Abstract This retrospective study aimed to determine if the number of cows exposed per bull affects pregnancy rates of cows returning to estrus after fixed-time artificial insemination (FTAI). Data were compiled over the course of 13 breeding seasons (6 fall and 7 spring seasons) between 2010 and 2017 from the Virginia Department of Corrections herd. Available records contained data from 17 farms and 324 groups of cows (average 47 cows/group). Multiparous cows and heifers (average age per group: 5.11 ± 0.14 years; n = 14,868) were exposed to FTAI. After FTAI, animals were placed on pasture with bulls diagnosed as fertile by a breeding soundness exam for natural service of cows who did not become pregnant to FTAI (n = 7,248; average 22 cows/group). Animals were classified as pregnant to FTAI, to natural service on first return to estrus, or to natural service on second or subsequent estrus determined by fetal aging at pregnancy diagnosis. The bull:cow ratio for the total number of cows exposed ranged from 1:9 to 1:73 with an average of 1:31. The bull:cow ratio considering only open cows exposed after FTAI ranged from 1:2 to 1:44 with an average of 1:14. There was significant negative, small correlation between the bull:cow ratio for total number of cows exposed and return to estrus pregnancy rate in fall breeding seasons (P = 0.01, r 2 = 0.04) but not in spring (P = 0.90). There was a significant negative, small correlation between bull:cow ratio of open cows exposed and pregnancy rates to first return to estrus in fall herds with a single sire (P < 0.001, r 2 = 0.11). There was no correlation in fall herds using multiple sires or spring herds (P ≥ 0.12). Bull:cow ratio accounted for only 1 – 11% of variation in the pregnancy rates, thus we conclude that a decreased bull:cow ratio (up to 1:73) did not affect natural service return to estrus pregnancy rate. Cattlemen may consider a reduced number of bulls needed for natural service breeding after FTAI, which can decrease bull related costs and increase the economic feasibility of adopting FTAI protocols.

There was no correlation between the bull:cow ratio and return to estrus pregnancy rate (r = -0.12), and there was no correlation between the number of open cows per bull and return to estrus conception rates (r = -0.17). Thus, higher bull:cow ratios (between 1:8 and 1:68) did not effect return to estrus pregnancy rate; producers may consider a lower number of bulls needed for natural service breeding post TAI, decreasing bull related costs and increasing the economic feasibility of adopting TAI protocols.

Abstract Clitoral stimulation after artificial insemination (AI) in beef cattle is a common practice utilized by AI technicians; however, the effect of clitoral stimulation during fixed-time AI (TAI) is still unknown. The objective of the present experiment was to evaluate the effect of clitoral stimulation on temperament responses and pregnancy rates to TAI in Bos indicus beef cows. A total of 1,186 multiparous Nellore cows across three different locations were assigned to an estradiol/progesterone-based estrus synchronization + TAI protocol (day −11 to 0). Cows were randomly assigned to receive either 3 s of clitoral stimulation (n = 602) or no clitoral stimulation (n = 584) immediately after TAI (day 0). Cow body condition score (BCS) was recorded on day −11. Estrus expression was evaluated based on estrus detection visual aid patch activation on day 0 (estrus, ≥50% activated; no estrus, <50% activated). Temperament was assessed by individual chute score based on a 5-point scale before TAI, and individual exit velocity was measured after clitoral stimulation. Pregnancy diagnosis was performed 30 d after TAI via transrectal ultrasonography. Pregnancy rate to TAI was positively affected by BCS (P < 0.01) and estrus expression (P = 0.03). Pregnancy rates of cows receiving clitoral stimulation did not differ (P = 0.39) from cows non-stimulated (47.5 ± 4.6% vs. 44.3 ± 4.6%, respectively). No interaction of clitoral stimulation and estrus expression was observed (P = 0.26). Chute score was positively correlated to exit velocity (P < 0.01; r = 0.29); however, clitoral stimulation did not affect exit velocity (P = 0.86). In summary, pregnancy rates to TAI are influenced by various factors and multiple strategies have the potential to increase the fertility of beef cows submitted to TAI; however, clitoral stimulation of Bos indicus beef cows did not improve TAI pregnancy rate.

AbstractThe objective was to evaluate different oestrous synchronization schemes in beef cattle under range conditions. In experiment 1, 202 heifers averaging 26 months of age were assigned randomly to three treatments: (a) GnRH-PG. (no. = 44) day 0 (D0): injection of GnRH; D7: injection of PGF2α; D0 to D25 oestrous detection (OD) and artificial insemination (AI); (b) OD-PG. (no. = 45) D0 to D4; OD + AI; D5: injection of PGF2α; D5 to D25 OD + AI; and (c)Ovsynch. (no. = 113) D0: injection of GnRH; D7: injection of PGF2α; D9: injection of GnRH and 16 h later AI at fixed time. In experiment 2, 318 non-suckling cows were assigned randomly to three treatments: (a) GnRH-PG. (no. = 106) D0: injection of GnRH; D7: injection of PGF2α; D0 to D25 OD + AI; (b) GnRH-MAP-PG. (no. = 106) D0: injection of a GnRH analogue and insertion of an intravaginal sponge impregnated with 250 mg of medroxyprogesterone acetate; D7: injection of PGF2α and sponge withdrawal; D0 to D25 OD + AI; (c) controls. (no. = 106) OD and AI. The experimental period lasted 25 days and, with exception of the Ovsynch treatment, oestrous detection was carried out twice a day (a.m./p.m.) and inseminations performed 12 h later. The pregnancy rates assessed by ultrasonography 30 days after AI were: experiment 1: (a) 30·0%; (b) 28·6% and (c) 62·3% (P < 0·05); experiment 2: (a) 60·2%; (b) 57·8% and (c) 45·5% (P < 0·05). It is concluded that in heifers and non-suckling cows, oestrous synchronization treatments result in higher pregnancy rates. In non-suckling cows, the addition of a progestagen did not improve the response.

