The aim of this study was to investigate the effects of hormonal support on the pregnancy rate in repeat breeder cows. Prostaglandin F2α + Ovsynch oestrus synchronization protocol was applied to the cows. Following the fixed time insemination (day 0), the cows were divided into 4 groups. In Group 1 (n = 42), progesterone releasing intravaginal device (PRID) was placed vaginally at 84 h and removed on the 9th day after the artificial insemination. In Group 2 (n = 40), the cows were administered human chorionic gonadotropin (hCG) on the 7th day. Group 3 (n = 45) was applied a combination of progesterone and hCG. Group 4 (n = 42) was not given any treatment. Blood samples were collected from all cows 4 times on days 3.5, 7, 12, and 18 to evaluate progesterone concentrations. The pregnancy rates were 40.47%, 37.50%, 44.44%, and 30.95% in Group 1, 2, 3, and 4, respectively (P > 0.05). In addition, in cows with progesterone concentrations <2 ng/ml on day 3.5, the pregnancy rates were found to be lower than in the cows with progesterone concentrations >2 ng/ml in Group 4 (P < 0.05). Progesterone supplementation in cows with progesterone concentrations < 2 ng/ml appeared to increase pregnancy rates (P < 0.05) in Groups 1 and 3. As a result, post-insemination hormonal applications in the repeat breeder cows did not increase the pregnancy rate. However, it was concluded that determination of progesterone concentrations on day 3.5 following artificial insemination and then hormonal support in the cows with low concentrations would increased the pregnancy rate.

This study focused on determining the success of inducing endogenous progesterone source (by formation of accessory corpus luteum) by human chorionic gonadotropin (hCG) administration on day 4 post timed artificial insemination (TAI) and on evaluating the effect of the day of hCG administration (day 4 or 5 post TAI) on pregnancy rates in repeat breeder cows according to their responses to hCG administrations. Non-pregnant cyclic lactating Holstein dairy cows with more than three services despite no clinical reproductive abnormalities were enrolled in the study. All cows (n = 192) received the Ovsynch protocol (GnRH-7d-PGF2α-56h-GnRH-16/18h-TAI) and hCG administration (1500 IU, i.m.) was performed in group hCG4 (n = 64) on day 4 and in group hCG5 (n = 68) on day 5 post TAI. The control group (CON, n = 60) did not receive any hCG treatment. The responses (ovulation rate) to hCG (~64%) were similar on days 4 and 5 post TAI. Pregnancy per AI (P/AI) at 31 days was similar among the groups (48.3% in CON, 48.4% in hCG4, and 42.6% in hCG5). Regardless of the day of the treatment (hCG4 + hCG5), the cows had an ovulatory response to hCG (54.1%), and a greater P/AI (P = 0.007) at 31 days than those without a response (29.8%). The administration of hCG on either day 4 or 5 post TAI did not increase overall pregnancy rates, however, the ovulatory response to the post TAI hCG administration on day 4 improved fertility in repeat breeder cows. Further studies are needed with a larger number of animals to reveal this potentiallly beneficial effect.

The aim of the present study was to determine the effects of one or two doses of prostaglandin F2α (PGF) on the pregnancy rate (PR) in Simmental cows subjected to the Ovsynch or Ovsynch + Controlled internal drug release (CIDR) protocols. On d 0, 100 µg gonadorelin diacetate tetrahydrate (GnRH) was injected into cows in group 1 (n = 50) and group 2 (n = 40) and CIDR was intravaginally inserted. On d 7, 25 mg dinoprost tromethamine (PGF2α, PGF) was administered, and GnRH was injected 56 h later. Fixed-time artificial insemination (TAI) was performed 16 h after GnRH injection. In group 3 (n = 62) and group 4 (n = 63), PGF was administered 7 d following GnRH treatment, and GnRH was injected 56 h later. TAI was performed 16 h after GnRH injection. In groups 2 and 4, a second dose of PGF was injected 24 h after the first dose. Pregnancy was determined by transrectal ultrasonography on d 30 and 60 post-TAI. The pregnancy rate (PR) on d 30 post-TAI was significantly different among the groups: 46%, 55%, 29%, and 36.5%, in groups 1, 2, 3, and 4, respectively (P = 0.048). In addition, the PR was 36.3% and 43.7% in the single PGF and double PGF injection groups, respectively. There was no statistical difference in the PR on d 60 post-TAI (P > 0.05). Pregnancy loss was between 4.34% and 11.11% (P > 0.05). The highest PR in the primiparous cows was observed in group 3 (57.7%). The PR was not significantly different among multiparous cows (P > 0.05). In conclusion, two injections of PGF administered 24 h apart in Simmental cows may increase the PR in the Ovsynch or Ovsynch + CIDR protocols.

