Nocardioides szechwanensis sp. nov. and Nocardioides psychrotolerans sp. nov., isolated from a glacier

Two Gram-positive, rod-shaped, non-spore-forming bacteria (strains RHLT1-17T and RHLT2-1T) were isolated from Hailuogou glacier in Szechwan province, PR China. Phylogenetic analysis based on 16S rRNA gene sequences revealed that the strains belonged to the genus Nocardioides and shared 97.8 % sequence similarity with each other and 97.6 and 98.4 % sequence similarity, respectively, with Nocardioides kribbensis KSL-2T. Strain RHLT1-17T grew at 0–35 °C and strain RHLT2-1T grew at 0–25 °C. The major cellular fatty acids of strain RHLT1-17T were C17 : 1ω8c (32.69 %) and iso-C16 : 0 (21.74 %). The major cellular fatty acids of strain RHLT2-1T were C18 : 1ω9c (28.72 %), summed feature 3 (17.14 %; comprising C16 : 1ω7c and/or C16 : 1ω6c), iso-C16 : 0 (14.35 %), C16 : 0 (9.96 %) and iso-C14 : 0 (8.34 %). Both strains contained ll-2,6-diaminopimelic acid in the cell-wall peptidoglycan and MK-8(H4) as the predominant menaquinone. On the basis of data obtained using a polyphasic approach, two novel species, Nocardioides szechwanensis sp. nov. (type strain RHLT1-17T  = CGMCC 1.11147T  = NBRC 108562T) and Nocardioides psychrotolerans sp. nov. (type strain RHLT2-1T  = CGMCC 1.11156T  = NBRC 108563T), are proposed.

A strain of thermotolerant actinomycete, designated YIM 75948T, was isolated from a soil sample in Yunnan province, China. The strain grew at 25–55 °C (optimum 37 °C). The substrate mycelium and aerial mycelium produced on Czapek’s agar were both pale yellow to white. The diagnostic diamino acid of the cell-wall peptidoglycan was meso-diaminopimelic acid, and the whole-cell sugars were mannose, ribose, glucose, galactose and arabinose. The major fatty acids were iso-C15 : 0, iso-C16 : 0 and C16 : 0 and the predominant respiratory quinone was MK-9(H4). The polar lipids consisted of phosphatidylethanolamine, phosphatidylethanolamine with hydroxy fatty acids, diphosphatidylglycerol, phosphatidylinositol and two unidentified phospholipids. The genomic DNA G+C content was 73.8 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain YIM 75948T belonged in the genus Actinokineospora and that its closest relative among recognized species was Actinokineospora fastidiosa DSM 43855T (97.6 % sequence similarity). The mean level of DNA–DNA relatedness between the novel strain and A. fastidiosa DSM 43855T was, however, only 47.8 %. Based on the phenotypic, chemotaxonomic and phylogenetic data and the results of the DNA–DNA hybridizations, strain YIM 75948T represents a novel species of the genus Actinokineospora for which the name Actinokineospora soli sp. nov. is proposed. The type strain is YIM 75948T ( = JCM 17695T = DSM 45613T). The description of the genus Actinokineospora is emended to reflect the fact that the genomic DNA G+C contents of A. fastidiosa DSM 43855T and the type strain of Actinokineospora soli sp. nov. recorded in the present study fell above the range given in previous descriptions of this genus.

Strain HNM0947T, representing a novel actinobacterium, was isolated from the coral Galaxea astreata collected from the coast of Wenchang, Hainan, China. The strain was found to have morphological and chemotaxonomic characteristics consistent with the genus Nocardiopsis . The organism formed abundant fragmented substrate mycelia and aerial mycelia which differentiated into non-motile, rod-shaped spores. Whole-cell hydrolysates contained meso-diaminopimelic acid and no diagnostic sugars. The major menaquinones were MK-10(H8), MK-10(H6) and MK-10(H4). The major phospholipids were phosphatidylcholine, phosphatidylglycerol, phosphatidylinositol and phosphatidylinositol mannosides. The major fatty acids were iso-C16:0, anteiso-C17:0, C18:0, C18:0 10-methyl (TBSA) and anteiso-C15:0. The G+C content was 71.3 mol%. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain HNM0947T belonged to the genus Nocardiopsis and shared highest sequence similarity to Nocardiopsis salina YIM 90010T (98.8%), Nocardiopsis xinjiangensis YIM 90004T(98.5%) and Nocardiopsis kunsanensis DSM 44524T (98.3%). The strain HNM0947T was distinguished from its closest type strain by low average nucleotide identity (90.8%) and dDDH values (60.4%) respectively. Based on genotypic, chemotaxonomic and phenotypic characteristics, it was concluded that strain HNM0947T represents a novel species of the genus Nocardiopsis whose name was proposed as Nocardiopsis coralli sp. nov. The type strain was HNM0947T (=CCTCC AA 2020015 T=KCTC 49525 T).

