ABSTRACT Nkx2.5 and Nkx2.6 are murine homologs of Drosophilatinman. Their genes are expressed in the ventral region of the pharynx at early stages of embryogenesis. However, no abnormalities in the pharynges of embryos with mutations in either Nkx2.5 or Nkx2.6 have been reported. To examine the function of Nkx2.5 and Nkx2.6 in the formation of the pharynx, we generated and analyzed Nkx2.5 and Nkx2.6 double-mutant mice. Interestingly, in the double-mutant embryos, the pharynx did not form properly. Pharyngeal endodermal cells were largely missing, and the mutant pharynx was markedly dilated. Moreover, we observed enhanced apoptosis and reduced proliferation in pharyngeal endodermal cells of the double-mutant embryos. These results demonstrated a critical role of the NK-2 homeobox genes in the differentiation, proliferation, and survival of pharyngeal endodermal cells. Furthermore, the development of the atrium was less advanced in the double-mutant embryos, indicating that these two genes are essential for both pharyngeal and cardiac development.

Successful seed development requires coordinated interaction of the endosperm and embryo. In most dicotyledonous seeds, the endosperm is crushed and absorbed by the expanding embryo in the later stages of seed development. Little is known about the metabolic interaction between the two filial tissues early in seed development. We examined the potential role of sucrose synthase (Sus) in the endosperm development of cotton. Sus was immunologically localised in the cellularising endosperm, but not in the heart-stage embryo at 10 days after anthesis. The activities of Sus and acid invertase were significantly higher in the endosperm than in the young embryos, which corresponded to a steep concentration difference in hexoses between the endosperm and the embryo. This observation indicates a role for the endosperm in generating hexoses for the development of the two filial tissues. Interestingly, Sus expression and starch deposition were spatially separated in the seeds. Silencing the expression of Sus in the endosperm using an RNAi approach led to the arrest of early seed development. Histochemical analyses revealed a significant reduction in cellulose and callose in the deformed endosperm cells of the Sus-suppressed seed. The data indicate a critical role of Sus in early seed development through regulation of endosperm formation.

AbstractRecent prevalence estimates for autism have been alarming as a function of the notable increase. Speech-language pathologists play a critical role in screening, assessment and intervention for children with autism. This article reviews signs that may be indicative of autism at different stages of language development, and discusses the importance of several psychometric properties—sensitivity and specificity—in utilizing screening measures for children with autism. Critical components of assessment for children with autism are reviewed. This article concludes with examples of intervention targets for children with ASD at various levels of language development.

Cardiac registries have filled many gaps in knowledge related to arrhythmogenic cardiovascular conditions. Despite the less robust level of evidence available in registries when compared with clinical trials, registries have contributed a range of clinically useful information. In this review, the authors discuss the role that registries have played – related to diagnosis, natural history, risk stratification, treatment, and genetics of arrhythmogenic cardiovascular conditions – in closing knowledge gaps, and their role in the future.