The objective of this trial was to evaluate different post-timed artificial insemination (TAI) reproductive managements in postpartum beef cows to produce crossbred calves from artificial insemination (AI). Nellore cows (n = 607), with 45 days postpartum, were inseminated at a fixed time, using a protocol that included an intravaginal progesterone-releasing device along with oestradiol benzoate, prostaglandin, equine chorionic gonadotropin, and oestradiol cypionate, followed TAI 48 h post-device removal. Four post-TAI treatments were evaluated: in CONTROL (T1, n = 161), cows were exposed to Nellore clean-up bulls until the end of the breeding season (75 days). In OBSERVATION (T2, n = 132), heat detection was performed for 15–25 days post-TAI, followed by AI. In RESYNC22 (T3, n = 157) and RESYNC30 (T4, n = 157), resynchronisation started after 22 or 30 days, following second TAI at Day 32 or 40 days after first TAI. In T2, T3 and T4, after the second AI, cows were exposed to Nellore clean-up bulls until the end of the breeding season (75 days). The pregnancy rate (PR) for the first TAI did not differ (54.6%, 53.0%, 59.2%, and 51.6% for CONTROL, OBSERVATION, RESYNC 22, and RESYNC 30, respectively; P = 0.66), and no difference was observed for the second TAI (RESYNC 22 = 45.31% and RESYNC30 = 46.05%; P = 0.137), in the PR at the end of the breeding season (86.33%, 86.36%, 78.98%, and 81.52%, P = 0.43), or embryonic losses (4.54%, 2.85%, 6.45% and 7.40%, respectively; P = 0.61), but the percentage of crossbred pregnancy was higher in groups with resynchronisation (RESYNC22 and RESYNC30) than CONTROL and OBSERVATION (98.38%, 90.62%, 63.30%, 78.95%, P &lt; 0.001). In conclusion, resynchronisation programs of 22 or 30 days are more efficient to produce AI products, and the final pregnancy rate is similar among the treatments, differing only in the amount of calves produced by AI.

Three experiments were conducted to determine the effects of presynchronization and treatment with equine chorionic gonadotropin (eCG) on corpus luteum (CL) and ovarian follicular development, plasma progesterone concentrations, and pregnancy rates in beef heifers subjected to a gonadotropin releasing hormone (GnRH)-based, fixed-timed AI (TAI) protocol. All heifers were given GnRH on day 0, prostaglandin F2α (PGF) on day 7, and a second GnRH on day 9 concurrent with TAI (54 h after PGF). In exp. 1 (N = 148), presynchronization with PGF (days -22 and -11) decreased the percentage of heifers with non-luteal plasma progesterone concentrations on day 0 (5.4 vs 29.7%) and day 7 (0 vs 11.6%; P < 0.05), but not on day 9 (74.3 vs. 66.2%; P > 0.20), and reduced the number of heifers in estrus and bred before TAI (P < 0.05). Although presynchronization reduced preovulatory follicle diameter (12.9 ± 0.3 vs. 14.9 ± 0.3 mm; mean ± SEM; P < 0.01), it did not affect TAI pregnancy rates (36.5 vs. 29.7%; P > 0.20). In exp. 2, heifers (N = 128) were presynchronized with melengestrol acetate (MGA) (days -27 to -12), and received a controlled internal drug release (CIDR) on day 0; on day 7, half were given 300 IU of eCG at CIDR removal. Treatment with eCG tended to increase preovulatory follicle diameter in heifers that did not ovulate to GnRH on day 0 (P = 0.06), but did not affect the percentage of heifers with non-luteal plasma progesterone concentrations on day 9 (57.8 vs. 57.8%) or TAI pregnancy rates (48.4 vs. 53.1%; P > 0.20). Experiment 3 was a 2 × 2 factorial arrangement of presynchronization (PGF concurrent with a CIDR on day -7) and eCG treatments (on day 7) applied to heifers in three herds (A, N = 150, B, N = 260 and C, N = 40). All heifers had a once-used CIDR from days 0 to 7. Presynchronization increased the percentage of heifers (Herd A) with low-luteal plasma progesterone concentrations on day 0 (70.7 vs. 22.7%) and day 7 (90.7 vs. 53.3%; P < 0.01), but did not affect the percentage of heifers with non-luteal concentrations of progesterone on day 9 (97.3 vs. 93.3%; P > 0.20). Combined for all herds, presynchronization reduced the prevalence of a CL on day 0 (23.5 vs. 73.7%; P < 0.01), and increased the prevalence of follicles ≥ 10 mm on day 7 (96.8 vs. 86.7%; P < 0.01); however, TAI pregnancy rates (195/439 = 44.4%) were not improved by either presynchronization or eCG treatment (P > 0.20).Key words: Presynchronization, equine chorionic gonadotropin, GnRH, fixed-time artificial insemination, progesterone

