A new entomopathogenic nematode, Steinernema tielingense n. sp. (Rhabditida: Steinernematidae), from north China

A new species of Steinernema was isolated from shrub soils in Liaoning province during a survey for entomopathogenic nematodes in north China. This nematode was obtained by the insect-baiting technique using last instar larvae of Galleria mellonella (Lepidoptera: Pyralidae). This new species is described herein as S. tielingense n. sp. It belongs to the ‘feltiae-kraussei-oregonense’ group and is characterised by infective third-stage juvenile (IJ) with a body length of 915 (824-979) μm, distance from head to excretory pore of 69 (64-73) μm, tail length of 81 (74-85) μm, ratio E=88 (85-94)%, lateral field with eight ridges at mid-body, first generation male spicule 88 (79-98) μm and gubernaculum 61 (49-70) μm long, and first generation female with a vulval protrusion and ratio D=41 (32-49)%. The new species distinctly differs from the related species S. kraussei, S. silvaticum, S. oregonense and S. cholashanense in the different number of ridges in the lateral fields and hyaline tail length as % of total tail length of IJ and male body length and distance from head to excretory pore. Cross hybridisation tests showed that these species were reproductively isolated. The sequences analyses of the internal transcribed spacers (ITS) and D2-D3 regions of the ribosomal DNA confirm this to be a new species.

Abstract Steinernema ashiuense sp. n. was collected by a Galleria baiting method from grassland along the riverbank of the Yura river, near the Ashiu Research Forest Station of Kyoto University. The new species is characterised by an infective juvenile body length of 768 (720-800) μm, lateral field with five equally developed ridges (i.e., six lines or incisures), head smooth lacking horn-like structures, excretory pore located at level of middle of pharynx, hyaline portion short (ca one-third of tail length). Males have 50-65 μm long, broad, slightly yellowish spicules; spicule length / spicule width = 4 (3.5-4.4), and two subventral and one subdorsal pair of genital papillae in the postcloacal region. The analysis of ITS-rDNA sequence placed S. ashiuense sp. n. in the 'feltiae-kraussei-oregonense' group in the clade containing S. robustispiculum and S. monticolum. It can be distinguished from these species by morphological characters of the infective juveniles and males.

Summary Aphelenchoides smolae n. sp. isolated in Ningbo Port, China, from medium soil and tissues of Lilium orientalis bulbs imported from The Netherlands, is described. It is characterised by the female body length of 826 (672-1002) μm and male body length of 785 (683-844) μm, lateral field with four lines (three bands), stylet 13.0-14.9 μm long with small basal swellings, excretory pore situated slightly posterior to nerve ring, vulval flap absent, post-vulval uterine sac well developed and extending for ca 35.7-62.4% of vulva to anus distance, conical tail with terminus bearing a single mucron with many tiny nodular protuberances under SEM, males common, with spicules 25.7-29.8 μm long (median curved line), the condylus and rostrum rounded, well developed, the dorsal limb with a hook-like tip, and tail bearing three pairs (2 + 2 + 2) of caudal papillae and a sharply pointed mucron. The new species belongs to the Group 2 category of Aphelenchoides species sensu Shahina. Phylogenetic analysis based on 18S, 28S D2-D3 of rDNA and COI mtDNA further characterised the new species.

AbstractA new species of entomopathogenic nematode, Steinernema phyllophagae n. sp., was isolated from a white grub (Phyllophaga sp.) in a live oak (Quercus virginiana) nursery. The new species is characterised by morphometrics of the infective Sjuvenile (IJ) with body length 1289 (1133-1395) μm, distance from anterior end to the excretory pore 99 (84-120) μm, tail 89 (77-109) μm, ratio a = 31 (27.6-35.5), H% = 57 (51-60) and E% = 110 (70-139). The pattern of the lateral field of IJ of the new species is 2, 5, 6, 8, 6, 2. The male of the first generation can be recognised by the length and shape of the spicule and gubernaculum, position of the excretory pore, D% = 75 (68-80), and GS% = 71 (61-77). The female can be recognised by the vulva having low epiptygma and by the presence of several papilla-like structures near the tail tip. Steinernema phyllophagae n. sp. differs further from other species of Steinernema by characteristics of ITS and D2D3 regions of rDNA.