The objective of this trial was to evaluate different post-timed artificial insemination (TAI) reproductive managements in postpartum beef cows to produce crossbred calves from artificial insemination (AI). Nellore cows (n = 607), with 45 days postpartum, were inseminated at a fixed time, using a protocol that included an intravaginal progesterone-releasing device along with oestradiol benzoate, prostaglandin, equine chorionic gonadotropin, and oestradiol cypionate, followed TAI 48 h post-device removal. Four post-TAI treatments were evaluated: in CONTROL (T1, n = 161), cows were exposed to Nellore clean-up bulls until the end of the breeding season (75 days). In OBSERVATION (T2, n = 132), heat detection was performed for 15–25 days post-TAI, followed by AI. In RESYNC22 (T3, n = 157) and RESYNC30 (T4, n = 157), resynchronisation started after 22 or 30 days, following second TAI at Day 32 or 40 days after first TAI. In T2, T3 and T4, after the second AI, cows were exposed to Nellore clean-up bulls until the end of the breeding season (75 days). The pregnancy rate (PR) for the first TAI did not differ (54.6%, 53.0%, 59.2%, and 51.6% for CONTROL, OBSERVATION, RESYNC 22, and RESYNC 30, respectively; P = 0.66), and no difference was observed for the second TAI (RESYNC 22 = 45.31% and RESYNC30 = 46.05%; P = 0.137), in the PR at the end of the breeding season (86.33%, 86.36%, 78.98%, and 81.52%, P = 0.43), or embryonic losses (4.54%, 2.85%, 6.45% and 7.40%, respectively; P = 0.61), but the percentage of crossbred pregnancy was higher in groups with resynchronisation (RESYNC22 and RESYNC30) than CONTROL and OBSERVATION (98.38%, 90.62%, 63.30%, 78.95%, P &lt; 0.001). In conclusion, resynchronisation programs of 22 or 30 days are more efficient to produce AI products, and the final pregnancy rate is similar among the treatments, differing only in the amount of calves produced by AI.

The aim of this study was to investigate the effects of hormonal support on the pregnancy rate in repeat breeder cows. Prostaglandin F2α + Ovsynch oestrus synchronization protocol was applied to the cows. Following the fixed time insemination (day 0), the cows were divided into 4 groups. In Group 1 (n = 42), progesterone releasing intravaginal device (PRID) was placed vaginally at 84 h and removed on the 9th day after the artificial insemination. In Group 2 (n = 40), the cows were administered human chorionic gonadotropin (hCG) on the 7th day. Group 3 (n = 45) was applied a combination of progesterone and hCG. Group 4 (n = 42) was not given any treatment. Blood samples were collected from all cows 4 times on days 3.5, 7, 12, and 18 to evaluate progesterone concentrations. The pregnancy rates were 40.47%, 37.50%, 44.44%, and 30.95% in Group 1, 2, 3, and 4, respectively (P > 0.05). In addition, in cows with progesterone concentrations <2 ng/ml on day 3.5, the pregnancy rates were found to be lower than in the cows with progesterone concentrations >2 ng/ml in Group 4 (P < 0.05). Progesterone supplementation in cows with progesterone concentrations < 2 ng/ml appeared to increase pregnancy rates (P < 0.05) in Groups 1 and 3. As a result, post-insemination hormonal applications in the repeat breeder cows did not increase the pregnancy rate. However, it was concluded that determination of progesterone concentrations on day 3.5 following artificial insemination and then hormonal support in the cows with low concentrations would increased the pregnancy rate.

