A method for demonstrating acidic polysaccharides in formalin fixed parafin embedded tissue sections using fluorescein labelled deacetylated chitin is described. Epithelial and connective tissue acidic polysaccharides have been studied in a number of organs. The distribution of acidic polysaccharides as seen with fluorescein-labelled deacetylated chitin is similar to results obtained with alcian blue-periodic acid Schiff and fluorescein-labelled Aspergillus polysaccharide. Previously reported fluorescent staining methods for acidic polysaccharides such as iron-hematoxylin-acridine orange and atabrine as well as fluorescein-labelled deacetylated chitin may prove to be useful adjuncts to the light microscopic methods of demonstrating mucins. Other possible uses of deacetylated chitin, a colorless macro-cation, in histochemistry and cytochemistry are noted.

A procedure was described for demonstrating acidic polysaccharides in formalin-fixed paraffin-embedded tissue sections using fluorescein-labelled Aspergillus polysaccharide. This material is a basic polysaccharide composed of units of galactosamine and N-acetyl galactosamine and is synthesized by the mold Aspergillus parasiticus. The distribution of acidic polysaccharides shown by the fluorescein-labelled Aspergillus polysaccharide method was similar to results with the alcian blue-periodic acid Schiff sequence and to those with fluorescein labelled deacetylated chitin. Both epithelial and connective tissue mucins were demonstrated by the fluorescein-labelled basic polysaccharide. Aspergillus polysaccharide may be considered a colorless polyvalent cation that acts as a carrier for the fluorochrome in this procedure. Since it can react with many types of biological cells as well as with various substances isolated from cells, it may have other important applications in histochemistry and cytochemistry.

The effect of basic polysaccharides on the staining characteristics of acidic polysaccharides in tissue sections of formalin-fixed paraffinembedded material has been studied. Acidic polysaccharides ordinarily do not stain with acidic dyes, but stain well with basic dyes. The complex formed when a basic polysaccharide such as Aspergillus polysaccharide or deacetylated chitin reacts with acidic polysaccharides assumes the staining characteristics of the basic polysaccharide, that is, it stains with acidic dyes. Thus, areas in tissue sections containing acidic polysaccharides can be colored with acidic dyes. Further, the basic polysaccharides were shown to temporarily block staining of acidic polysaccharides by alcian blue and mucicarmine.

The objective of this investigation was to study morphological aspects of the Harderian, paraorbital, gland in domestic geese. Tissues were obtained from 9 male and 9 female adult domestic geese and processed for light microscopy. Tissue sections were stained with Mallory’s trichrome, silver impregnation, methyl green-pyronin, periodic acid Schiff, alcian blue (pH 2.5), alcian blue/periodic acid Schiff (pH 2.5) and Sudan black B staining techniques. Results revealed the Harderian gland of domestic geese was covered by a capsule, which contributed trabeculae that divide the organ into lobes and lobules. The glandular epithelium was multilobular, compound tubuloalveolar. The secretion of Harderian gland was mucous and the secretion type was apocrine; the gland emptied to the inner surface of the nicticating membrane by a single duct.

This study was conducted to investigate the histological characteristic, type, and distribution of connective tissue in Sunda porcupine skin. The investigation was carried out in three adult of sunda porcupines at microscopic level using hematoxylin eosin, Masson thrichrome, Verhoeffs van Gieson, alcian blue pH 2.5 and periodic acid Schiff staining methods. Skin consists of epidermis, dermis hypodermis, and subcutaneous muscle. Quill follicles were the main and dominant structure as well as the specific characteristic on Sunda porcupine skin. The connective tissue was distributed well in basal membrane, dermis, quill follicle, and hypodermis with various intensity and density. The collagen was the main fiber found in the skin while the elastin fiber was not observed. The acid carbohydrate was found distributed well in the skin while the neutral carbohydrate was not detected in this study. In addition the fibers of connective tissue associated with the adipose tissue which found plentifully in quill follicles and hypodermis. The present results showed that the wide distribution of connective tissue might have an important role on the wound healing physiology of Sunda porcupine skin.