AbstractA new species of entomopathogenic nematode, Steinernema khoisanae n. sp. is described from South Africa. The new species is characterised by morphometrics of the infective juvenile with body length 1076 μm, narrow body diameter of 33 μm, excretory pore 94 μm from anterior end, tail 85 μm long, a = 33, D% = 68, H% = 57, and E% = 111. The lateral field pattern of the new species is 2, 7, 8, 6, 4 and 2. The male of the first generation can be recognised by the spicule and the gubernaculum shape, excretory pore located posteriorly near the end of the pharynx, D% = 88, and SW% = 199. The first generation female can be recognised by the non-protruding vulva and tail bearing a prominent mucron. Steinernema khoisanae n. sp. is characterised genetically by sequences of the internal transcribed spacers and D2/D3 regions of 28S ribosomal DNA, by composition of their sequences and by numerous unique, derived, nucleotide character states. Phylogenetic trees show that S. khoisanae n. sp. and other members of the S. glaseri-group form a monophyletic assemblage.

A new species of the genus Nothotylenchus, N. persicus n. sp. was collected around the roots of grapevine and is described and illustrated herein based on morphological and molecular studies. The new species is characterised by a body length of 776-900 μm, delicate stylet 5-6 μm long, six lines in the lateral field, post-vulval uterine sac short, 10-18 μm long, female tail elongate-conoid with pointed terminus, and bursa covering 40-45% of tail length. Morphologically, N. persicus n. sp. appears closer to four known species of the genus, namely: N. hexaglyphus, N. affinis, N. medians and N. taylori. The results of phylogenetic analyses based on sequences of D2-D3 expansion region of 28S rRNA gene confirmed the close molecular relationship between N. persicus n. sp. and other anguinids, but Nothotylenchus claded separately from Ditylenchus species.

AbstractTwo new species of Steinernema (Rhabditida), Steinernema loci sp. n. and S. thanhi sp. n., were isolated from beach soil in Thanhhoa and Hatinh provinces in the northern part of Vietnam. A combination of morphological, morphometric and rDNA-RFLP features indicated the distinctness of both species from other Steinernema spp. Diagnostic characters of Steinernema loci sp. n. include: total body length (896-1072 μm), distance from anterior end to excretory pore (EP = 71-86 μm), tail length (66-83 μm), lateral field in mid-body with eight ridges (nine distinct lines) and rounded, broad and smooth anterior end of the third stage infective juvenile; first generation males were characterised by body width, spicule length and the ratio spicule length : anal body width (SW). Diagnostic characters of Steinernema thanhi sp. n. third stage infective juveniles include: total body length (720-960 μm), EP (68-84 μm), tail length (52-72 μm), lateral field in mid-body with eight ridges (nine distinct lines) and rounded, broad and smooth anterior end. Spicule length, ratio SW and arrangement of genital papillae characterised the first generation males.

Bursaphelenchus singaporensis sp. n. isolated in China from packaging wood of deciduous trees, imported from Singapore is described and illustrated. This new species clearly belongs to the B. xylophilus group, having males with the typically shaped spicules with a cucullus at their distal extremity, the typical position and number of caudal papillae (three pairs and one single) and the anterior vulval lip of the females developed as a distinct flap. The new species is characterized by a body length of 792 (553–950) µm and 850 (690–961) µm of males and females, respectively, robust body (a= 34 and 31, resp.), 15–16 µm-long stylet, lateral field with four lines, long postuterine branch (averaging 102 µm) and a strongly conoid female tail (c= 20) with a finely rounded, only slightly ventrally-bent terminus, male with very strong spicules (41–48 µm long), distinct rostrum and small cucullus, and a dorso-ventral visible terminal bursa. Bursaphelenchus singaporensis sp. n. is closely related to other species of the B. xylophilus group (B. xylophilus, B. mucronatus, B. kolymensis, B. fraudulentus, B. conicaudatus, B. baujardi and B. luxuriosae) and similar to B. abruptus. The morphological differentiation is mainly based on the shape of the female tail. However, B. singaporensis sp. n. differs from all other species of the B. xylophilus group by larger spicules. The new species can be differentiated from B. abruptus, B. xylophilus, B. mucronatus, B. fraudulentus, B. conicaudatus and B. luxuriosae by means of ITS-RFLP patterns.

