Identification and characterisation of resistance to the cereal cyst nematode Heterodera filipjevi in winter wheat

The aim of this study was to search for new sources of resistance against the cereal cyst nematode,Heterodera filipjevi, in a collection of 290 wheat accessions. The plants were inoculated with juveniles and assessed for the number of females and cysts. One percent of the wheat accessions were ranked as resistant, 16% as moderately resistant, 41% as moderately susceptible, 26% as susceptible and 15% as highly susceptible. The infection rate and the number of females and cysts per plant were significantly lower in the resistant accession Nudakota and three moderately resistant accessions Ekonomka, Katea and Lantian 12 compared with susceptible cv. Bezostaya 1. Nematode development was reduced in resistant and moderately resistant accessions. The size of females and the total number of eggs and second-stage juveniles were reduced only in Ekonomka. No significant difference in plant height, plant weight, root length, root weight and root volume were recorded for inoculated plants compared to non-inoculated plants. This study has identified four resistant wheat accessions offering new material for breeding the resistance toH. filipjevi.

This study was undertaken to identify new sources of resistance against downy mildew of cucumber caused by Pseudoperonospora cubensis [(Berk. and Cart.) Rostow]. Pot experiments were conducted with 16 accessions each of Cucumis sativus and C. hardwickii during the years 2017 and 2018 using 12 isolates of P. cubensis representing different agroclimatic zones of India. The accession, IC331627 from Dehradun, Uttarakhand showed plant disease index (PDI) ranging from 5.55-20.35 per cent with an average PDI of 11.56 and 11.87 per cent during the years, 2017 and 2018, respectively. Hence, IC331627 was identified to be resistant against the five isolates of P. cubesis (Pc12, Pc17, Pc19, Pc21 and Pc24) and moderately resistant against one isolate (Pc22). This resistant accession C. hardwickii (IC331627) could be utilized to develop mapping population to map genomic regions conferring the resistance to downy mildew in cucumber.

The most globally recognized and economically important nematode on wheat is the cereal cyst nematode (CCN) complex (1). One of the most important species of this group is Heterodera filipjevi (Madžidov, 1981) Mulvey and Golden, 1983. During regular soil quarantine control in September 2010, Heterodera sp. cysts were found in soil samples originating from a wheat field in Gunaroš, Vojvodina Province, in northern Serbia. The wheat was a winter crop grown in a dryland production system and had an average cyst density of 2.50/100 cm3 of soil. Morphologically, the cysts were golden brown and lemon shaped with a posterior protuberance. The vulval cone was bifenestrate with horseshoe-shaped semifenestra, bullae, and underbridge. Cyst measurements (n = 30) ranged as follows: cyst length (without neck): 511.50 to 899.00 μm, cyst width: 201.50 to 682.00 μm, fenestral length: 44.80 to 65.60 μm, fenestral width: 24.00 to 40.00 μm, vulval bridge length: 12.80 to 20.80 μm, vulval bridge width: 6.40 to 14.40 μm, vulval slit: 6.00 to 12.80 μm, and underbridge length: 60.00 to 112.00 μm. The second-stage juveniles had an offset head, stylet with characteristic anchor-shaped basal knobs, four incisures, and a conical tail with a rounded tip. The J2 morphometrics (n = 30) were: length: 447.30 to 611.10 μm, width: 22.40 to 25.60 μm, stylet: 20.80 to 24.00 μm, tail length: 56.00 to 68.80 μm, tail width: 14.40 to 19.20 μm, and hyaline length: 35.20 to 44.80 μm. The ITS region was used for molecular analysis. Each DNA sample was extracted from a single cyst. Sequencing was done with primers TW81 and AB28 (2). In comparison with other H. filipjevi populations, the obtained sequence (GenBank Accession No. JX235959) revealed 99 to 100% similarity. Morphological and molecular data confirmed the existence of H. filipjevi. This is, to our knowledge, the first report of H. filipjevi from Serbia. Since wheat has important socioeconomic value for Serbia, after extensive surveys, additional phytosanitary measures may be necessary to prevent the spread of this parasite. References: (1) J. M. Several soybean (Glycine max) cropping sequences were planted for 12 years in a field that, at the beginning of the test, was infested with race 14 of the soybean cyst nematode, Heterodera glycines. Continuous soybean cropping sequences included H. glycines-susceptible cultivars Forrest, J82-21, Peking × Centennial breeding line, and moderately resistant cultivars Bedford and J81-116. Forrest treated with aldicarb or pentachloronitrobenzene (PCNB) plus metalaxyl and resistant breeding line JS83-236 followed by resistant cultivars Cordell and Hartwig were additional continuous soybean sequences. Rotations included two sequences each of Bedford with J81-116 or J82-21, and three sequences of Bedford with corn (Zea mays) and susceptible Essex soybean. Rotations of Bedford, corn, and Essex had 12-year mean yields significantly greater than continuous Bedford or Forrest. The female index (FI) of H. glycines on five cultivars and lines was used to bioassay changes in parasitic potential in each cropping sequence. The FI on Bedford bioassay plants increased significantly over time for all field treatments involving Bedford. When J82-21 was the bioassay plant, FI decreased significantly in treatments involving Bedford. There were no significant changes in FI for any treatment when Forrest, J81-16, and Peking were used as bioassays. Rotations of soybean cultivars with different sources of resistance and rotations of resistant and susceptible cultivars with a nonhost crop were not successful practices to manage the nematode's ability to parasitize the resistant cultivar Bedford. However, rotation of resistant and susceptible cultivars with a nonhost crop produced greater mean soybean yields and slowed the shift toward greater parasitism of the resistant cultivar sufficiently to warrant adoption of this practice.

Meloidogyne graminicola causes significant damage to rice fields worldwide. Sources of resistance to M. graminicola reported in Oryza sativa are limited. Resistance to this species has been found in other Oryza species such as O. glaberrima and O. longistaminata. This study aimed to evaluate the reaction of four wild species of Oryza from the Embrapa Rice and Bean Germplasm Bank (Goiás, Brazil) to a pool of M. graminicola populations and determine the resistance mechanism in O. glumaepatula. Two genotypes of O. glaberrima, one of O. alta, three of O. glumaepatula, one of O. grandiglumis, one of O. longistaminata, and one of O. sativa (control) were included in the study. The results showed that O. glumaepatula was highly resistant (reproduction factor [RF] < 1). O. glaberrima, O. alta, and O. grandiglumis were considered moderately resistant. O. longistaminata was susceptible, although values of RF remained lower than the control O. sativa ‘BR-IRGA 410’, considered highly susceptible. Histological observations on the interaction of O. glumaepatula and M. graminicola showed reduced penetration of second-stage juveniles (J2s) when this resistant wild accession was compared with O. sativa. An intense hypersensitivity response-like reaction occurred at 2 days after inoculation in the root cortex of the resistant accession. Few J2s established in the central cylinder, and rare collapsed giant cells were observed surrounded by degenerate females. Fluorescence microscopy in O. glumaepatula revealed giant cells and the female body presumably exhibiting accumulation of phenolic compounds. Our study suggests that wild rice accessions, especially from the AA genotype (e.g., O. glumaepatula), are of great interest for use in future breeding programs with Oryza spp.