The objective of this work was to evaluate the effect of insulin alone or in association with equine chorionic gonadotropin (eCG) on the fertility of postpartum beef cows subjected to synchronization. A total of 340 cows was subjected to fixed time artificial insemination. In the trial 1, the cows were subjected to temporary weaning (TW), while in the trial 2 the same protocol was tested without TW. The addition of an insulin injection to a progesterone/eCG-based protocol without TW increased the pregnancy rate of beef cows with body condition score (BCS) equal to or lower than 2.5. Insulin had no effect on cows submitted to TW or with BCS equal to or higher than 3.0.

The most important factor in bovine embryo transfer programs is the low efficiency in the utilization of the recipients; this low efficiency is associated with low response to synchronization protocols and failures in estrus detection. It has been shown that cows transferred at fixed time with in vivo-derived embryos resulted in high rates of recipients selected for transfer and high overall pregnancy rates (recipients pregnant/recipients treated) (Tribulo et al. 2002 Theriogenology 57, 563). An experiment was designed to evaluate the pregnancy rate in recipients transferred with in vivo (fresh and frozen), IVF, and cloned-derived embryos without estrus detection. A total of 1555 non-lactating Bos Taurus crossbred beef cows was divided into two groups. Cows from group 1 (n = 421) were synchronized with a progesterone intravaginal releasing device (1 g P4; DIB, Syntex®, Buenos Aires, Argentina) plus 2 mg of estradiol benzoate (EB) i.m. (Syntex®) on Day 0. On Day 5, they received 400 IU of eCG (Novormon 5000, Syntex®) i.m. and 150 μg of D-Cloprostenol (PGF2α) (Bioprost-D, Biotay®, Buenos Aires, Argentina). The DIB devices were removed on Day 8 and on Day 9, 1 mg of EB was injected. Day 10 was arbitrarily considered as the day of estrus. Cows from group 2 (n = 1134) received 2 doses of PGF2α 14 days apart and were checked for heat during 5 days after the second PGF2α dose. Cows of both groups were examined 7 days after estrus by ultrasonography (Pie Medical Scanner 200®) and those with a corpus luteum >10 mm of diameter were transferred nonsurgically with in vivo (fresh and frozen), IVF, and cloned-derived embryos. In group 1, 360 cows were transferred, and in group 2, 726 cows were transferred (Table 1). Pregnancy was diagnosed 23 days later by ultrasonography (Pie Medical Scanner 200®). The pregnancy rates were compared statistically between groups 1 and 2 by analysis of variance (Infostat, LSD Fisher). There was no significant statistic difference (P > 0.05) between pregnancy rate in group 1 and 2 with in vivo (fresh), IVF, and cloned-derived embryos. However, pregnancy rate of frozen in vivo-derived embryos was lower in group 1 than in group 2 (P < 0.05). Results showed that treatment using DIB combined with EB, PGF2α, and eCG associated with embryo transfer without estrus detection (group 1) had no difference in pregnancy rate when compared with the treatment where synchronization with PGF2α and heat detection were used (group 2). Another important advantage is the use the group 1 treatment for increasing the flexibility and efficiency in the management of the recipients of in vivo, IVF, and cloned-derived embryo transfer programs. Abstract Activation of physiological stress responses have been associated with decreased pregnancy rates in beef cattle. Therefore, alternative cattle restraint methods have been developed to reduce this negative impact. However, little evidence exists comparing pregnancy outcomes of differing methods. Thus our objective was to determine if a difference was present in pregnancy rate to fixed-time artificial insemination in Bos taurus beef females when using a breeding box (b) versus a squeeze chute (c). Cows and heifers from two separate locations (Virginia Tech University, VT; and R.A. Brown Ranch, RAB) were exposed to FTAI protocols at location of origin while restrained in a squeeze chute (n = 169) or a breeding box (n = 162). Females were exposed to the 7 day Co-Sync + CIDR protocol pre-breeding. At day of breeding, females were randomly assigned to treatment. Immediately following breeding, blood was collected via coccygeal venipuncture for assessment of plasma cortisol concentration using an automated ELISA assay (Immulite 2000 XPi, Siemens). Chute and Exit Score (1–5) were assessed at the time of breeding by a trained technician at each location. Pregnancy was verified thirty days post-breeding via rectal ultrasonography. Pregnancy rate (PROC FREQ) and cortisol (PROC GLIMMIX) were analyzed using SAS version 9.4. Also, a Linear Regression Model (SAS version 9.4) was used to determine relationships between chute score, exit score, and cortisol. It was found that there was no difference in pregnancy rate between the breeding box and the squeeze chute (61.73% vs 64.71%, P = 0.31, respectively). Cortisol showed no difference between b and c (59.44±10.17 vs 58.61±10.16, P = 0.68, respectively). There was no linear relationship between cortisol, exit score, and chute score (P = 0.14). It can be concluded that stress responses are similar between the breeding box and squeeze chute. Therefore females will have similar pregnancy rates when exposed to FTAI in either restraint mechanism.