Two novel actinomycetes, strains TA4-1T and TA4-8T, were isolated from the South-East Asian stingless bee (Tetragonilla collina Smith 1857), collected from Chiang Mai Province, Thailand. The morphological and chemotaxonomic properties of strains TA4-1T and TA4-8T were consistent with the genus Streptomyces , i.e. the formation of aerial mycelia bearing spiral spore chains, the presence of the ll-isomer of diaminopimelic acid in cell walls, iso- and anteiso-branched fatty acids with carbon chain lengths 14–17 atoms as the major fatty acids and MK-9(H8) as the predominant menaquinone plus minor amounts of MK-9(H6) and MK-9(H10). Analysis of 16S rRNA gene sequences showed that strains TA4-1T and TA4-8T exhibited 98.8 and 98.1 % sequence similarity, respectively, with Streptomyces chromofuscus NRRL B-12175T and 98.9 % sequence similarity with each other. This study suggested that strains TA4-1T and TA4-8T were distinct from previously described species of the genus Streptomyces . In addition, the low degrees of DNA–DNA relatedness between the isolates and S. chromofuscus JCM 4354T warranted assigning strains TA4-1T and TA4-8T to two novel species. The names Streptomyces chiangmaiensis sp. nov. (type strain TA4-1T  = JCM 16577T  = TISTR 1981T) and Streptomyces lannensis sp. nov. (type strain TA4-8T  = JCM 16578T  = TISTR 1982T) are proposed. The species names indicate the geographical locations where the stingless bees reside.

A novel filamentous bacterium, designated Nari11AT, was isolated from soil collected from a salt lake named Chott Melghir, located in north-eastern Algeria. The strain is an aerobic, halophilic, thermotolerant, Gram-stain-positive bacterium, growing at NaCl concentrations between 5 and 20 % (w/v) and at 43–60 °C and pH 5.0–10.0. The major fatty acids were iso-C15 : 0, anteiso-C15 : 0 and iso-C17 : 0. The DNA G+C content was 53.4 mol%. ll-Diaminopimelic acid was the diamino acid of the peptidoglycan. The major menaquinone was MK-7, but MK-6 and MK-8 were also present in trace amounts. The polar lipid profile consisted of phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylmonomethylethanolamine and three unidentified phospholipids. Results of molecular and phenotypic analyses led to the description of the strain as a novel member to the genus Melghirimyces , family Thermoactinomycetaceae . Strain Nari11AT shows 96.7 % 16S rRNA gene sequence similarity to the type strain of Melghirimyces algeriensis . On the basis of phenotypic, physiological and phylogenetic data, strain Nari11AT ( = DSM 45514T  = CCUG 60050T) represents the type strain of a novel species, for which the name Melghirimyces thermohalophilus sp. nov. is proposed.

A Gram-stain-negative, aerobic, non-motile and rod-shaped marine bacterium, CW2-9T, was isolated from algae collected from Fujian Province in PR China. 16S rRNA gene sequence analysis showed that this strain was affiliated with the genus Tamlana in the family Flavobacteriaceae of the class Flavobacteriia and was very similar to the type strain Tamlana sedimentorum MCCC 1A10799T (96.3 % sequence similarity). The whole genome of strain CW2-9T comprised 3 997 513 bp with a G+C content of 34.3 mol%. The average nucleotide identity value between strain CW2-9T and T. sedimentorum MCCC 1A10799T was 73.8 %. Growth was observed from 15 to 40 °C (optimum, 30 °C), at pH from pH 5.0 to 10.0 (pH 8.0) and in the presence of 0–4 % (w/v) NaCl (0–1 %). The major fatty acids (>10 % of the total) were iso-C15 : 0, iso G-C15 : 1, iso-C17 : 0 3-OH and anteiso-C15 : 0. The predominant menaquinone was MK-6. The combined phylogenetic, physiological and chemotaxonomic data indicate that strain CW2-9T represents a novel species in the genus Tamlana , for which the name Tamlana fucoidanivorans sp. nov. is proposed. The type strain is CW2-9T (=CICC 24749T=KCTC 72389T).

