Dyadobacter arcticus sp. nov., isolated from Arctic soil

A psychrotolerant, Gram-negative, rod-shaped, yellow-pigmented bacterium, designated strain R-S7-29T, was isolated from a soil sample collected from the Svalbard Archipelago in Norway (78° N). Cells were non-motile, aerobic, and catalase- and oxidase-positive. Growth occurred at 4–28 °C (optimum, 18 °C). A phylogenetic tree based on 16S rRNA gene sequences showed that strain R-S7-29T belongs to the genus Dyadobacter (family ‘ Flexibacteraceae ’) with sequence similarity to related members of the genus ranging from 95.2 to 96.7 %. The major fatty acids were C16 : 1ω5c, iso-C17 : 0 3-OH and summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) and the predominant respiratory quinone was MK-7. The DNA G+C content of strain R-S7-29T was 50 mol%. On the basis of its phenotypic, phylogenetic and chemotaxonomic properties, strain R-S7-29T represents a novel species of the genus Dyadobacter , for which the name Dyadobacter arcticus sp. nov. is proposed. The type strain is R-S7-29T ( = CCTCC AB 2011022T = NRRL B-59659T).

A Gram-negative bacterium, designated DCY13T, was isolated from soil of a ginseng field in South Korea. Comparative analysis of 16S rRNA gene sequences showed that strain DCY13T shared the highest sequence similarity (95.0 %) with Sediminibacterium salmoneum NBRC 103935T and 87.6–91.4 % sequence similarity with other members of the family Chitinophagaceae . Cells were non-spore-forming rods, catalase- and oxidase-positive, motile by gliding and facultatively anaerobic. The only respiratory quinone was menaquinone 7 (MK-7) and the major fatty acids were iso-C17 : 0 3-OH, iso-C15 : 0 and iso-C15 : 1 G. The G+C content of the genomic DNA was 47.5±1.0 mol%. In addition to phosphatidylethanolamine, the major polar lipids were two unidentified aminophospholipids, one unidentified aminolipid and three unidentified polar lipids. The major cell-wall sugars were ribose, xylose and galactose. It is proposed that strain DCY13T represents a novel species in the genus Sediminibacterium , for which the name Sediminibacterium ginsengisoli sp. nov. is proposed. The type strain is DCY13T ( = KCTC 12833T  = JCM 15794T  = DSM 22335T). Emended descriptions of the genus Sediminibacterium and of Sediminibacterium salmoneum are also proposed.

Strain M2T, isolated from marine sediment collected at Jeju Island, was an aerobic, Gram-stain-negative, oxidase- and catalase-positive, motile, rod-shaped bacterium that formed circular, raised, yellow colonies. Strain M2T grew at 15–42 °C, pH 5.5–9.0 and with 1–9 % (w/v) NaCl. Phylogenetic analysis based on its 16S rRNA gene sequences indicated that strain M2T was closely related to Kineobactrum sediminis F02T (98.0 % sequence similarity). Ubiquinone-8 was determined to be the sole respiratory quinone. Summed feature 3 (C16 : 1  ω6c/C16 : 1  ω7c) and summed feature 8 (C18 : 1  ω7c/C18 : 1  ω6c) were identified as the predominant fatty acids. The DNA G+C content and digital DNA–DNA relatedness between strain M2T and K. sediminis F02T were 60.7 mol% and 19.5 %, respectively. Phosphatidylglycerol and phosphatidylethanolamine were identified as the major polar lipids. Thus, polyphasic characterization revealed that strain M2T represents a novel species in the genus Kineobactrum , for which the name Kineobactrum salinum sp. nov. is proposed. The type strain is M2T (=KCTC 72815T=VTCC 910108T).

A Gram-stain-negative, aerobic, non-motile and rod-shaped bacterium, designated KIS59-12T, was isolated from a soil sample collected on Hodo island, Boryeong, Republic of Korea. The strain grew at 10–33 °C, pH 6.0–7.5 and with 0–4 % NaCl (w/v). Results of phylogenetic analysis based on 16S rRNA gene sequences showed that strain KIS59-12T was in the same clade as Arachidicoccus rhizosphaerae Vu-144T and Arachidicoccus ginsenosidivorans Gsoil809T with 97.5 and 97.2 % sequence similarity, respectively. Comparative genome analysis between strain KIS59-12T and A. rhizosphaerae Vu-144T showed that average nucleotide identity value was 69.4 % and the digital DNA–DNA hybridization value was 19.1 %. The major respiratory quinone was menaquinone-7. The major polar lipids were phosphatidylethanolamine and an unknown polar lipid. The predominant cellular fatty acids were iso-C15 : 0, iso-C15 : 1 G and iso-C17 : 0 3-OH, which supported the affiliation of strain KIS59-12T with the genus Arachidicoccus . The major polyamines were homospermidine and putrescine. The genomic DNA G+C content was 36.4 mol%. On the basis of phylogenetic, physiological and chemotaxonomic characteristics, strain KIS59-12T represents a novel species of the genus Arachidicoccus , for which the name Arachidicoccus soli sp. nov. is proposed. The type strain of Arachidicoccus soli is KIS59-12T (=KACC 17340T=NBRC 113161T).

