An adaptation of a moist-chamber method is described whereby Verticillium albo-atrum, V. dahliae, and V. nigrescens can readily be isolated from herbaceous and woody hosts and easily identified. Segments of infected stems, petioles, and roots cut transversely can be observed microscopically at daily or frequent intervals to determine the origin of outgrowing fungal hyphae in the absence of troublesome contaminants.

AbstractThe research about the observation of diseases that attack Coffee plants (Coffea sp.) in Mekar Jaya village Betara subdistrict Tanjung Jabung Barat Regency,  and Agriculture Quarantine Laboratory Jambi had been done for six months. The aim of this research is to identification the disease that attack coffee plants. The research was conducted by observing the disease which in coffea plants area, and  identificaton pathogenic fungi in laboratory by moist chamber method using microscop and also fungi identifications book. The result of this research showed that there were three pathogenic fungies which attack Coffee plants namely: 1) Hemileia sp wich caused coffee leaft rust disease, 2) Cercospora coffeicola wich caused Brown eye-spot disease, and 3) Upasia salmanicolor wich caused  Upas fungi disease.  Keywords : Pathogenic fungi, Cooffee Plants Disease, Coffee Plants. AbstrakPenelitian tentang pengamatan beberapa penyakit yang menyerang tanaman Kopi (Coffea sp) di desa Mekar Jaya kecamatan Betara kabupaten Tanjung Jabung Barat dan di Laboratorium Karantina Pertanian Jambi telah dilakukan selama 6 bulan.  Tujuan penelitian adalah untuk mengetahui penyakit yang menyerang tanaman kopi.  Penelitian dilakukan dengan mengamati gejala penyakit di lapangan dan mengidentifikasi jamur patogen di laboratorium secara moist chamber dengan menggunakan mikroskop dan buku identifikasi jamur. Hasil penelitian menunjukkan bahwa ditemukan tiga jamur patogen yang menyerang tanaman kopi yaitu: 1) . Hemileia sp penyebab penyakit Karat Daun 2) Cercospora coffeicola penyebab penyakit Bercak Daun, 3) Upasia salmanicolor penyebab penyakit Jamur upas. Kata Kunci : Jamur Patogen, Penyakit tanaman kopi, Tanaman kopi

Samples of heparinized blood, or bone marrow aspirates, or cell suspensions prepared from biopsied tissues (nodes, spleen, etc. ) are routinely prepared, after Ficoll-Hypaque concentration of the mononuclear leucocytes, for scanning electron microscopy. One drop of the cell suspension is placed in a moist chamber on a poly-l-lysine pretreated plastic coverslip (Mazia et al., J. Cell Biol. 66:198-199, 1975) and fifteen minutes allowed for cell attachment. Fixation, started in 2. 5% glutaraldehyde in culture medium at room temperature for 30 minutes, is continued in the same fixative at 4°C overnight or longer. Ethanol dehydration is immediately followed by drying at the critical point of CO2 or of Freon 13. An efficient alternative method for ethanol dehydrated cells is to dry the cells at low temperature (-75°C) under vacuum (10-2 Torr) for 30 minutes in an Edwards-Pearse freeze-dryer (de Harven et al., SEM/IITRI/1977, 519-524). This is preceded by fast quenching in supercooled ethanol (between -90 and -100°C).

A new species of Cribraria, described herein as C. bicolor, appeared in moist chamber cultures on samples of the bark of Eucalyptus sp. collected at two localities in Australia. The morphology of representative specimens was examined by light and scanning electron microscopy, and micrographs of relevant morphological details of sporocarps and spores are provided. The species has a number of distinct and unique morphological features, including a glossy bright-violet globose sporotheca and a two-colored long stalk which is bright-red over the lower one-third and light yellow or lemon-yellow over the upper two-thirds. The combination of these characteristics as well as a shallow calyculus which is dark-violet when viewed under a dissecting microscope and bright red in transmitted light when mounted in lactophenol makes C. bicolor a well-defined morphospecies when compared to all other species of Cribraria. The stability of the taxonomic characters of the species was confirmed by an examination of a number of specimens.

The peatlands of the West Siberian Lowlands, comprising the largest pristine peatland area of the world, have not previously been covered by continuous measurement and monitoring programs. The response of peatlands to climate change occurs over several decades. This paper summarizes the results of peatland carbon balance studies collected over ten years at the Mukhrino field station (Mukhrino FS, MFS) operating in the Middle Taiga Zone of Western Siberia. A multiscale approach was applied for the investigations of peatland carbon cycling. Carbon dioxide fluxes at the local scale studied using the chamber method showed net accumulation with rates from 110, to 57.8 gC m−2 at the Sphagnum hollow site. Net CO2 fluxes at the pine-dwarf shrubs-Sphagnum ridge varied from negative (−32.1 gC m−2 in 2019) to positive (13.4 gC m−2 in 2017). The cumulative May-August net ecosystem exchange (NEE) from eddy-covariance (EC) measurements at the ecosystem scale was −202 gC m−2 in 2015, due to the impact of photosynthesis of pine trees which was not registered by the chamber method. The net annual accumulation of carbon in the live part of mosses was estimated at 24–190 gC m−2 depending on the Sphagnum moss species. Long-term carbon accumulation rates obtained by radiocarbon analysis ranged from 28.5 to 57.2 gC m−2 yr−1, with local extremes of up to 176.2 gC m−2 yr−1. The obtained estimates of various carbon fluxes using EC and chamber methods, the accounting for Sphagnum growth and decomposition, and long-term peat accumulation provided information about the functioning of the peatland ecosystems at different spatial and temporal scales. Multiscale carbon flux monitoring reveals useful new information for forecasting the response of northern peatland carbon cycles to climatic changes.

AbstractInfluence of fungi on multiplication and distribution of the pine wood nematode (PWN), Bursaphelenchus xylophilus, was investigated in Pinus thunbergii cuttings. Axenized nematodes and/or one of two fungi isolated from healthy and PWN-killed P. thunbergii were inoculated together into autoclaved cuttings. A close relationship between the existence and distribution of fungal hyphae, and the multiplication and distribution of PWN was observed. The PWN did not multiply when only axenized nematodes were inoculated in the absence of fungi. When fungi were present, PWN population size increased markedly. The number of nematodes was high at sites where fungal hyphae were distributed. It is suggested that the restriction of a large portion of the nematode population near the inoculation site during the early stage of disease development is closely related to restricted distribution of fungal hyphae.

The present study was conducted to determine the effectiveness of vitamin A eye ointment (VAEO) and moist chamber (MC) in preventing ocular surface disorders (OSD) in intensive care unit (ICU) patients. A total of 38 eligible patients were selected for participation in the present clinical trial. All the patients were randomly administered VAEO in one eye every 6 hr for 5 days and had a polyethylene cover (PC) placed on their other eye to create an MC that was replaced every 12 hr as well. The results of Schirmer’s test also increased by 2.06 mm in the VAEO group ( p < .001), while they showed a slight reduction by 0.15 mm in the MC group ( p = .669). VAEO was more effective in preventing OSD in ICU patients than MC and is, therefore, recommended to be used as a method of preventing OSD.
