Efficiency of mating, artificial insemination or resynchronisation at different times after first timed artificial insemination in postpartum Nellore cows to produce crossbred calves

The objective of this trial was to evaluate different post-timed artificial insemination (TAI) reproductive managements in postpartum beef cows to produce crossbred calves from artificial insemination (AI). Nellore cows (n = 607), with 45 days postpartum, were inseminated at a fixed time, using a protocol that included an intravaginal progesterone-releasing device along with oestradiol benzoate, prostaglandin, equine chorionic gonadotropin, and oestradiol cypionate, followed TAI 48 h post-device removal. Four post-TAI treatments were evaluated: in CONTROL (T1, n = 161), cows were exposed to Nellore clean-up bulls until the end of the breeding season (75 days). In OBSERVATION (T2, n = 132), heat detection was performed for 15–25 days post-TAI, followed by AI. In RESYNC22 (T3, n = 157) and RESYNC30 (T4, n = 157), resynchronisation started after 22 or 30 days, following second TAI at Day 32 or 40 days after first TAI. In T2, T3 and T4, after the second AI, cows were exposed to Nellore clean-up bulls until the end of the breeding season (75 days). The pregnancy rate (PR) for the first TAI did not differ (54.6%, 53.0%, 59.2%, and 51.6% for CONTROL, OBSERVATION, RESYNC 22, and RESYNC 30, respectively; P = 0.66), and no difference was observed for the second TAI (RESYNC 22 = 45.31% and RESYNC30 = 46.05%; P = 0.137), in the PR at the end of the breeding season (86.33%, 86.36%, 78.98%, and 81.52%, P = 0.43), or embryonic losses (4.54%, 2.85%, 6.45% and 7.40%, respectively; P = 0.61), but the percentage of crossbred pregnancy was higher in groups with resynchronisation (RESYNC22 and RESYNC30) than CONTROL and OBSERVATION (98.38%, 90.62%, 63.30%, 78.95%, P < 0.001). In conclusion, resynchronisation programs of 22 or 30 days are more efficient to produce AI products, and the final pregnancy rate is similar among the treatments, differing only in the amount of calves produced by AI.

The objective of this work was to evaluate the effect of insulin alone or in association with equine chorionic gonadotropin (eCG) on the fertility of postpartum beef cows subjected to synchronization. A total of 340 cows was subjected to fixed time artificial insemination. In the trial 1, the cows were subjected to temporary weaning (TW), while in the trial 2 the same protocol was tested without TW. The addition of an insulin injection to a progesterone/eCG-based protocol without TW increased the pregnancy rate of beef cows with body condition score (BCS) equal to or lower than 2.5. Insulin had no effect on cows submitted to TW or with BCS equal to or higher than 3.0.

This study evaluated the efficiency application of low dose of equine chorionic gonadotropin (eCG) using Hou Hai acupoint for fixed-term artificial insemination (FTAI) in beef cows. Seventy cows received intravaginal devices with progesterone, and 3 mg of estradiol benzoate on day zero (D0) of FTAI. On D9, the devices were removed, and 150 ?g of prostaglandin F2? was applied; the animals were then randomly distributed to three treatment groups: T1 (n = 23), 300 IU of eCG intramuscularly (IM); T2 (n = 23), 90 IU of eCG at acupoint Hou Hai; and T3 (n = 25), 90 IU of eCG at false acupoint IM. On D10, the animals received 1 mg of gonadotropin-releasing hormone IM, and FTAI was performed, 52 h after the devices were removed. The quantitative variables with normal distribution were assessed using analysis of variance at 5% probability. For variables that did not show normal distribution, Kruskal-Wallis test at 5% probability was used. No significant difference was noted among the groups (P > 0.05) for the interval between device withdrawal at ovulation (58.2 ± 12.2 h), diameter of the largest follicle on D9 (9.9 ± 2.2 mm), diameters of the ovulatory follicle (12.2 ± 3.0 mm) and the second largest follicle (6.7 ± 2.1 mm), follicular growth rate (0.8 ± 0.3 mm/d), ovulation rate (82%), corpus luteum size (2.32 ± 0.35 cm2), and pregnancy rate (58.67%). The protocol cost per animal was US$ 10.67 (T1) and US$ 8.50 (T2 and T3). The use of 90 IU of eCG applied at Hou Hai acupoint or at false acupoint caused satisfactory synchronization of estrus in beef cows. In addition, this procedure was cost-effective.