Aoruroides chubudaigaku n. sp. (Nematoda: Oxyuridomorpha: Thelastomatidae) is described from the wood-burrowing cockroach Panesthia angustipennis spadica (Shiraki, 1906) (Blattodea: Blaberidae: Panesthiinae) from Aichi prefecture, Japan. Females of A. chubudaigaku n. sp. are similar to A. costaricensis Carreno & Tuhela, 2011 by the position of the nerve ring at level of the first third of the isthmus, the body length and the comparative measurements of the oesophagus and tail. They differ by the position of the excretory pore and the vulva. The males of A. chubudaigaku n. sp. are characterized by the absence of ornamentations in the cervical cuticle and the nerve ring located at the posterior third of the corpus. In addition, the males of the new species can be differentiated by the length of the body and the comparative lengths of the oesophagus and the tail. The phylogeny of A. chubudaigaku n. sp. is inferred by the D2-D3 domains of the LSU rDNA and the validity of the genus Aoruroides Travassos & Kloss, 1958 is discussed on the basis of both molecular and morphological data. This constitutes the first species of the genus Aoruroides described from the Japanese archipelago.

During a survey for entomopathogenic nematodes in northern China, a new species of Steinernema was isolated from soil samples collected from Xinbin county, Liaoning province. This nematode was obtained by the insect-baiting technique using last instar larvae of Galleria mellonella. It is described herein as S. xinbinense n. sp. The nematode can be separated from other described species of the group by morphological and morphometric characteristics of the different stages and by characterisation and phylogeny of DNA sequences of the D2D3 domain of the LSU or ITS regions of rDNA. This new species is characterised by the following morphological characters: infective third-stage juvenile with a body length of 694 (635-744) μm, distance from head to excretory pore of 51 (46-53) μm, tail length of 73 (61-81) μm, E = 71 (65-78)%, presence of eight unevenly spaced and developed ridges in middle lateral field (i.e., nine lines). First generation male with well curved, yellowish spicules 56 (49-62) μm long and gubernaculum 35 (30-41) μm long, small mucron mostly present, first generation female with protruding vulva, tail conical with one or two small mucrons and D = 45 (41-50)%. Cross hybridisation tests with S. tielingense, S. kraussei, S. feltiae and S. hebeiense showed that this species was reproductively isolated. The analyses of ITS-rDNA and D2D3 sequence confirm that the studied nematode isolate is a valid new species belonging to the ‘feltiae-kraussei-oregonense’ group.

During a survey of soil nematodes in Iran, a new species of Ditylenchus Filipjev, 1936 was discovered. Ditylenchus sarvarae sp. n. is characterised by its body length (1.0–1.4 mm), lateral field with seven incisures and without areolation, long postuterine sac (1.8–2.8 times the corresponding body diameter) and conical female tail with pointed tip (68–89 μm long, c = 13.7–18.2, c' = 4.2–5.1). Male specimens have conical tails (64–70 μm long, c = 14.5–16.7, c' = 4.0–4.5), spicules (22–26 μm long) and gubernaculum (8–10 μm long). Measurements and illustrations are provided for this new species. A molecular study of the 28S rDNA region of D. sarvarae sp. n. demonstrates that the Iranian species belongs to a separate group compared with the other molecularly characterized species of genus Ditylenchus.