A Gram-stain-negative, non-spore-forming, aerobic, curved rod-shaped bacterium, designed strain R142T, was isolated from a coralline algae Tricleocarpa sp. in the Beibu Gulf, China. Optimal growth occurred with 0–0.5 % (w/v) NaCl, at 25 °C and at pH 8. Global alignment based on 16S rRNA gene sequences indicated that strain R142T shared 93.8 % similarity with its closest type strain, Pseudomaricurvus alkylphenolicus KU14GT. Phylogenetic analyses showed that strain R142T forms a distinct branch alongside Maricurvus nonylphenolicus KU41ET, Pseudoteredinibacter isoporae SW-11T, Pseudomaricurvus alkylphenolicus KU14GT, Pseudomaricurvus alcaniphilus MEBiC06469T and Aestuariicella hydrocarbonica SM-6T. The major polar lipids of strain R142T were phosphatidylethanolamine and phosphatidylglycerol. The primary cellular fatty acids were C16 : 0, C16 : 1ω7c, C18 : 1ω7c, C18 : 0 and C14 : 0. The genome DNA G+C ratio was 56.4 mol%. The only detected respiratory quinone was ubiquinone 8. The low 16S rRNA gene sequence similarity and differences in cellular fatty acids readily distinguished strain R142T from all validly published type strains. Strain R142T is therefore suggested to represent a novel species of a new genus, for which the name Exilibacterium tricleocarpae gen. nov., sp. nov. is proposed. The type strain of Exilibacterium tricleocarpae is R142T (=MCCC 1K03816T=KCTC 72138T).

Two Gram-stain-negative, aerobic, non-motile bacterial strains, 36D10-4-7T and 30C10-4-7T, were isolated from bark canker tissue of Populus × euramericana, respectively. 16S rRNA gene sequence analysis revealed that strain 36D10-4-7T shows 98.0 % sequence similarity to Sphingomonas adhaesiva DSM 7418T, and strain 30C10-4-7T shows highest sequence similarity to Sphingobacterium arenae H-12T (95.6 %). Average nucleotide identity analysis indicates that strain 36D10-4-7T is a novel member different from recognized species in the genus Sphingomonas . The main fatty acids and respiratory quinone detected in strain 36D10-4-7T are C18 : 1  ω7c and/or C18 : 1  ω6c and Q-10, respectively. The polar lipids are diphosphatidylglycerol, phosphatidylcholine, phosphatidylglycerol, aminolipid, phosphatidylethanolamine, sphingoglycolipid, two uncharacterized phospholipids and two uncharacterized lipids. For strain 30C10-4-7T, the major fatty acids and menaquinone are iso-C15 : 0, C16 : 1  ω7c and/or C16 : 1  ω6c and iso-C17 : 0 3-OH and MK-7, respectively. The polar lipid profile includes phosphatidylethanolamine, phospholipids, two aminophospholipids and six unidentified lipids. Based on phenotypic and genotypic characteristics, these two strains represent two novel species within the genera Sphingomonas and Sphingobacterium . The name Sphingomonas corticis sp. nov. (type strain 36D10-4-7T=CFCC 13112T=KCTC 52799T) and Sphingobacterium corticibacterium sp. nov. (type strain 30C10-4-7T=CFCC 13069T=KCTC 52797T) are proposed.

Two aerobic, Gram-stain-positive, rod-shaped, endospore-forming, thermophilic bacterial strains, designated FJAT-54423T and FJAT-54424, were isolated from hyperthermophilic compost sampled in Shanxi Province, PR China. Growth was observed at 30–60 °C (optimum, 50 °C) and pH 6.0–9.0 (optimum, pH 7.0), with up to 2.0 % (w/v) NaCl (optimum, 0 % NaCl). The 16S rRNA gene sequence similarity between FJAT-54423T and FJAT-54424 was 99.9%, and the maximum similarity to a valid taxon was observed with Brevibacillus borstelensis (98.3%). Further, in phylogenetic and phylogenomic trees, strains FJAT-54423T and FJAT-54424 branched with members of the genus Brevibacillus . The menaquinone was MK-7, and the major fatty acids were iso-C15 : 0 and anteiso-C15 : 0. The main polar lipids included phosphatidylmethylethanolamine, phosphatidylethanolamine, diphosphatidylglycerol and phosphatidylglycerol. The cell-wall peptidoglycan was found to contain meso-diaminopimelic acid. The DNA G+C content of strains FJAT-54423T and FJAT-54424 were 54.3 and 54.4 mol%, respectively. The average nucleotide identity (ANI) and digital DNA–DNA hybridization (dDDH) values of strain FJAT-54423T and its most closely related reference strain B. borstelensis DSM 6347T were 77.7 and 21.5 %, respectively, which were lower than the recommended species delineation thresholds of ANI (95%) and dDDH (70%). Based on the observed physiological properties, chemotaxonomic characteristics and ANI and dDDH values, FJAT-54423T and FJAT-54424 belong to a novel species of the genus Brevibacillus , for which the name Brevibacillus composti sp. nov. is proposed. The type strain is FJAT-54423T (=GDMCC 1.2054T=KCTC 43273T).

