Free amino acids of sunflower hypocotyl tissue during crown gall tumor development

The concentration of each free amino acid present in hypocotyl tissue of sunflower seedlings was determined during the period following both sterile wounding and inoculation with either tumorigenic (strain 1025) or non-tumorigenic (strain IIBNV6) Agrobacterium tumefaciens. The concentration of the total free amino acids of hypocotyl tissue reached a maximum 68 hours after infection with strain 1025 and this level was 50% greater than that found in plants treated with strain IIBNV6. The total content of free basic amino acids increased to a peak value at 20 hours after infection by strain 1025 and this represented 1.4 to 3 times the level found in strain IIBNV6 inoculated seedlings at 20 hours.The amount of free proline decreased with time and it was essentially absent 164 hours after infection with strain 1025. At 2 months the concentration of free proline in the tumor tissue was greatly increased with 75% of the total free amino acids accounted for as proline.The free amino acid content of strain 1025 growing in either the presence or absence of an extract from wounded seedlings was measured and the results indicated no significant relationship between the plant and bacterial systems with regard to their free amino acid pools.

During 6-day exposures of cells of Saccharomyces cerevisiae to acetate sporulation medium, the content of free amino acids declined to approximately one-third of that of vegetative cells, but proline was exceptional in that it increased conspicuously in amount. The content of combined amino acids also diminished to about one-third, ammonia was evolved, and amino acids (not including proline) passed out of the cells into the medium. When dihydroxyacetone replaced acetate in the sporulation medium, the results were similar except that the decline in content of free and combined amino acids was much greater, more ammonia was evolved, and only very small amounts of amino acids could be detected in the medium. Transfer of sporulated cells to growth medium led to an increase in the pool of free amino acids, except for proline, which declined in amount.In two other species of Saccharomyces the free proline content also increased on exposure to sporulation medium, but in Schizosaccharomyces pombe and Torulopsis famata no such increase was observed.

This paper studies the changes that occur in free amino acid and biogenic amine contents of raw meats (beef, pork, lamb, chicken and turkey) during storage (2 °C, 10 days). The meat cuts samples were harvested from a retail outlet (without getting information on the animals involved) as the following: Beef leg (four muscles), pork leg (five muscles), lamb leg (seven muscles), turkey leg (four muscles), and chicken breast (one muscle). Meat composition varied according to meat types. In general, pH, microbiology counts, biogenic amine (BA), and free amino acid (FAA) contents were also affected by meat types and storage time (p < 0.05). Chicken and turkey presented the highest levels (p < 0.05) of FAAs. Total free amino acids (TFAA) were higher (p < 0.05) in white meats than in red ones. The behavior pattern, of the total free amino acids precursors (TFAAP) of Bas, was saw-toothed, mainly in chicken and turkey meat during storage, which limits their use as quality indexes. Spermidine and spermine contents were initially different among the meats. Putrescine was the most prevalent BA (p < 0.05) irrespective of species. In general, chicken and turkey contained the highest (p < 0.05) levels of BAs, and TFAAP of BAs. In terms of the biogenic amine index (BAI), the quality of chicken was the worst while beef meat was the only sample whose quality remained acceptable through the study. This BAI seems to be more suitable as a quality index for white meat freshness than for red meat, especially for beef.

This study evaluated the effects of the canning process and different filling media on the free amino acid and biogenic amine contents of eels. The main free amino acids were histidine, taurine and arginine, which constituted 72% of the free amino acids in raw eels. All steps in the canning process significantly altered the free amino acid content of eels, relative to raw samples. The changes were influenced by the step, the composition of the frying or filling medium and the storage time. The biogenic amine contents were very low in all samples. Histamine was not detected in either raw eels or canned eels. The highest values were obtained for 2-phenylethylamine. The step of the canning process, the composition of the frying or filling medium and storage time also determined the changes in the biogenic amine contents. The biogenic amines indices were low, indicating the good quality of canned eels.

Nuptial gifts that are manufactured by the male are found in numerous insect species and some spiders, but there have been very few studies of the composition of such gifts. If, as has been proposed recently, nuptial gifts represent sensory traps, males will be selected to produce gifts that are attractive to females but such gifts will not necessarily provide the female with nutritional benefits (the ‘Candymaker’ hypothesis). We examined the free amino acid content of the spermatophylax of the cricket Gryllodes sigillatus (Orthoptera: Gryllidae) using high performance liquid chromatography (HPLC). The spermatophylax (dry weight) consisted of approximately 7 per cent free amino acids. The free amino acid composition was highly imbalanced, with a low proportion of essential amino acids (18.7%) and a high proportion of proline and glycine. The main free amino acids found in the spermatophylax appeared to act as phagostimulants: the duration of feeding on artificial gels by females was positively related to the free amino acid content of the gels. The results therefore suggest that males use free amino acids to ‘sweeten’ a relatively low-value food item. A possible function of glycine in inhibiting female movement is also proposed.