A Gram-stain-negative, short rod-shaped, non-motile, non-spore-forming bacterial strain, designated H7T, was isolated from the Gobi desert, Xinjiang Province of China. The temperature, NaCl and pH ranges for growth were 8–40 °C (optimum 30 °C), 0–5 % (w/v) NaCl and pH 6–10 (optimum pH 7), respectively. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain H7T belonged to the genus Sphingobacterium and showed highest sequence similarity (91 %) to Sphingobacterium composti DSM 18850. The genomic DNA G+C content was 44.3 mol%. MK-7 was identified as the predominant respiratory quinone. Strain H7T contained C16 : 1ω7c and/or C16 : 1ω6c, iso-C15 : 0 and iso-C17 : 0 3-OH as major fatty acids. These data support the affiliation of strain H7T to the genus Sphingobacterium while phenotypic data indicated it is a representative strain of a novel species for which the name Sphingobacterium gobiense sp. nov. is proposed. The type strain is H7T ( = ACCC 05757T = KCTC 32293T).

A Gram-staining-negative, non-spore-forming, non-gliding and rod-shaped bacterial strain KYW566T, was isolated from seawater of the Suncheon Bay, Korea, and its taxonomic position was investigated by using a polyphasic study. The cells contained MK-6 as the only respiratory quinone and contained iso-C15 : 0 (13.8 %), iso-C16 : 0 3-OH (13.1 %), anteiso-C15 : 0 (9.3 %), iso-C15 : 0 3-OH (8.7 %), iso-C15 : 1 G (6.3 %) and summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) (6.0 %) as the major fatty acids. The DNA G+C content of strain KYW566T was 41.6±0.8 mol% (mean±sd of three determinations). A phylogenetic tree based on 16S rRNA gene sequences showed that strain KYW566T forms an evolutionary lineage within the radiation enclosing the members of the genus Lutibacter with Lutibacter flavus IMCC1507T as its nearest neighbour (96.7 % sequence similarity). A number of phenotypic characteristics distinguished strain KYW566T from described members of the genus Lutibacter . On the basis of the evidences presented in this study, strain KYW566T represents a novel species, for which the name Lutibacter agarilyticus sp. nov. is proposed. The type strain is KYW566T ( = KCTC 23842T = JCM 18281T).

A novel orange-pigmented strain, designated R2-35T, was isolated from a glacier till near Ny-Alesund, Svalbard Archipelago, Norway. The cells were aerobic, Gram-negative, rod-shaped and sometimes filamentous. Growth occurred at 4–28 °C (optimum, 20 °C), at pH 7.0–9.0 (optimum, pH 8.0) and with 0–1 % NaCl. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain R2-35T belonged to the genus Spirosoma with sequence similarity to related species ranging from 91.65 to 95.19 %. Strain R2-35T contained C16 : 0 (10.7 %), C18 : 0 (9.2 %), C16 : 1ω5c (16.5 %) and summed feature 3 (C16 : 1ω6c and/or C16 : 1ω7c) (24.6 %) as the major cellular fatty acids, MK-7 as the major respiratory quinone, and phosphatidylethanolamine as the main polar lipid. The DNA G+C content of strain R2-35T was 54.9 mol%. On the basis of phylogenetic, physiological and chemotaxonomic data, strain R2-35T is considered to represent a novel species of the genus Spirosoma , for which the name Spirosoma arcticum sp. nov., is proposed, The type strain is R2-35T ( = CCTCC AB 2012849T = LMG 28141T).