Four aerobic, Gram-stain-positive, rod-shaped bacteria (HY60T, HY54, HY82T and HY89) were isolated from bat faeces of Hipposideros and Rousettus species collected in PR China. Phylogenetic analyses based on 16S rRNA gene sequences indicated that the four novel strains formed two separate but adjacent subclades close to Microbacterium agarici CGMCC 1.12260T (97.6–97.7 % similarity), Microbacterium humi JCM 18706T (97.3–97.5 %) and Microbacterium lindanitolerans JCM 30493T (97.3–97.4 %). The 16S rRNA gene sequence similarity was 98.3 % between strains HY60T and HY82T, and identical within strain pairs HY60T/HY54 and HY82T/HY89. The DNA G+C contents of strains HY60T and HY82T were 61.9 and 63.3 mol%, respectively. The digital DNA–DNA hybridization and average nucleotide identity values between each novel strain and their closest relatives were all below the 70 % and 95–96 % thresholds for species delimitation, respectively. All four novel strains contained anteiso-C15 : 0, anteiso-C17 : 0, iso-C16 : 0 and iso-C15 : 0 as the main fatty acids, MK-11 and MK-12 as the major respiratory quinones, and diphosphatidylglycerol, phosphatidylglycerol and one unidentified glycolipid as the predominant polar lipids. The cell-wall peptidoglycan was of B type and contained alanine, glutamate, glycine and ornithine. The acyl type of the muramic acid was glycolyl. The whole-cell sugars were rhamnose and ribose. Based on the foregoing polyphasic analyses, it was concluded that the four uncharacterized strains represented two novel species of the genus Microbacterium , for which the names Microbacterium chengjingii sp. nov. [type strain HY60T (=CGMCC 1.17468T=GDMCC 1.1951T=KACC 22102T)] and Microbacterium fandaimingii sp. nov. [type strain HY82T (=CGMCC 1.17469T=GDMCC 1.1949T=KACC 22101T)] are proposed, respectively.

Two strains, 8-4-E12T and 8-4-E13T, were isolated from a biowaste composting reactor. Based on 16S rRNA gene sequences, both strains belong to the genus Bacillus . Strain 8-4-E12T was most closely related to the type strains of Bacillus shackletonii , B. acidicola , B. sporothermodurans and B. oleronius (96.4, 96.3, 96.0 and 95.6 % 16S rRNA gene similarity, respectively), whereas strain 8-4-E13T was most closely related to the type strain of Bacillus humi (96.5 % sequence similarity). Strains 8-4-E12T and 8-4-E13T shared 94 % 16S rRNA gene sequence similarity. The fatty acid profile of strain 8-4-E12T was dominated by saturated iso- and anteiso-branched fatty acids (iso-C15 : 0, anteiso-C15 : 0, anteiso-C17 : 0 and iso-C16 : 0), and also contained considerable amounts of C16 : 0. The fatty acid profile of strain 8-4-E13T showed a predominance of iso-C15 : 0 (65 %), with smaller amounts of other saturated branched-chain fatty acids along with an unsaturated alcohol. Both strains contained diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine as major polar lipids. Additionally, strain 8-4-E12T contained an unknown lipid and strain 8-4-E13T two unknown (amino-)phospholipids. The diagnostic diamino acid found in the cell-wall peptidoglycan of 8-4-E12T and 8-4-E13T was meso-diaminopimelic acid. The predominant menaquinone was MK-7. The results of physiological and biochemical tests also allowed phenotypic differentiation of the two strains from each other and from related Bacillus species. On the basis of their phylogenetic, phenotypic and chemotaxonomic properties, strains 8-4-E12T and 8-4-E13T represent novel species of the genus Bacillus , for which the names Bacillus pervagus sp. nov. (type strain 8-4-E12T = DSM 23947T = LMG 27601T) and Bacillus andreesenii sp. nov. (type strain 8-4-E13T = DSM 23948T = LMG 27602T) are proposed.
