This experiment was conducted to assess the effect of Trichoderma harzianum, soil solarization, and biofumigant on southern blight dis-ease of carrot incited by Sclerotium rolfsii isolate CS 5. A series of in-vitro and in-vivo trials laid out to select a virulent isolate of S. rolfsii against the carrot variety New Kuroda and evaluated the mustard, cabbage, cauliflower, and broccoli leaf extracts for choosing an effec-tive biofumigant against test pathogen. Mustard was the most effec-tive in inhibiting the radial growth, and sclerotia formation (80.37 and 83.37%) of S. rolfsii (isolate CS 5) at 40% level of concentration followed by cabbage leaf extract (62.22 and 68.69%). On the contrary, a total of 10 isolates of T. harzianum were screened against the test pathogen on Potato Dextrose Agar (PDA) medium for choosing a dominant isolate of T. harzianum. The isolate Th-6 was found as the most active in inhibiting the radial growth (84.44%) of S. rolfsii fol-lowed by Th-1 (75.56%). In the application of T. harzianum, biofumi-gation, and soil solarization, the treatment appeared to be the most superior in reducing pre- and post-emergence mortality of carrot during secondary field trials. The lowest southern blight disease inci-dence (10.77%), and disease severity (12.78%) were found at the same treatment. Subsequently, the yield of carrot was increased 155.18% which might be due to the reduction of carrot disease as well as the addition of organic materials in the soil.

Laboratory and production bed observations reveal that the major fungal competitors of the Chinese straw mushroom growing on sugarcane bagasse are Sclerotium rolfsii Sacc., Corticium sp., Coprinus sp., Aspergillus flavus Link ex Fries and Chaetomium globosum Kunze ex Steud. Because of their perceived importance, these five fungi were selected for control studies. The selectivity of fungicides for the control of these fungi in Chinese straw mushroom beds was tested in vitro. Benomyl, captan, carboxin, chloroneb, mancozeb and penthachloronitrobenzene (PCNB) were tested in poison agar tests at the rates of 1, 10 and 100 p/m a.i. with Chinese straw mushroom and the selective competitors. Mushroom radial growth was reduced in all treatments of carboxin and at 10 and 100 p/m of chloroneb. Compared to these, PCNB was intermediately toxic. Chinese straw mushroom was tolerant to all concentrations of benomyl, captan and mancozeb. A. flavus radial growth was highly reduced with benomyl and slightly reduced by carboxin, chloroneb and PCNB. S. rolfsii showed growth reduction at 10 and 100 p/m of PCNB, chloroneb and carboxin. Growth of Coprinus sp., Corticium sp., C. globosum and A. flavus was reduced over 90% with treatments of benomyl at 100 p/m. Fungicidal sprays were evaluated in vivo on sugarcane bagasse and coffee pulp beds. Poor pasteurization of the coffee pulp was associated with rampant development of Aspergillus fumigatus Fries. and Mucor sp. Partial superficial control of these fungi was obtained with mancozeb alone (50 to 75% reduction of visible growth). Benomyl alone and in combination with mancozeb gave excellent suppression of fungi on the mushroom beds (90% reduction of visible growth). None of the treatments controlled fungi within the beds.

In the search for antifungal agents from marine resources, we recently found that the culture filtrate of Trichoderma longibrachiatum SFC100166 effectively suppressed the development of tomato gray mold, rice blast, and tomato late blight. The culture filtrate was then successively extracted with ethyl acetate and n-butanol to identify the fungicidal metabolites. Consequently, a new compound, spirosorbicillinol D (1), and a new natural compound, 2′,3′-dihydro-epoxysorbicillinol (2), together with 11 known compounds (3–13), were obtained from the solvent extracts. The chemical structures were determined by spectroscopic analyses and comparison with literature values. The results of the in vitro antifungal assay showed that of the tested fungal pathogens, Phytophthora infestans was the fungus most sensitive to the isolated compounds, with MIC values ranging from 6.3 to 400 µg/mL, except for trichotetronine (9) and trichodimerol (10). When tomato plants were treated with the representative compounds (4, 6, 7, and 11), bisvertinolone (6) strongly reduced the development of tomato late blight disease compared to the untreated control. Taken together, our results revealed that the culture filtrate of T. longibrachiatum SFC100166 and its metabolites could be useful sources for the development of new natural agents to control late blight caused by P. infestans.

Anthracnose and blight were recorded on Houttuynia cordata Thunb. during April 2013 to December 2013. The isolated fungi from the symptomatic plants were identified as Alterneria alternata (Fr.) Keissler and Colletotrichum gloeosporoides (Penz.) Sacc. Ethanol leaf extracts of five plants viz.,Azadirachta indica L., Citrus limon L., Datura metel L., Sennaalata L. and Tagetes erecta L.were evaluated against the pathogenic fungi A. alternata and C. gloeosporoides at 5%, 10% and 20% concentrations in vitro. A. indica recorded as good inhibitor against the test fungi followed by C. limon, S. alata, D. metel and T.erecta. In vivo treatment also showed that A.indica is the most effective in controlling diseases at 10% concentration. The plants treated with A. indica were fresh and healthy up to one month of observation.J. Asiat. Soc. Bangladesh, Sci. 42(1): 41-48, June 2016

Typhoidal salmonella infections remain a challenge in the health care system in sub-Saharan Africa. Carrier status and advent of multi-drug resistant S. Typhi strains have necessitated the search for new drug leads. Hence, this study aims at investigating P. guajava and A. indica leaves for anti-salmonella activities. Guava and neem leaves were extracted by maceration in methanol and fractionated by solvent partitioning. In vitro activities were assessed by agar well diffusion and broth micro-dilution methods. Sixty male rats were randomized to 10 groups of 6 animals each for the in vivo experiments. Groups of rats except, normal control, were induced with 0.5McFarland of S. Typhi suspension orally. Treatment groups received 200 mg/kg body weight of extracts and fractions, and the control groups were treated with 14.29mg/kg body weight of ciprofloxacin and 1%v/v DMSO for 7 days post-infection. Biochemical parameters were determined spectrophotometrically. Hematological parameters were analyzed with automated hematology diagnostic machine. All fractions of P. guajava and three of A. indica inhibited S. Typhi growth with Zone of Inhibition (ZI) ranging from 11-15 mm. Active fractions inhibited 48.60-62.45% of S. Typhi biofilm formation at 25 mg/mL with Minimum Bactericidal Inhibitory Concentration (MBIC) of 0.39-12.5 mg/mL. All fractions improved body weight of treated rats and inhibited bacteremia at 44.75 and 95.94%. Hematological parameters improved in all fractions-treated rats. MDA was not significantly (p<0.05) altered in all groups. One fraction of P. guajava (ePg) lowered the elevated level in concentration of Nitric oxide (NO) while all fractions enhanced the lowered activity of SOD. Elevated (lactate dehydrogenase (LDH), aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) and bilirubin (BIL) were lowered by all fractions to various extents in treated rats. Fractions of P. guajava, and A. indica could be further considered for identification of active anti-salmonella principle(s).