An orange, rod-shaped, Gram-reaction-negative, aerobic and gliding bacterial strain devoid of flagella, designated strain KYW614T, was isolated from seawater collected from Gwangyang Bay, Republic of Korea. Zeaxanthin was the major carotenoid pigment produced and flexirubin-type pigments were not produced. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain KYW614T belonged to the family Flavobacteriaceae and it was most closely related to Mesoflavibacter zeaxanthinifaciens TD-ZX30T (96.5 %, sequence similarity). The predominant cellular fatty acids of strain KYW614T were iso-C15 : 1 G (10.5 %), summed feature 3 (C16 : 1ω7c/C16 : 1ω6c; 10.0 %), iso-C15 : 0 (9.5 %), C15 : 0 (7.5 %) and iso-C17 : 0 3-OH (7.4 %). MK-6 was the only isoprenoid quinone and the DNA G+C content was 32.6 mol%. Data from a polyphasic taxonomic study suggested that the isolate represents a novel species in the genus Mesoflavibacter , for which the name Mesoflavibacter aestuarii sp. nov. is proposed. The type strain is KYW614T ( = KCTC 32269T = JCM 19524T).

A Gram-stain-negative, non-motile and aerobic bacterium, designated CF17T, was isolated from coastal planktonic seaweeds, East China Sea. The isolate grew at 18–37 °C (optimum 25–28 °C), pH 6.5–9.0 (optimum 7.0–8.0) and with 0–5 % NaCl (optimum 1–2 %, w/v) and 0.5–10 % sea salts (optimum 2–3 %, w/v). Growth of strain CF17T could be stimulated prominently by supplementing the growth medium with the autoclaved supernatant of a culture of strain CF5, which was isolated from the same sample along with strain CF17T. The cell morphology of strain CF17T was a bean-shaped rod consisting of a swollen end and a long prostheca. The phylogenetic analysis of 16S rRNA gene sequences indicated that strain CF17T clustered with Gemmobacter nectariphilus DSM 15620T within the genus Gemmobacter . The DNA G+C content of strain CF17T was 61.4 mol%. The respiratory quinone was ubiquinone Q-10. The major fatty acids included C18 : 1ω7c and C18 : 0. The polar lipids of strain CF17T consisted of phosphatidylethanolamine, phosphatidylglycerol, phosphatidylcholine, two uncharacterized phospholipids, one uncharacterized aminolipid, three uncharacterized glycolipids and one uncharacterized lipid. On the basis of phenotypic, phylogenetic and chemotaxonomic data, strain CF17T ( = CGMCC 1.11024T = JCM 18498T) is considered to represent a novel species of the genus Gemmobacter , for which the name Gemmobacter megaterium sp. nov. is proposed.

A novel Gram-negative strain, designated M-SX103T, was isolated from a fresh leaf of Nicotiana tabacum Linn. collected at Yuxi City, Yunnan province, China, and subjected to a polyphasic taxonomic study. Growth occurred at 10–40 °C, at pH 6.0–9.0 and with 0–3 % (w/v) NaCl. The predominant isoprenoid quinone was MK-7. The major cellular fatty acids of strain M-SX103T were iso-C15 : 0, iso-C17 : 0 3-OH and summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c). The G+C content of the genomic DNA was 40.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that the isolate belongs to the genus Sphingobacterium , sharing 95.4–89.7 % sequence similarity with type strains of members of the genus Sphingobacterium ; it was most closely related to Sphingobacterium composti T5-12T. On the basis of the taxonomic evidence, a novel species, Sphingobacterium nematocida sp. nov., is proposed. The type strain is M-SX103T ( = JCM 17339T = CCTCC AB 2010390T).

A non-motile and rod-shaped bacterium, designated strain 02SUJ3T, was isolated from freshwater collected from the Juam Reservoir (Republic of Korea). Cells were Gram-stain-negative, aerobic, oxidase-negative and catalase-positive. The major fatty acids were iso-C15 : 0, iso-C15 : 1 G and iso-C17 : 0 3-OH. The strain contained MK-7 as the major isoprenoid quinone. The main polar lipid was phosphatidylethanolamine. The DNA G+C content was 46.4 mol%. A phylogenetic tree based on 16S rRNA gene sequences showed that strain 02SUJ3T forms an independent lineage within the genus Flavisolibacter with low sequence similarity to Flavisolibacter ginsengiterrae Gsoil 492T and Flavisolibacter ginsengisoli Gsoil 643T (95.7 %). Phenotypic characteristics distinguished strain 02SUJ3T from members of the genus Flavisolibacter . On the basis of the evidence presented in this study, strain 02SUJ3T is considered to represent a novel species, for which the name Flavisolibacter rigui sp. nov. is proposed. The type strain is 02SUJ3T ( = JCM 17515T = KCTC 23328T). An emended description of the genus Flavisolibacter is also provided.
