Domibacillus robiginosus gen. nov., sp. nov., isolated from a pharmaceutical clean room

A novel red-pigmented bacterial strain, designated WS 4628T, was isolated from a pharmaceutical clean room of a vaccine-producing company and was investigated in a taxonomic study using a polyphasic approach. The strain was Gram-stain-positive, strictly aerobic, motile, catalase-positive and produced spherical to slightly ellipsoidal endospores in rods. The genomic DNA G+C content was 44.1 mol%. The major fatty acids were anteiso-C15 : 0, iso-C15 : 0 and anteiso-C17 : 0 and the predominant quinone was MK-6. The polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, an unidentified phosphoglycolipid and an unidentified phospholipid. meso-diaminopimelic acid (type A1γ) was present in the cell-wall peptidoglycan and the major whole-cell sugars were glucose and ribose. The closest phylogenetic neighbours were identified as Bacillus badius ATCC 14574T (95.8 % 16S rRNA gene sequence similarity), Bacillus indicus Sd/3T (94.8 %), Jeotgalibacillus alimentarius YKJ-13T (94.8 %) and Bacillus cibi JG-30T (94.8 %). Phylogenetic, physiological, biochemical and morphological differences between strain WS 4628T and its closest relatives in the families Bacillaceae and Planococcaceae suggest that this strain represents a novel species in a new genus in the family Bacillaceae for which the name Domibacillus robiginosus gen. nov., sp. nov. is proposed; the type strain of the type species is WS 4628T ( = DSM 25058T = LMG 26645T).

A novel Gram-staining-positive, rod-shaped, motile, strictly aerobic, endospore-forming bacterium, designated WCC 4585T, was isolated from a pharmaceutical production line. The organism grew optimally at 30 °C, at pH 8 and in the presence of 0.5 % (w/v) NaCl. Oval endospores were formed subterminally and terminally in swollen sporangia. The cell-wall diamino acid was meso-diaminopimelic acid (type A1γ) and the genomic DNA G+C content was 38.7 mol%. The major menaquinone was MK-7. The cellular fatty acid profile contained major amounts of iso-C15 : 0, anteiso-C15 : 0 and anteiso-C17 : 0, and the cellular phospholipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and aminophospholipid. The isolate was most closely related to Bacillus oceanisediminis H2T, Bacillus infantis SMC 4352-1T, Bacillus firmus NCIMB 9366T, Bacillus circulans ATCC 4513T and Bacillus horneckiae DSM 23495T with which it shared less than 98.0 % 16S rRNA gene sequence similarity. DNA–DNA relatedness values between strain WCC 4585T and five type strains of related species were ≤27 % and sequence similarity values based on groEL sequences were ≤88.7 %. On the basis of the characteristics presented, strain WCC 4585T is proposed to represent a novel species, Bacillus gottheilii sp. nov. The type strain is WCC 4585T( = DSM 23668T = CCUG 59876T = LMG 25856T).

A novel bacterium, designated strain F051-1T, isolated from a seawater sample collected from the coast at Damupo beach in Pohang, Korea, was investigated in a polyphasic taxonomic study. Cells were yellow-pigmented, strictly aerobic, Gram-staining-negative and rod-shaped. The temperature, pH and NaCl ranges for growth were 4–30 °C, pH 6.0–9.0 and 1.0–6.0 % (w/v), respectively. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain F051-1T belongs to the genus Psychroserpens in the family Flavobacteriaceae . Its closest relatives were Psychroserpens burtonensis ACAM 188T (96.8 % 16S rRNA gene sequence similarity) and Psychroserpens mesophilus KOPRI 13649T (95.7 %). The major cellular fatty acids were iso-C15 : 0, iso-C15 : 1 G and anteiso-C15 : 0. The polar lipid profile consisted of a mixture of phosphatidylethanolamine, two unidentified aminolipids, one unidentified phospholipid and eight unidentified lipids. The major respiratory quinone was menaquinone-6 and the genomic DNA G+C content of the strain was 33.5 mol%. On the basis of phenotypic, phylogenetic and genotypic data, strain F051-1T represents a novel species within the genus Psychroserpens , for which the name Psychroserpens damuponensis sp. nov. is proposed. The type strain is F051-1T ( = KCTC 23539T  = JCM 17632T).