A dermal mucinoisis, visualized as dermal alcianophilic material, is occasionally present in canine hypothyroidism (myxedema). Various histochemical reactions (alcian blue/periodic acid–Schiff [PAS], alcian blue at pH 2.6, alcian blue at pH 1.0, critical electrolytical concentrations with and without dimethylsulfoxide, differential hydrolysis by hyaluronidases) were performed on skin biopsies from six dogs (four females and two males ranging from 8 to 13 years) affected by hypothyroidism, all of them presenting dermal mucinosis in hematoxylin and eosin–stained sections. In these dogs, the only polysaccharide compound involved in the dermal mucinosis was hyaluronic acid. In this study, hyaluronic acid dermal deposits of hypothyroid dogs were significantly different from those of controls in subepidermal connective tissue and loose reticular connective tissue but not in periadnexal zones. We recommend the combined alcian blue/PAS reaction as a routine technique to assess dermal mucinosis in hypothyroid dogs.

Correlated histochemical and autoradiographic studies were conducted on rat, rabbit, cat and dog salivary glands. Mucous acini of rat minor sublingual (SL) rabbit SL and cat posterior SL and submandibular (SM) glands were stained by Alcian Blue at pH 1.0, aldehyde fuchsin or the high iron diamine technique. The distribution of cells with this type of basophilia corresponded with that of cells shown autoradiographically to incorporate S35O4= and accordingly to contain sulfomucin. The lack of periodic acid-Schiff and alcohol-resistant metachromasia at pH 0.5 of rabbit SL mucin resembles connective tissue mucopolysaccharide. Some mucous acini of the dog posterior SL gland incorporate S35-sulfate and the distribution of isotope-labeled acini correlates with that of acini showing histochemical staining indicative of sulfomucin. The remainder of mucous acini in this gland do not show uptake of radiosulfate, but have alcianophilia that is reduced by sialidase digestion. All of the mucous acini of dog SM gland show correspondence of staining for sulfomucin with S35O4–, incorporation. The seromucous demilunes of this gland manifest sialidase-labile alcianophilia. The staining of sulfomucin and S35 labeling in each of the salivary glands studied are eliminated by a 4-hr methylation at 60°C.

AbstractPathology is practiced by visual inspection of histochemically stained tissue slides. While the hematoxylin and eosin (H&E) stain is most commonly used, special stains can provide additional contrast to different tissue components. Here, we demonstrate the utility of supervised learning-based computational stain transformation from H&E to special stains (Masson’s Trichrome, periodic acid-Schiff and Jones silver stain) using kidney needle core biopsy tissue sections. Based on the evaluation by three renal pathologists, followed by adjudication by a fourth pathologist, we show that the generation of virtual special stains from existing H&E images improves the diagnosis of several non-neoplastic kidney diseases, sampled from 58 unique subjects (P = 0.0095). A second study found that the quality of the computationally generated special stains was statistically equivalent to those which were histochemically stained. This stain-to-stain transformation framework can improve preliminary diagnoses when additional special stains are needed, also providing significant savings in time and cost.

Abstract Context.—We have observed intraluminal crystalloid morphology in seminal vesicles that is superficially similar to that seen in prostate neoplasia, but found little information on such morphology in the literature. Design.—Two hundred fifty-three prostate specimens (163 needle biopsies, 75 radical prostatectomies with prostate carcinoma, 11 prostates from autopsy, and 4 cystoprostatectomies without prostate carcinoma) were examined for seminal vesicle secretions, which were categorized as (a) dense platelike inspissated, (b) fluidlike, (c) crystalloid morphology, and (d) absent. Histochemical stains (periodic acid–Schiff with and without diastase, Alcian blue at pH 2.5, and mucicarmine) were performed to characterize the nature of secretions. Results.—Proteinaceous secretions were identified in 82% of seminal vesicles examined. Of these, 61% had predominantly dense, platelike, inspissated secretions, 15% had predominantly fluidlike secretions, and 24% had predominantly crystalloid morphology. Although in some cases the crystalloid morphology resembled that of prostatic intraluminal crystalloids, the seminal vesicle crystalloids differed in that they were invariably multiple, had curved edges, and had varied forms (elliptical, cylindrical, rodlike, and rectangular). Seventy-one percent of seminal vesicle crystalloids were associated with dense, platelike, inspissated secretions and appeared to be created by fracturing within platelike secretions. There was no relationship between seminal vesicle crystalloid morphology and associated malignancy in the prostate gland, as it was seen in 24% of cases with prostate carcinoma and 25% of cases without prostate carcinoma (P = 1.0000). Fluidlike secretions were positive for Alcian blue (pH 2.5) and mucicarmine, whereas dense platelike secretions and crystalloid morphology were negative for Alcian blue (pH 2.5) and mucicarmine. Conclusions.—Seminal vesicle secretions are fairly common and, when fluidlike, are composed of acid mucopolysaccharides. Inspissation of secretions appears to be associated with loss of acidity, presumably resulting in dense platelike secretions and crystallization. Awareness of both the crystalloid morphology in seminal vesicle tissue and the distinguishing features from prostatic crystalloids may be important while interpreting prostate needle biopsies in which seminal vesicle epithelium may be confused for prostate carcinoma because of a small acinar morphology with accompanying cytologic atypia and crystalloid morphology.

