Abstract Central nervous system glucagon-like peptide-1-(7–36) amide (GLP-1) administration has been reported to acutely reduce food intake in the rat. We here report that repeated intracerebroventricular (icv) injection of GLP-1 or the GLP-1 receptor antagonist, exendin-(9–39), affects food intake and body weight. Daily icv injection of 3 nmol GLP-1 to schedule-fed rats for 6 days caused a reduction in food intake and a decrease in body weight of 16 ± 5 g (P < 0.02 compared with saline-injected controls). Daily icv administration of 30 nmol exendin-(9–39) to schedule-fed rats for 3 days caused an increase in food intake and increased body weight by 7 ± 2 g (P < 0.02 compared with saline-injected controls). Twice daily icv injections of 30 nmol exendin-(9–39) with 2.4 nmol neuropeptide Y to ad libitum-fed rats for 8 days increased food intake and increased body weight by 28 ± 4 g compared with 14 ± 3 g in neuropeptide Y-injected controls (P < 0.02). There was no evidence of tachyphylaxis in response to icv GLP-1 or exendin-(9–39). GLP-1 may thus be involved in the regulation of body weight in the rat.

Abstract Besides an established medication for hypercholesterolemia, bile acid binding resins (BABRs) present antidiabetic effects. Although the mechanisms underlying these effects are still enigmatic, glucagon-like peptide-1 (GLP-1) appears to be involved. In addition to a few reported mechanisms, we propose prohormone convertase 1/3 (PC1/3), an essential enzyme of GLP-1 production, as a potent molecule in the GLP-1 release induced by BABRs. In our study, the BABR colestimide leads to a bile acid-specific G protein-coupled receptor TGR5-dependent induction of PC1/3 gene expression. Here, we focused on the alteration of intestinal bile acid composition and consequent increase of total TGR5 agonistic activity to explain the TGR5 activation. Furthermore, we demonstrate that nuclear factor of activated T cells mediates the TGR5-triggered PC1/3 gene expression. Altogether, our data indicate that the TGR5-dependent intestinal PC1/3 gene expression supports the BABR-stimulated GLP-1 release. We also propose a combination of BABR and dipeptidyl peptidase-4 inhibitor in the context of GLP-1-based antidiabetic therapy.

A avaliação da dispneia tem sido feita por meio de instrumentos como escala de Borg modificada, a escala de cores e a escala do Medical Research Council modificada (mMRC). O objetivo deste estudo foi correlacionar a frequência respiratória com a sensação de dispneia, através das escalas citadas, correlacioná-las entre si e verificar se o grau de alfabetização influenciou na resposta do paciente sobre a sensação de dispneia. Para avaliar o esforço físico utilizou-se o teste de caminhada de seis minutos. Este foi um estudo prospectivo, transversal e analítico-descritivo composto por 124 voluntários com diagnóstico de doença pulmonar. Para comparar as variáveis categóricas entre os grupos foram utilizados os testes Qui-Quadrado e exato de Fisher. Para comparar as variáveis contínuas foi utilizado o teste Kruskal-Wallis e para análise de correlação foi utilizado o coeficiente de correlação de Spearman. A idade média foi de 55,9 (± 13,08 anos), 14% eram analfabetos. Nos tempos de análise houve correlação positiva entre as escalas mMRC e Borg, r = 0,43, r = 0,61 e r = 0,55. Entre as escalas mMRC e Cores, observou-se correlação negativa. Concluiu-se que a frequência respiratória correlacionou-se com as três escalas. O grau de alfabetização não modificou a resposta do paciente em relação à sensação de dispneia.Palavras-chave: dispneia, fisioterapia, avaliação.