A Gram-stain-negative, non-spore-forming, strictly aerobic, orange-pigmented bacterial strain, motile by gliding, designated CAU 1044T, was isolated from a green seaweed and its taxonomic position was investigated using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain CAU 1044T formed a distinct lineage within the genus Maribacter and was most closely related to Maribacter antarcticus JCM 15445T and Maribacter arcticus KOPRI 20941T (96.3 and 95.7 % similarity, respectively). Strain CAU 1044T contained menaquinone 6 as the only isoprenoid quinone and iso-C15 : 0, summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c and/or iso-C15 : 0 2-OH), iso-C17 : 0 3-OH and iso-C15 : 1 G as the major fatty acids. The cell wall peptidoglycan of strain CAU 1044T contained meso-diaminopimelic acid and the major whole-cell sugars were glucose and ribose. The polar lipids were composed of phosphatidylethanolamine, one unidentified phospholipid, six unidentified aminolipids and four unidentified lipids. The DNA G+C content was 40.2 mol%. On the basis of phenotypic data and phylogenetic inference, strain CAU 1044T should be classified as a representative of a novel species in the genus Maribacter for which the name Maribacter chungangensis sp. nov. is proposed. The type strain is CAU 1044T ( = KCTC 23735T  = CCUG 61948T). Emended descriptions of the genus Maribacter and the species M. arcticus KCTC 22053T are also proposed.

A novel rod-shaped, Gram-stain-negative, strictly aerobic and alginate-degrading marine bacterium, designated CCB-QB4T, was isolated from a surface of algal turf collected from a coastal area of Penang, Malaysia. The cells showed motility by a lateral flagellum. The rod-shaped cells formed long chains end-to-end. Phylogenetic analysis based on the 16S rRNA gene sequence of strain CCB-QB4T showed 94.07, 92.69, 91.52 and 90.90 % sequence similarity to Algibacillus agarilyticus RQJ05T, Catenovulum maritimum Q1T, Catenovulum agarivorans YM01T and Catenovulum sediminis D2T, respectively. Strain CCB-QB4T formed a cluster with A. agarilyticus RQJ05T. Strain CCB-QB4T was catalase-negative, oxidase-positive, and degraded agar, alginate, and starch. Cell growth was observed at 15–40 °C, at pH 7.0–10.0 and in the presence of 1–6 % (w/v) NaCl and glucose. The major fatty acids were summed feature 3 (C16 : 1 ω7c/iso-C15 : 0 2-OH), C16 : 0 and C18 : 1 ω7c. The polar lipids were phosphatidylethanolamine, two unidentified aminolipids, two unidentified glycolipids, an unidentified phospholipid and unidentified lipid. The major respiratory quinone was ubiquinone-8. The genomic DNA G+C content was 46.7 mol%. Based on the phenotypic, chemotaxonomic and phylogenetic data, strain CCB-BQ4T represents a novel species in a new genus, for which the name Saccharobesus litoralis gen. nov., sp. nov. is proposed. The type strain is CCB-QB4T (=JCM 33513T=CCB-MBL 5008T).

Strain HMF4947T, isolated from the bark of a ginkgo tree, was a pale-pink coloured, Gram-stain-negative, non-motile, strictly aerobic and rod-shaped bacterium. The isolate grew optimally on Reasoner's 2A agar at 30 °C, pH 7.0 and with 0 % NaCl. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain HMF4947T belonged to the genus Hymenobacter and was most closely related to Hymenobacter metalli A2-91T (96.9 % sequence similarity) and Hymenobacter pomorum 9-2-1-1T (96.5 %). The average nucleotide identity and estimated DNA–DNA hybridization values between strain HMF4947T and Hymenobacter arizonensis DSM 17870T were 74.3 and 20.5 %, respectively. The major fatty acids were summed feature 3 (C16 : 1  ω7c and/or C16 : 1  ω6c), iso-C15 : 0 and C16 : 1  ω5c. The predominant isoprenoid quinone was menaquinone-7. The polar lipids comprised phosphatidylethanolamine, one unidentified aminoglycolipid, three unidentified aminophospholipids, one unidentified phospholipid, three unidentified aminolipids, two unidentified glycolipids and three unidentified polar lipids. The genomic DNA G+C content was 59.3 mol%. Thus, based on phylogenetic, phenotypic and chemotaxonomic data, strain HMF4947T represents a novel species of the genus Hymenobacter , for which the name Hymenobacter ginkgonis sp. nov. is proposed. The type strain of the species is strain HMF4947T (=KCTC 72780T=NBRC 114271T).

