Prognostic Value of Long Non-Coding RNA Long Intergenic Non-Protein Coding RNA 1586 in Pancreatic Cancer

This study investigated the prognostic value of the long non-coding RNA (lncRNA) LINC01586 in pancreatic cancer. Three pancreatic cancer patients who received pancreaticoduodenectomies in our department in January 2019 were retrospectively selected. Cancer tissue and adjacent tissue samples were collected for high-throughput sequencing of lncRNAs. Among them, 221 lncRNAs were up-regulated and 235 were down-regulated. The expression of LINC01586 was decreased in pancreatic cancer patients (logFC = -3.308). An additional 74 tissue specimens were collected from pancreatic cancer patients from January 2011 to December 2016 for low-throughput validation. Patient samples were categorized into overexpression and low expression groups, based on the median LINC01586 expression level. The LINC01586 low expression group exhibited larger tumor size than the overexpression group (P < 0.001), while the low expression group exhibited a lower cancer-related survival rate than the overexpression group (one-year cancer-related survival rate: 55.6% vs. 89.2%, P < 0.001). Further analysis confirmed that low expression of LINC01586 was associated with poor prognosis for pancreatic cancer patients (OR = 0.169, 95% CI 0.066-0.437, P = 0.000). KEGG signaling pathway analysis was used to enrich LINC01586 target genes, and were mainly related to two metabolic pathways: insulin secretion (P = 0.011) and dopaminergic synapses (P = 0.0129), with SNAP25 as the core gene. The expression of LINC01586 and SNAP25were positively correlated in pancreatic cancer (R = 0.81 and P < 0.001). Together, our results indicate that LINC01586 may be used as a biomarker for prognosis predictions in pancreatic cancer patients, and its low expression is associated with poor prognosis.

AbstractLong intergenic non-coding RNA 01,087 (LINC01087) has been concerned as an oncogene in breast cancer, while its mechanism in glioma has been little surveyed. Thus, we searched the prognostic value and functional action of LINC01087 in glioma. Glioma patients after preoperative MRI diagnosis were enrolled, and LINC01087, microRNA (miR)-1277-5p, and alkaline ceramidase 3 (ACER3) levels were tested in glioma cancer tissue. The correlation between LINC01087 expression and the survival of patients were analyzed. LINC01087, miR-1277-5p, and ACER3 levels in U251 cells were altered via transfection, and cell malignant phenotypes were monitored. The relationship between miR-1277-5p and LINC01087 or ACER3 was detected. The LINC01087 and ACER3 expression was in up-regulation and the miR-1277-5p expression was in down-regulation in clinical glioma samples. High expression of LINC01087 was associated with poor prognosis of glioma patients with preoperative MRI. LINC01087 silencing restrained tumor malignancy in glioma cells. Mechanistically, LINC01087 directly interacted with miR-1277-5p. ACER3 was a known target of miR-1277-5p. Moreover, rescue assays reveal that miR-1277-5p overexpression (or ACER3 overexpression) reversed the effects of LINC01087 upregulation (or miR-1277-5p upregulation) on glioma cells. LINC01087 has prognostic significance in glioma and silencing LINC01087 deters glioma development through elevating miR-1277-5p to reduce ACER3 expression.

Background & Aims: Down-regulation of the growth arrest specific transcript 5 (GAS5) (long non-coding RNA) is associated with cell proliferation of gastric cancer (GC) and a poor prognosis. We aimed to investigate whether the variant rs145204276 of GAS5 is associated with the prognosis of GC in the Chinese population, and to unveil the regulatory mechanism underlying the GAS5 expression in GC tissues.Method: 1,253 GC patients and 1,354 healthy controls were included. The frequency of the genotype del/del and the allele del of rs145204276 were compared between the patients and the controls and between different subgroups of patients classified by clinicopathological variables. The overall survival rate was analyzed according to the Kaplan-Meier method using the log-rank test.Results: The frequency of genotype del/del was significantly lower in patients than in the controls (7.0% vs. 9.1%, p = 0.001). Kaplan-Meier analysis showed that genotype del/del was significantly associated with a higher survival rate (p = 0.01). Patients with late tumor stage were found to have a significantly lower rate of genotype del/del than those with an early tumor stage (4.9% vs. 8.8%, p = 0.01). Patients with UICC III and IV were found to have a significantly lower rate of genotype del/del than those with UICC I and II (5.3% vs. 8.1%, p = 0.02).Conclusion: The variant rs145204276 of GAS5 is associated with the development and prognosis of GC. The allele del of rs145204276 is associated with a remarkably lower incidence of cancer progression and metastasis.