Abstract The objective of this study was to assess how well estrus detection patch readings correlated to successful AI breedings and to characterize the within-cow repeatability of estrus detection patch readings across breeding seasons. Data from the Virginia Department of Corrections beef cattle herds were collated for 7 locations over 7 years, with 2 calving seasons per year. Data from Spring of 2011 were missing. The full dataset contained 19,253 individual animal observations, of which 2,389 observations were omitted for failure to report estrus detection patch data. The relationship between estrus patch reading and pregnancy rate to fixed-time AI and subsequent natural service pregnancy rate was assessed by calculating the sensitivity and specificity of patch readings within each location during each breeding season. A true positive was defined as a patch activating and a cow being confirmed pregnant. A true negative was a patch remaining in the non activated state and a cow being confirmed open. A false positive was defined as a patch activating and a cow being confirmed open. A false negative was a patch failing to activate and a cow being confirmed pregnant. Pregnancy rate to AI sensitivity ranged from 0.2 to 1 with a mean of 0.583. Specificity ranged from 0 to 1 with a mean of 0.525. Subsequent natural service pregnancy rate), ranges in sensitivity (0.197 to 1.00, mean 0.563) and specificity (0 to 1, mean 0.545) were similar. Of the 6,249 animals with usable patch data, 934 never presented with an activated patch and 2,064 presented with an activated patch every breeding season. The remaining 3,251 animals averaged presenting with an activated patch 47% of the time and a non-activated patch 46% of the time. Odds of getting pregnant by AI for cows that always flag with the heat patch were 0.415.

Successful establishment of pregnancy in the cow depends on early rise of progesterone (P4) to improve embryo development. The present study was carried out to determine the corpus luteum (CL) function and pregnancy rate following the administration of 1.500 IU of hCG in suckled beef cows on Day 4 after fixed-time AI (FTAI). Crossbred Angus cows (n = 429), 40 to 70 days postpartum, were synchronized using 2 mg of oestradiol benzoate (EB) i.m. and an intravaginal progesterone-releasing device containing 1 g of P4 for 7 days. At device removal, prostaglandin F2a (150 mg of D-cloprostenol) was injected i.m.; 24 h later, 1 mg of EB was given and the FTAI were done 30 to 36 h after EB using semen of just 1 bull. Cows were assigned alternatively to receive 1.500 IU of hCG i.m. (n = 200) or 1 mL of saline i.m. (n = 229) on Day 4 after FTAI. Ovaries were examined by transrectal ultrasonography (n = 71) on Days 4 and 7 to detect the presence and to measure the area of CL. Blood samples were collected on Days 4, 7, 10, 14, and 30 post-oestrus to determine P4 concentration by RIA. Pregnancy diagnoses were done 30 days after FTAI. The effects of treatments on CL area and serum P4 levels were analysed by ANOVA (Proc GLM, SAS Institute Inc., Cary, NC, USA) and pregnancy rates compared by chi-square (Proc CATMOT, SAS). The induction of accessory CL determined on Day 7 was 44% in cows treated with hCG and not observed in the control group. The area of the CL on Day 7 was significantly higher (P < 0.01; 3.37 ± 2.56 v. 2.56 ± 0.22 cm2) in the hCG group. Despite the increase in CL size, the mean levels of P4 on Days 4, 7, 10, 14, and 30 post-AI indicated no difference (P > 0.05) in CL function between control and hCG-treated cows. On Day 30, pregnancy rates were not different (P = 0.08), at 46.7 and 55% for control and hCG-treated cows, respectively. In conclusion, administration of 1.500 IU of hCG on Day 4 post-FTAI did not improve pregnancy rate in suckled beef cows.

Abstract Advancements in reproductive technologies have improved pregnancy rates in postpartum cows; however, undesirable consequences leading to reduced fertility still occur. Thus, the objectives were to determine if presence of a corpus luteum (CL) at the start of estrous synchronization (AIM1) or if the length of proestrus (interval from progesterone removal to a GnRH-induced LH surge; AIM2) alters fixed timed artificial insemination (FTAI) pregnancy rates. Cycling postpartum cows (n = 285) at two locations were synchronized using the CO-Synch+CIDR® FTAI protocol. Ultrasonographic measurements of luteal area were recorded at CIDR insertion. To determine the impact of proestrus length, cows were randomly assigned to either undergo CIDR removal at the time of PGF2a injection (long proestrus, n = 145), or CIDR removal 24 hours post PGF2a injection (short proestrus, n = 140). Blood samples were collected at CIDR insertion and at insemination for progesterone (P4) analysis. Pregnancy status to FTAI was assessed using ultrasonographic diagnosis at 30 d post insemination. Data were analyzed using a MIXED procedure of SAS and examined for effects of length of proestrus, P4 concentration, luteal area, sire, location, and age. Statistical significance was determined at P0.05) pregnancy rates. Pregnancy rates were greater in cows with >1 ng/mL P4 compared to cows with 0.05). Cows > 4 y old had higher AI pregnancy rates compared to 2–3 y cows (70.2±7.4 vs. 51±5.8%, respectively; P > 0.05). Based on these data, incorporating a presynchronization program to ensure the presence of a functional CL at the start of a FTAI protocol may improve pregnancy success in postpartum beef cows.

The objective of this work was to evaluate the effect of insulin alone or in association with equine chorionic gonadotropin (eCG) on the fertility of postpartum beef cows subjected to synchronization. A total of 340 cows was subjected to fixed time artificial insemination. In the trial 1, the cows were subjected to temporary weaning (TW), while in the trial 2 the same protocol was tested without TW. The addition of an insulin injection to a progesterone/eCG-based protocol without TW increased the pregnancy rate of beef cows with body condition score (BCS) equal to or lower than 2.5. Insulin had no effect on cows submitted to TW or with BCS equal to or higher than 3.0.