The objective of this study was to compare the effects of 1 and 2 prostaglandin doses in a timed AI (TAI) resynchronization protocol on the pregnancy rate. This study was performed during the 2017-2018 breeding season on 10 commercial farms located in the central and southwest region of Rio Grande do Sul, Brazil. A total of 2347Bos taurus beef cows (Angus, Hereford) with a body condition score 2.90±0.15 (1=thin, 5=obese) and 40±5.1 days postpartum were used. In the beginning of first synchronization TAI protocol, on Day −11, the animals received 2mg of oestradiol benzoate IM (Gonadiol®, Zoetis Saúde Animal, São Paulo, SP, Brazil) and a progesterone (P4) intravaginal device (CIDR®, Zoetis). On Day −4, the cows received 12.5mg of dinoprost tromethamine IM (PGF, Lutalyse®, Zoetis). On Day −2, the animals received 1.0mg of oestradiol cypionate IM (ECP®, Zoetis) associated with the treatment with 300IU of eCG IM (Novormon®, Zoetis) and the P4 device was removed. The TAI was done on Day 0 (48 h after P4 device removal). The resynchronization protocol was started on Day 23 without pregnancy or oestrus detection; all cows were randomly assigned to receive a new, once-used, or twice-used P4 device (CIDR®) and given 2mg of oestradiol benzoate. On Day 30, pregnancy diagnosis was performed with transrectal ultrasonography. Pregnant cows (n=1021) had the P4 device withdrawn and received no further treatment. Nonpregnant cows (n=1326) were randomly assigned into 2 groups. The 1PGF group (n=674) received a single dose of PGF on Day 30, and the 2PGF group (n=652) received PGF on Days 30 and 32. On Day 32, all cows received eCG and ECP, and the P4 device was removed. In both groups the TAI was performed 48h after the P4 device removal (Day 34). Pregnancy diagnosis was performed by ultrasound 30 days after TAI (Day 64 of the breeding season). Data were analysed using the GLIMMIX procedure of SAS (SAS Institute Inc., Cary, NC, USA). The pregnancy rate obtained in the first synchronization protocol was 43.5% (1021/2347). The pregnancy rate in the resynchronization protocol was similar (P=0.49) between PGF doses [1PGF=39.5% (266/674); 2PGF=40.6% (265/652). No interactions were observed between P4 device use and PGF dose (P=0.38). The pregnancy rates were similar when comparing 1PGF and 2PGF using a new P4 device [1PGF=43.2% (35/81); 2PGF=56% (93/166); P=0.67], once-used P4 device [1PGF=39% (163/418); 2PGF=40.6% (140/345); P=0.64], or twice-used P4 device [1PGF=38.9% (68/175); 2PGF=22.7% (32/141); P=0.14]. In conclusion, a second dose of prostaglandin in the resynchronization protocol was not necessary to achieve high resynchronization pregnancy rates with TAI in Bos taurus beef cows.

The aim of this study was to investigate the effects of hormonal support on the pregnancy rate in repeat breeder cows. Prostaglandin F2α + Ovsynch oestrus synchronization protocol was applied to the cows. Following the fixed time insemination (day 0), the cows were divided into 4 groups. In Group 1 (n = 42), progesterone releasing intravaginal device (PRID) was placed vaginally at 84 h and removed on the 9th day after the artificial insemination. In Group 2 (n = 40), the cows were administered human chorionic gonadotropin (hCG) on the 7th day. Group 3 (n = 45) was applied a combination of progesterone and hCG. Group 4 (n = 42) was not given any treatment. Blood samples were collected from all cows 4 times on days 3.5, 7, 12, and 18 to evaluate progesterone concentrations. The pregnancy rates were 40.47%, 37.50%, 44.44%, and 30.95% in Group 1, 2, 3, and 4, respectively (P > 0.05). In addition, in cows with progesterone concentrations <2 ng/ml on day 3.5, the pregnancy rates were found to be lower than in the cows with progesterone concentrations >2 ng/ml in Group 4 (P < 0.05). Progesterone supplementation in cows with progesterone concentrations < 2 ng/ml appeared to increase pregnancy rates (P < 0.05) in Groups 1 and 3. As a result, post-insemination hormonal applications in the repeat breeder cows did not increase the pregnancy rate. However, it was concluded that determination of progesterone concentrations on day 3.5 following artificial insemination and then hormonal support in the cows with low concentrations would increased the pregnancy rate.

