Clarireedia, but none of those are rapid enough and characterize down to the species level. This study produced a co-dominant cleaved amplified polymorphic sequences (CAPS) test that differentiates between C. jacksonii and C. monteithiana, the two species that cause dollar spot disease within the United States. The calmodulin gene (CaM) was targeted to generate Clarireedia spp. specific PCR primers. The CAPS assay was optimized and tested for specificity and sensitivity using DNA extracted from pure cultures of two Clarireedia spp. and other closely related fungal species. The results showed that the newly developed primer set could amplify both species and was highly sensitive as it detected DNA concentrations as low as 0.005 ng/µL. The assay was further validated using direct PCR to speed up the diagnosis process. This drastically reduces the time needed to identify the dollar spot pathogens. The resulting assay could be used throughout turfgrass settings for a rapid and precise identification method in the US.

Abstract The dioecious white campion Silene latifolia (syn. Melandrium album) has heteromorphic sex chromosomes, XX in females and XY in males, that are larger than the autosomes and enable their separation by flow sorting. The group of MROS genes, the first male-specifically expressed genes in dioecious plants, was recently identified in S. latifolia. To localize the MROS genes, we used the flow-sorted X chromosomes and autosomes as a template for PCR with internal primers. Our results indicate that the MROS3 gene is located in at least two copies tandemly arranged on the X chromosome with additional copy(ies) on the autosome(s), while MROS1, MROS2, and MROS4 are exclusively autosomal. The specificity of PCR products was checked by digestion with a restriction enzyme or reamplification using nested primers. Homology search of databases has shown the presence of five MROS3 homologues in A. thaliana, four of them arranged in two tandems, each consisting of two copies. We conclude that MROS3 is a low-copy gene family, connected with the proper pollen development, which is present not only in dioecious but also in other dicot plant species.

The plant-parasitic genus Microbotryum (Pucciniomycotina) has been used as a model for various biological studies, but fundamental aspects of its life history have not been documented in detail. The smut fungus is characterized by a dimorphic life cycle with a haploid saprophytic yeast-like stage and a dikaryotic plant-parasitic stage, which bears the teliospores as dispersal agents. In this study, seedlings and flowers of Silene latifolia Poir. (Caryophyllaceae) were inoculated with teliospores or sporidial cells of Microbotryum lychnidis-dioicae (DC. ex Liro) G. Deml & Oberw. and the germination of teliospores, the infection process, and the proliferation in the host tissue were documented in vivo using light and electron microscopy. Although germination of the teliospore is crucial for the establishment of Microbotryum, basidium development is variable under natural conditions. In flowers, where the amount of nutrients is thought to be high, the fungus propagates as sporidia, and mating of compatible cells takes place only when flowers are withering and nutrients are decreasing. On cotyledons (i.e., nutrient-depleted conditions), conjugation occurs shortly after teliospore germination, often via intrapromycelial mating. After formation of an infectious hypha with an appressorium, the invasion of the host occurs by direct penetration of the epidermis. While the growth in the plant is typically intercellular, long distance proliferation seems mediated through xylem tracheary elements. At the beginning of the vegetation period, fungal cells were found between meristematic shoot host cells, indicating a dormant phase inside the plant. By using different microscopy techniques, many life stages of Microbotryum are illustrated for the first time, thereby allowing new interpretations of laboratory data.

Silene latifolia is a model dioecious plant with morphologically distinguishable XY sex chromosomes. The end of the Xq arm is quite different from that of the Yp arm, although both are located at opposite ends of their respective chromosomes relative to a pseudo-autosomal region. The Xq arm does not seem to originate from the same autosome as the Yp arm. Bacterial artificial chromosome clone #15B12 has an insert containing a 130-kb stretch in which a 313-bp satellite DNA is repeated 420 times. PCR with a single primer revealed that this 130-kb stretch consists of three reversals of the orientation of the satellite DNA. A non-long terminal repeat retroelement and two sequences that share homology with an Oryza sativa RING zinc finger and a putative Arabidopsis thaliana protein, respectively, were found in the sequences that flank the satellite DNA. Fluorescence in situ hybridization carried out using this low-copy region of #15B12 as a probe confirmed that these sequences originated from the X chromosome and that homologous sequences exist at the end of chromosome 7. The region distal to DD44X on the Xq arm is postulated to have recombined with a region containing satellite DNA on chromosome 7 during the process of sex chromosome evolution.