Aim: This study aimed to evaluate the pregnancy rates in hair ewes using an Ovsynch synchronization protocol under a breeding system that combines fixed-time insemination plus a 10-day mating period as an alternative. Materials and Methods: Through an experimental study (n=27), ewes were randomly located into one of three treatments: (1) Pre-synch (n=9): Prostaglandin F2α (PGF2α)+Gonadotropin-releasing hormone (GnRH)+PGF2α+GnRH; (2) Ovsynch (n=9): GnRH+PGF2α+GnRH; and (3) control: Ewes bred by natural mating (NM) (n=9). Ewes were fixed-time inseminated (fixed-time artificial insemination [FTAI]) with fresh semen, collected just before the insemination time through vaginoscopy at 16 h after the second GnRH (gonadorelin) injection. Each experimental group was placed separately during 15 days and, after this time, fertile rams were allowed back with ewes for a 10-day mating period. Control group ewes remained with the rest of the herd suitable for breeding and were bred under NM. Pregnancy diagnosis was performed by ultrasound at 28-, 56-, and 84-day post-breeding to differentiate between FTAI and NM pregnancies. Total (FTAI±NM) pregnancy rates at 56-day post-breeding were used to compared Pre-synch, Ovsynch, and control. For this purpose, two-tailed proportions comparison z-test was used with a 95% confidence level, for testing as the null hypothesis whether two proportions were equal. Results: Pregnancy rates were higher in control ewes (66.4%) than FTAI (46.6%). When pregnancy rates after a 10-day mating period (40%) were added, the final rate (86.6%) was significantly (p<0.05) higher in Ovsynch-based protocols. The pregnancy rate was significantly lower in FTAI ewes compared to FTAI +10-day mating group (p<0.05). The overall pregnancy rate was 88.0, 85.7, and 67.0 (p>0.05) for Pre-synch, Ovsynch, and control ewes, respectively. Conclusion: These results provide evidence on the benefits of combined FTAI protocols for improving the reproductive efficiency of sheep.
