By an agar dilution method, the antimicrobial susceptibilities of antral sinus puncture isolates were studied. Pneumococci were generally susceptible to amoxicillin, azithromycin, and clarithromycin, but 17% of pneumococcal isolates were resistant to cefuroxime.Haemophilus influenzae isolates were resistant to amoxicillin and clarithromycin. β-Lactamase production occurred in 69% of Prevotella species. One-third ofPeptostreptococcus magnus isolates were resistant to azithromycin and clarithromycin. Cefuroxime had limited activity against Prevotella species and P. magnus. Levofloxacin was active against most isolates except peptostreptococci. Amoxicillin-clavulanate was active against all isolates, with the MIC at which 90% of the isolates were inhibited being ≤1 μg/ml.

ABSTRACT The antimicrobial activities of garenoxacin and eight other antibiotics against 641 anaerobic isolates were evaluated with the NCCLS agar dilution method. Overall, the MICs of garenoxacin for 50 and 90% of the strains tested (in micrograms per milliliter) were as follows: Bacteroides fragilis group, 0.5 and 2; Prevotella spp., 0.25 and 2; Fusobacterium spp., 0.25 and 0.5; Porphyromonas spp., 0.125 and 0.25; Bilophila wadsworthia, 0.5 and 1; Veillonella spp., 0.25 and 0.5; Clostridium spp., 0.25 and 1; Clostridium difficile, 2 and >64; Bifidobacterium spp., 1 and 2; Eggerthella lenta, 0.25 and 1; Propionibacterium spp., 0.5 and 0.5; gram-positive cocci, 0.125 and 0.25.

ABSTRACT The in vitro susceptibilities of 678 Campylobacter jejuni and Campylobacter coli strains isolated from stool samples of the same number of Finnish subjects were studied. A total of 523 patients, representing inhabitants from throughout Finland, had not traveled abroad within the 2 weeks prior to becoming ill, whereas 155 persons had presumably acquired their infections abroad. The antimicrobial agents studied were erythromycin, ciprofloxacin, levofloxacin, trovafloxacin, and moxifloxacin. The MICs of these antimicrobial agents were determined by the agar dilution method. The growth of all domestic isolates was inhibited by erythromycin at concentrations of 4 μg/ml, and for these isolates the fluoroquinolone MICs at which 90% of isolates are inhibited (MIC90s) ranged from 0.06 to 0.5 μg/ml. For the foreign isolates, the erythromycin MIC90 was still low (4 μg/ml), but their susceptibilities to fluoroquinolones were clearly reduced (MIC90s, 8 to 64 μg/ml). Of the four different fluoroquinolones studied, ciprofloxacin was the least active (MIC90, 64 μg/ml).

ABSTRACT By using an agar dilution method, the in vitro activities of ramoplanin, teicoplanin, vancomycin, linezolid, and five other agents were determined against 300 gram-positive and 54 gram-negative strains of intestinal anaerobes. Ramoplanin was active at ≤2 μg/ml against 287 of 300 (95.7%) gram-positive organisms, including 18 strains of Clostridium difficile for which MICs of ramoplanin were 0.25 to 0.5 μg/ml; for 3 of these, linezolid MICs were 8 to 16 μg/ml. Nineteen Clostridium innocuum strains for which the vancomycin MIC at which 90% of strains were inhibited was 16 μg/ml were susceptible to ramoplanin at 0.06 to 0.25 μg/ml and to teicoplanin at 0.125 to 1.0 μg/ml. All strains of Eubacterium, Actinomyces, Propionibacterium, and Peptostreptococcus spp. were inhibited by ≤0.25 μg of ramoplanin per ml and ≤1 μg of vancomycin per ml. Ramoplanin was also active at ≤4 μg/ml against 15 of 22 of the Prevotella and Porphyromonas strains tested, but ramoplanin MICs for all 31 strains of the Bacteroides fragilis group, the Fusobacterium mortiferum-Fusobacterium varium group, and Veillonella spp. were ≥256 μg/ml. Ramoplanin displays excellent activity against C. difficile and other gram-positive enteric anaerobes, including vancomycin-resistant strains; however, it has poor activity against most gram-negative anaerobes and thus potentially has a lesser effect on the ecological balance of normal fecal flora.

The susceptibility to 9 antimicrobial agents of 32 aerobic bacterial isolates and to 10 antimicrobial agents of 37 anaerobic bacterial isolates from 23 cases of bovine summer mastitis (16 Actinomyces pyogenes isolates, 8 Streptococcus dysgalactiae isolates, 3 S. uberis isolates, 3 S. acidominimus isolates, 2 Streptococcus spp., 15 Peptostreptococcus indolicus isolates, 10 Fusobacterium necrophorum isolates, and 12 isolates of anaerobic gram-negative rods) was determined by the agar dilution method. All isolates except one Bacteroides fragilis isolate (beta-lactamase producer) were susceptible to penicillin G, amoxicillin, amoxicillin-clavulanate, cefoxitin, clindamycin, and chloramphenicol (the B. fragilis strain was susceptible to the last four), which had MICs at which 90% of isolates were inhibited (MIC90s) of < or = 0.06, < or = 0.06, < or = 0.06 0.25, < or = 0.06, and 4.0 micrograms/ml, respectively. Spiramycin was active against the gram-positive aerobes (MIC90, 1.0 microgram/ml) but not against the anaerobes (MIC90, 16.0 micrograms/ml). Similar trends were noted for susceptibilities of aerobic and anaerobic bacteria to ofloxacin (MIC90s, 2.0 and 8 micrograms/ml, respectively). Occasional strains of aerobic streptococci were resistant to oxytetracycline, but all anaerobes were susceptible. Tinidazole was active against all anaerobes (MIC90, 2.0 micrograms/ml). beta-Lactamase was produced only by the B. fragilis isolate.