Larvae of the lamprey, Entosphenus lamottei (Le Sueur), and rainbow trout, Salmo gairdneri Richardson, were exposed to the sodium salt of 3-trifluormethyl-4-nitrophenol (TFM) in concentrations of 0.75 p.p.m., 3.00 p.p.m., and 6.00 p.p.m. Microscopic examination of changes induced in the gills, liver, cloacal region, and musculature were made on 7-micron sections stained with Harris' haematoxylin and Bowie's eosin, and in the gill region with periodic acid Schiff reagent. A comparison of the degree of the effects in the two species was made by planimetry of the vascular, cellular, and edematous areas from enlarged drawings of sections.Upon exposure to lethal concentrations of TFM, the body of the larval lamprey becomes distended at the pharyngeal level and heavy cords of mucus emerge from the external gill clefts. A deep red coloration is evident in the pharyngeal region consequent upon vasodilatation of the arterioles and capillaries of the gill filaments. Trout exhibit a similar vasodilatation of the gills together with increased mucous secretion. Edema in the connective tissue between the respiratory epithelium and the vascular endothelium is induced in both species. After prolonged exposure to TFM, the mucous cells in the lining of the branchial chamber and covering the tips of the gill filaments are actively discharging their secretions or completely spent.Certain effects induced by TFM in the larval lamprey are not evident in the trout. The cloacal region takes on a deep red coloration due to dilatation of the venous sinuses and the liver becomes reddish because of sinusoidal dilatation. Extensive edema of the fibrous connective tissue of the skeletal musculature is characteristically present. A slightly increased secretory activity of mucous-secreting cells may occur in the epidermis.With the techniques employed in this study, there was no evidence in either species of cytological or histological changes in the nervous tissue, cardiac musculature, notochord, alimentary canal (including the haemopoietic typhlosole of the lamprey), or mesonephros.

Background: This study was undertaken to evaluate the preferred method (Giemsa or periodic acid Schiff-Alcian blue [PAS-AB] stains) of detecting Helicobacter pylori (H. pylori) in gastric mucosal biopsies in terms of sensitivity, specificity and applicability. To the best of my knowledge, this is the first report comparing Giemsa and PAS-AB staining for the detection of H. pylori in such biopsies. Methods: The formalin-fixed paraffin-embedded blocks of 49 gastric biopsies from different patients were collected from the archive of anatomical pathology at King Abdulaziz Medical City, National Guard, Riyadh, Saudi Arabia. From each block, three slides were prepared and analysed using the hematoxylin and eosin (H&E), Giemsa and PAS-AB stains to detect the presence/absence of H. pylori, and the results were compared in terms of sensitivity, specificity and applicability. Results: The majority of the biopsies in this study showed antrum-type gastric mucosa. Only 15 biopsies showed active gastritis, whereas the rest showed chronic gastritis. Three biopsies showed intestinal metaplasia. All were detected by PAS-AB stain, but only two-thirds were detected by H&E stain. Fifteen gastric biopsies showed H. pylori infection in general and in 13 of them, active gastritis cases were discovered. Fourteen out of these 15 H. pylori infection cases were detected by Giemsa stain, whereas only 13 cases were detected by H&E stain. PAS-AB stain showed the worst results since it demonstrated only 40% sensitivity and 67.65% specificity in H. pylori detection. Conclusion: Giemsa stain has better sensitivity and specificity in gastric H. pylori infection detection than PAS-AB. Therefore, using PAS-AB stain to detect H. pylori infection is not recommended.