A novel actinobacterium, designated strain DW4-2T, was isolated from duckweed (Spirodela sp.) collected from an agricultural pond in Kasetsart University, Bangkok, Thailand. The morphological, chemotaxonomic and phylogenetic characteristics were consistent with its classification in the genus Streptomyces . Strain DW4-2T showed the highest 16S rRNA gene sequence similarity values to Streptomyces qinglanensis DSM 42035T (98.5 %), S treptomyces smyrnaeus DSM 42105T (98.4 %) and Streptomyces oryzae S16-07T (98.4 %). Digital DNA–DNA hydridization and average nucleotide identity values between the genome sequences of strain DW4-2T with S. qinglanensis DSM 42035T (29.8 and 87.8 %), S. smyrnaeus DSM 42105T (33.1 and 89.0 %) and S. oryzae S16-07T (33.0 and 88.9 %) were below the thresholds of 70 and 95–96 % for prokaryotic conspecific assignation. Chemotaxonomic data revealed that strain DW4-2T possessed MK-9(H6) and MK-9(H8) as the predominant menaquinones. It contained ll -diaminopimelic acid as the diagnostic diamino acid and glucose, ribose and trace amount of madurose in whole-cell sugars. The polar lipids consisted of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, phosphatidylinositol mannoside, an unidentified aminolipid, an unidentified lipid and an unidentified phospholipid. The predominant cellular fatty acids were anteiso-C17 : 0, anteiso-C15 : 0 and iso-C16 : 0. The genomic DNA size of the strain DW4-2T was 7 310 765 bp with DNA G+C content 71.0 mol%. Genomic analysis of the genome indicated that the strain DW4-2T had the potential to produce bioactive compounds. On the basis of these genotypic and phenotypic data, it is supported that strain DW4-2T represents a novel species of the genus Streptomyces , for which the name Streptomyces spirodelae sp. nov. is proposed. The type strain is strain DW4-2T (=TBRC 13095T=NBRC 114803T).

A novel red-pigmented bacterial strain, designated NIO-1016T, was isolated from a sediment sample from Chorao Island, India and was investigated by a polyphasic taxonomic approach. The strain was Gram-reaction-positive, strictly aerobic, motile and catalase-positive and produced spherical to slightly ellipsoidal endospores with subterminal position on swollen sporangia. The genomic DNA G+C content was 46.9 mol%. The major fatty acids were anteiso-C15 : 0, anteiso-C17 : 0, iso-C15 : 0 and C16 : 0. The predominant quinones were MK-6 (89.3 %) and MK-7 (8.7 %). The polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, and an unidentified phospholipid. meso-Diaminopimelic acid (type A1γ) was present in the cell-wall peptidoglycan and the major whole-cell sugars were glucose and ribose. The closest phylogenetic neighbours were identified as Domibacillus robiginosus DSM 25058T (98.0 % similarity) and Domibacillus indicus DSM 28032T (97.2 % similarity), other species of the genus Bacillus displayed below 96 % similarity. Phylogenetic, physiological, biochemical and morphological differences between strain NIO-1016T and its closest neighbours indicate that this strain represents a novel species in the genus Domibacillus in the family Bacillaceae for which the name Domibacillus enclensis sp. nov. is proposed with the type species NIO-1016T ( = DSM 25145T = NCIM 5462T = CCTCC AB 2011121T).