ABSTRACT Daptomycin has in vitro activity against gram-positive anaerobic bacteria, although limited numbers of species have been tested. We studied the in vitro activities of daptomycin, vancomycin, and penicillin against more than 100 strains each of Clostridium difficile, C. perfringens, Finegoldia magna, and Propionibacterium acnes. Daptomycin Etest MICs and results from time-kill studies were determined for selected strains. For 392 of 421 strains (93%), daptomycin was inhibitory at ≤1 μg/ml, including 15 of 16 strains of C. difficile with elevated linezolid MICs of 8 and 16 μg/ml, all 32 strains with moxifloxacin MICs of ≥4 μg/ml, and all 16 strains resistant to clindamycin. Daptomycin MICs were also ≤1 μg/ml for all 16 F. magna strains resistant to clindamycin and all 32 strains resistant to tetracycline. Only one strain, a C. perfringens strain, had a MIC of >2 μg/ml to daptomycin. Eighty-five and 92.5% of the Etest MICs were within 1 dilution of the agar dilution method for all drugs at 24 and 48 h, respectively. In time-kill studies, a C. difficile strain was inhibited by both daptomycin and vancomycin at 1, 2, 4, 8, and 24 h; colony counts were decreased by 2.3 to 2.9 log at 24 h. Vancomycin was not bactericidal for C. perfringens; however, daptomycin showed bactericidal activity as early as 1 h at four and eight times the MIC and at 2 and 4 h at two and four times the MIC.

The activities of DU-6859a, levofloxacin, ofloxacin, sparfloxacin, and ciprofloxacin against bite wound isolates were determined by the agar dilution method. DU-6859a was the most active compound (MICs, < or = 0.125 microg/ml) against all Pasteurella species, Staphylococcus aureus, and streptococci; anaerobes were susceptible to < or = 0.5 microg/ml, except fusobacteria, which were susceptible to < or = 2 microg/ml. Against aerobes, levofloxacin was more active than ofloxacin (MIC at which 90% of isolates are inhibited [MIC90], < or = 1.0 microg/ml for both) and sparfloxacin and ciprofloxacin were also active (MIC90s, < or = 0.25 and < 1 microg/ml, respectively).

During this study, thein vitroantifungal and antibacterial activities of different extracts (aqueous and organic) obtained from a French propolis batch were evaluated. Antifungal activity was evaluated by broth microdilution on three pathogenic strains:Candida albicans, C. glabrata, andAspergillus fumigatus. Antibacterial activity was assayed using agar dilution method on 36 Gram-negative and Gram-positive strains includingStaphylococcus aureus. Organic extracts showed a significant antifungal activity againstC. albicansandC. glabrata(MIC80between 16 and 31 µg/mL) but only a weak activity towardsA. fumigatus(MIC80= 250 µg/mL). DCM based extracts exhibited a selective Gram-positive antibacterial activity, especially againstS. aureus(SA) and several of its methicillin-resistant (MRSA) and methicillin-susceptible (MSSA) strains (MIC10030–97 µg/mL). A new and active derivative of catechin was also identified whereas a synergistic antimicrobial effect was noticed during this study.

The increase in dermatophytoses and the fact that some patients do not respond well to therapy make it necessary to find new antifungal agents. As part of our ongoing studies on medicinal plants from Iran, we studied antidermatophytic activities of Ferula latisecta essential oil, which had shown considerable antifungal activity in preliminary antimicrobial screening. Antifungal activity was evaluated by determination of MIC values using the agar dilution method on type strains of Candida albicans and dermatophytes. The composition of the oil was characterized by GC and GC/MS analyses. The essential oil was rich in polysulfides (75.2%) and exhibited good activity against Trichophyton rubrum and T. verrucosom for about three weeks, with a MIC value 96 μg/mL. The oil showed antifungal activity, especially against dermatophytes, and the activity is probably related to the sulfur-containing components of the oil. This study has identified that the polysulfides-rich essential oil of Ferula latisecta fruits has activity against a range of human pathogenic dermatophytes, justifying future clinical trials to validate its use as a therapeutic alternative for dermatophytosis.

ABSTRACT The in vitro susceptibilities of 170 clinical isolates plus 12 American Type Culture Collection strains of Pasteurella species comprising nine species and three Pasteurella multocida subspecies were studied by an agar dilution method. Garenoxacin (BMS-284756), a new des-fluoro(6) quinolone, was active at ≤0.06 μg/ml against all isolates, including four β-lactamase-producing strains, with >90% of the strains susceptible to ≤0.008 μg/ml. Garenoxacin was generally 1 to 2 dilutions more active than levofloxacin and moxifloxacin and was the most active agent tested. Cefoxitin required 1 μg/ml for inhibition of 51 of 182 (29%) of strains, and 3 strains (also β-lactamase producers) were resistant to doxycycline.