We have previously shown that expression of oestrus significantly influences the time of ovulation and pregnancy rates to timed-AI (P/TAI) in beef cattle synchronized with progesterone devices and oestradiol (Bó et al. 2016 Theriogenology 86, 388–396). Furthermore, delaying TAI from 48 h to 54–58 h after device removal improved P/TAI of those animals not showing oestrus by 48 h. The objective of the present study was to determine whether the administration of GnRH to those animals not showing oestrus by 48 h after device removal had an impact on P/TAI. Non-lactating and suckled beef cows and heifers (Bonsmara, Brangus, and Braford; n = 868), with a corpus luteum (CL) or a follicle ≥8 mm in diameter detected by ultrasonography (Mindray DP 30 Vet, 7.5 MHz, China) and body condition score 2 to 4 (1 to 5 scale) were synchronized in 3 replicates. On Day 0, animals received a progesterone device (DIB 0.5 g, Zoetis, Argentina) and 2 mg of oestradiol benzoate (Von Franken, Argentina). On Day 8, DIB were removed and cows received 0.150 mg of D+cloprostenol (Río de Janeiro, Argentina) plus 0.5 mg oestradiol cypionate (Cipiosyn, Zoetis). In addition, all cows were tail-painted on the sacrococcygeal area (CeloTest, Biotay, Argentina) at DIB removal and were observed to determine the expression of oestrus, according to the percentage of paint loss (PL) 48 h later. The animals that showed oestrus (PL > 30%) were recorded and TAI at that time while those with PL ≤ 30% (not in oestrus) were randomly allocated to receive 100 μg of gonadorelin (GnRH; Gonasyn, Zoetis) or no treatment and TAI between 54 to 58 h after DIB removal. Pregnancy was diagnosed by ultrasonography at 45 days after TAI, and data were analysed by logistic regression. There were no significant differences among replicates, inseminators, semen source or animal class (dry cows, suckled cows or heifers; P > 0.11). However, animals showing oestrus by 48 h after DIB removal had higher P/TAI (371/666; 55.7; P < 0.04) than those that did not show oestrus (96/202; 47.5%). Within animals not showing oestrus at 48 h, those that were in oestrus by the TAI at 54 to 58 h had higher P/TAI (71/124, 57.3%; P < 0.01) than those that did not show oestrus within that period (27/78; 34.6%). In addition, P/TAI was higher (P < 0.01) in animals that received GnRH (66/113; 58.4%) than in those that did not receive GnRH (30/89; 33.7%). In conclusion, the expression of oestrus significantly affected P/TAI in beef cattle synchronized with progesterone devices and oestradiol. Furthermore, treatment of animals not showing oestrus by 48 h after DIB removal with GnRH resulted in improved P/TAI performed 6 to 10 h later.

Three experiments were conducted to determine the effects of presynchronization and treatment with equine chorionic gonadotropin (eCG) on corpus luteum (CL) and ovarian follicular development, plasma progesterone concentrations, and pregnancy rates in beef heifers subjected to a gonadotropin releasing hormone (GnRH)-based, fixed-timed AI (TAI) protocol. All heifers were given GnRH on day 0, prostaglandin F2α (PGF) on day 7, and a second GnRH on day 9 concurrent with TAI (54 h after PGF). In exp. 1 (N = 148), presynchronization with PGF (days -22 and -11) decreased the percentage of heifers with non-luteal plasma progesterone concentrations on day 0 (5.4 vs 29.7%) and day 7 (0 vs 11.6%; P < 0.05), but not on day 9 (74.3 vs. 66.2%; P > 0.20), and reduced the number of heifers in estrus and bred before TAI (P < 0.05). Although presynchronization reduced preovulatory follicle diameter (12.9 ± 0.3 vs. 14.9 ± 0.3 mm; mean ± SEM; P < 0.01), it did not affect TAI pregnancy rates (36.5 vs. 29.7%; P > 0.20). In exp. 2, heifers (N = 128) were presynchronized with melengestrol acetate (MGA) (days -27 to -12), and received a controlled internal drug release (CIDR) on day 0; on day 7, half were given 300 IU of eCG at CIDR removal. Treatment with eCG tended to increase preovulatory follicle diameter in heifers that did not ovulate to GnRH on day 0 (P = 0.06), but did not affect the percentage of heifers with non-luteal plasma progesterone concentrations on day 9 (57.8 vs. 57.8%) or TAI pregnancy rates (48.4 vs. 53.1%; P > 0.20). Experiment 3 was a 2 × 2 factorial arrangement of presynchronization (PGF concurrent with a CIDR on day -7) and eCG treatments (on day 7) applied to heifers in three herds (A, N = 150, B, N = 260 and C, N = 40). All heifers had a once-used CIDR from days 0 to 7. Presynchronization increased the percentage of heifers (Herd A) with low-luteal plasma progesterone concentrations on day 0 (70.7 vs. 22.7%) and day 7 (90.7 vs. 53.3%; P < 0.01), but did not affect the percentage of heifers with non-luteal concentrations of progesterone on day 9 (97.3 vs. 93.3%; P > 0.20). Combined for all herds, presynchronization reduced the prevalence of a CL on day 0 (23.5 vs. 73.7%; P < 0.01), and increased the prevalence of follicles ≥ 10 mm on day 7 (96.8 vs. 86.7%; P < 0.01); however, TAI pregnancy rates (195/439 = 44.4%) were not improved by either presynchronization or eCG treatment (P > 0.20).Key words: Presynchronization, equine chorionic gonadotropin, GnRH, fixed-time artificial insemination, progesterone