Blood from the larval, prepupal, and early pupal stages of Calliphora augttr' (F.) contains the same 18 free amino acids. In addition, hydroxyproline is present in larval and prepupal blood. The quantitative differenccs in the concent,ratiolls of these amino acids arc reported. Larval blood has the highest free amino acid content (6'6 mgjml) followcd by early pupal blood (4'6 mgjml) and prepnpal blood (3'3 mgjml). As the larva matures to the prepupa, the greatest decreases occm in the concentrations in the blood of alanine, glutamic acid, glutamine, glycine, proline, and tyrosine. The metabolic significance of these changes are discussed.

Potatoes of six cultivars (Solanum tuberosum L.) with red, purple, and yellow flesh were stored at 2 and 5 °C for 3 and 6 months, and the influence of these factors on the content of free amino acids was determined. The potato cultivar and storage time had the greatest impact on the free amino acid content. The tubers of red-fleshed (Rote Emma) and purple-fleshed (Blue Congo) potatoes contained over 28 mg/g DM of free amino acids, and the Blaue Annelise cultivar with purple flesh had over 18 mg/g DM. After 6 months, the highest increase in their content (by 36%) was recorded in tubers of the Fresco cultivar (yellow-fleshed). In the analysed potatoes, the content of alanine, proline, serine, γ-aminobutyric acid, and α-aminoadipic acid increased, while that of asparagine, aspartic acid, and glutamine decreased. Asparagine decreased to the greatest extent in “Blaue Annelise” potatoes (by 24%) and that of glutamine in tubers of Rote Emma and Vineta by 18%.

Composition of free amino acid in waxy maize DH lines was analysed and the nutritional quality of waxy maize germplasm resources was evaluated by o-phthaladehyde (OPA) pre-column derivatization reversed-phase high performance liquid chromatography (HPLC). Results obtained showed that there were at least 14 kinds of free amino acids, DH57 line had the highest (25.185 mg/g), and DH21 line had the lowest (6.203 mg/g) and the average was 12.226 mg/g. The contents of essential amino acids varied from 2.59 to 14.09 mg/g, accounting for 29.55 - 75.64% of the total free amino acids. The quality of free amino acids was evaluated by comprehensive indices with principal component analysis and the first top three DH lines were DH57, DH59 and DH55. The contents of free amino acids in 60 waxy maize DH lines had obvious genetic diversity. As to the free amino acid content, the amino acids quality of DH57, DH59 and DH55 were the best, while DH21 was the worst. Results provide a theoretical guidance for the cultivation of new varieties of waxy maize with good quality.

AbstractWild soybean (Glycine soja) is a valuable genetic resource for soybean improvement. Seed composition profiles provide beneficial information for the effective conservation and utilization of wild soybeans. Therefore, this study aimed to assess the variation in free amino acid abundance in the seeds of wild soybean germplasm collected in Japan. The free amino acid content in the seeds from 316 accessions of wild soybean ranged from 0.965 to 5.987 mg/g seed dry weight (DW), representing a 6.2-fold difference. Three amino acids had the highest coefficient of variation (CV): asparagine (1.15), histidine (0.95) and glutamine (0.94). Arginine (0.775 mg/g DW) was the predominant amino acid in wild soybean seeds, whereas the least abundant seed amino acid was glutamine (0.008 mg/g DW). A correlation network revealed significant positive relationships among most amino acids. Wild soybean seeds from different regions of origin had significantly different levels of several amino acids. In addition, a significant correlation between latitude and longitude of the collection sites and the total free amino acid content of seeds was observed. Our study reports diverse phenotypic data on the free amino acid content in seeds of wild soybean resources collected from throughout Japan. This information will be useful in conservation programmes for Japanese wild soybean and for the selection of accessions with favourable characteristics in future legume crop improvement efforts.

Using intragastric titration in dogs with gastric fistulas, dose-response studies were carried out with liver extract and with a mixture of amino acids that matched the free amino acids found in liver extract. All solutions were adjusted to pH 7.0 and osmolality to 290 mosmol x kg-1. Doses are expressed as the sum of the concentrations of all free amino acids. At each dose studied (free amino acid concentration: 2.8, 5.6, 11, 23, and 45 mM), acid secretion in response to the free amino acid mixture was not significantly different from that of liver extract. The peak response to both liver extract and the free amino acid mixture occurred with the 23-mM dose and represented about 60% of the maximal response to histamine. The serum concentrations of gastrin after liver extract and the amino acid mixture were not significantly different. It is concluded that in dogs with gastric fistula, gastric acid secretion and release of gastrin were not significantly different in response to liver extract and to a mixture of amino acids that simulated the free amino acid content of liver extract.

Protamex was selected to prepare the hydrolysate. E-tongue, free amino acid combined with soluble peptide analysis were used to detect the flavor changes of Aloididae aloidi during enzymolysis. Degree of proteolysis increased with the prolongation of enzymolysis time, and reached the maximum value at 8 hours. The content of soluble peptide of hydrolysate increased firstly and then decreased in the later process. The E-tongue could effectively distinguish the taste difference of hydrolysates at different enzymolysis time, and the hydrolysate presented strong bitterness and astringency during the whole enzymolysis. The total amount of free amino acids in the hydrolysate increased gradually, and some sweet, umami and bitter amino acids increased in varying degrees during the process of enzymolysis.