A novel bacterial strain designated HME7863T was isolated from a freshwater reservoir in Goheung, Republic of Korea. Cells of strain HME7863T were Gram-staining-negative, strictly aerobic, orange-pigmented, rod-shaped and motile by gliding. Phylogenetic analysis based on 16S rRNA gene sequence showed that strain HME7863T formed a lineage within the genus Sediminibacterium . Strain HME7863T was closely related to Sediminibacterium ginsengisoli DCY13T (96.9 % sequence similarity) and Sediminibacterium salmoneum NJ-44T (96.4 %). The major cellular fatty acids were iso-C15 : 1 G, iso-C15 : 0, iso-C17 : 0 3-OH and iso-C16 : 0 3-OH. Polar lipid analysis revealed the presence of phosphatidylethanolamine, five unidentified aminolipids, two unidentified aminophospholipids, one unidentified phospholipid and two unidentified polar lipids. The only respiratory quinone was MK-7. The DNA G+C content was 40.5 mol%. On the basis of the evidence presented in this study, strain HME7863T represents a novel species of the genus Sediminibacterium , for which the name Sediminibacterium goheungense sp. nov., is proposed. The type strain is HME7863T ( = KCTC 23945T = CECT 8100T).

A Gram-stain-positive, strictly aerobic, short-rod-shaped, non-motile strain (designated MJ32T) was isolated from a sludge sample of the Daejeon sewage disposal plant in South Korea. A polyphasic approach was applied to study the taxonomic position of strain MJ32T. Strain MJ32T showed highest 16S rRNA gene sequence similarity to Gordonia hirsuta DSM 44140T (98.1 %) and Gordonia hydrophobica DSM 44015T (97.0 %); levels of sequence similarity to the type strains of other recognized Gordonia species were less than 97.0 %. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain MJ32T belonged to the clade formed by members of the genus Gordonia in the family Gordoniaceae . The G+C content of the genomic DNA of strain MJ32T was 69.2 mol%. Chemotaxonomically, strain MJ32T showed features typical of the genus Gordonia . The predominant respiratory quinone was MK-9(H2), the mycolic acids present had C56–C60 carbon atoms, and the major fatty acids were C16 : 0 (34.6 %), tuberculostearic acid (21.8 %), C16 : 1ω7c (19.5 %) and C18 : 1ω9c (12.7 %). The peptidoglycan type was based on meso-2,6-diaminopimelic acid as the diagnostic diamino acid with glycolated sugars. On the basis of phylogenetic inference, fatty acid profile and other phenotypic properties, strain MJ32T is considered to represent a novel species of the genus Gordonia , for which the name Gordonia caeni sp. nov. is proposed. The type strain is MJ32T ( = KCTC 19771T = JCM 16923T).

A Gram-staining-negative, strictly aerobic, rod-shaped, non-flagellated, non-spore-forming and gliding marine bacterium designated strain CC-AMZ-301T was isolated from coastal surface seawater near Taichung harbour, Taiwan. Strain CC-AMZ-301T predominantly synthesized zeaxanthin and thus formed yellow colonies on marine agar. The novel strain showed high pairwise 16S rRNA gene sequence similarity to Kordia periserrulae IMCC1412T (95.0 %), K. algicida KCTC 8814PT (94.4 %) and K. antarctica IMCC3317T (94.2 %), and formed a phylogenetic lineage tightly associated with species of the genus Kordia . The polar lipid profile of strain CC-AMZ-301T consisted of three unidentified lipids, three unidentified aminolipids and one unidentified phospholipid. The major fatty acids (>5 % of total) were iso-C17 : 0 3-OH, iso-C15 : 0, summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) and summed feature 9 (iso-C17 : 1ω9c and/or 10-methyl C16 : 0). The DNA G+C content was 36.4 mol%. Menaquinone 6 (MK-6) was the predominant respiratory quinone. The major polyamine was the triamine sym-homospermidine. Based on evidence from the present polyphasic study, strain CC-AMZ-301T is proposed to represent a novel species of the genus Kordia , for which the name Kordia aquimaris sp. nov. is proposed; the type strain is CC-AMZ-301T ( = JCM 18556T = BCRC 80464T). An emended description of the genus Kordia is also proposed.
