Histone post-translational modification (PTM) antibodies are essential research reagents in chromatin biology. However, they suffer from variable properties and insufficient documentation of quality. Antibody manufacturers and vendors should provide detailed lot-specific documentation of quality, rendering further quality checks by end-customers unnecessary. A shift from polyclonal antibodies towards sustainable reagents like monoclonal or recombinant antibodies or histone binding domains would help to improve the reproducibility of experimental work in this field.

India, being a country with a majority youth population, has a good opportunity to strengthen its economy in the next few decades. However, this could only be possible, if the youth population is trained with the required skill-set for the job market with improved opportunities for higher education. Unfortunately, the percentage of higher education enrolment does not possess encouraging numbers due to various social & economic reasons. The ratio in the coming years can be improved by offering the higher education courses in open & distance learning mode (ODL). Recently University Grants Commission (UGC also eased the norms for the higher educational institutes to offer ODL programs. However, the present ODL programs that are being offered were challenged with (i) lack of quality checks (ii) lack of paradigm shift in the course delivery, which is still through correspondence mode (iii) the degrees offered by the distance mode are not on par with regular courses. In this view, it is time to assess the quality of the present ODL programs and the necessary steps have to be initiated to improve the quality of the program. Due to the COVID 19 pandemic condition, major parts of the world dependent on online classes for the delivery of the regular courses, in this view, various tools for delivery and assessment are being widely used/developed. The integration of these tools with the present corresponding courses shall improve the quality of the courses. Herein, we comprehensively present the above-mentioned problems and solutions to improve the quality of ODL courses in India.

Due to the ongoing problems with the quality of foods in European markets, the public authorities try to take some measures, which would reduce the amount of substandard food products offered to consumers in retailing. Three such measures – the higher frequency of quality checks conducted by public authorities, the imposition of some import barriers on the foodstuffs from countries known for their frequent breaches of quality standards in the food industry and the establishment of an information platform serving for the public pillorying of poor quality food – are studied in this paper. The experimental policy analysis was used as the research method. The results obtained show that only the higher frequency of quality checks can improve the situation of consumers. This measure is also effective in punishing the dishonest retailers, while not harming the honest ones.

Accurate identification of bovine Parainfluenza type 3 virus in bovine respiratory disease requires dependable, sensitive, and specific techniques for detection in affected animals. Immunohistochemical testing can be a rapid and reliable means of demonstration of virus in tissues from suspect cases; however, this procedure is dependent upon the quality of the antisera directed against the viral antigens. The production of rabbit polyclonal and murine monoclonal antibodies directed against bovine Parainfluenza type 3 virus and techniques for their use in fresh-frozen and formalin-fixed paraffin-embedded tissues in immunofluorescence and immunoperoxidase-based immunohistochemical tests are described.

We have established a certification system for antibodies to be used in chromatin immunoprecipitation assays coupled to massive parallel sequencing (ChIP-seq). This certification comprises a standardized ChIP procedure and the attribution of a numerical quality control indicator (QCi) to biological replicate experiments. The QCi computation is based on a universally applicable quality assessment that quantitates the global deviation of randomly sampled subsets of ChIP-seq dataset with the original genome-aligned sequence reads. Comparison with a QCi database for >28,000 ChIP-seq assays were used to attribute quality grades (ranging from ‘AAA’ to ‘DDD’) to a given dataset. In the present report we used the numerical QC system to assess the factors influencing the quality of ChIP-seq assays, including the nature of the target, the sequencing depth and the commercial source of the antibody.  We have used this approach specifically to certify mono and polyclonal antibodies obtained from Active Motif directed against the histone modification marks H3K4me3, H3K27ac and H3K9ac for ChIP-seq. The antibodies received the grades AAA to BBC (www.ngs-qc.org). We propose to attribute such quantitative grading of all antibodies attributed with the label “ChIP-seq grade”.

Advances in standardization of observational healthcare data have enabled methodological breakthroughs, rapid global collaboration, and generation of real-world evidence to improve patient outcomes. Standardizations in data structure, such as use of Common Data Models (CDM), need to be coupled with standardized approaches for data quality assessment. To ensure confidence in real-world evidence generated from the analysis of real-world data, one must first have confidence in the data itself. The Data Quality Dashboard is an open-source R package that reports potential quality issues in an OMOP CDM instance through the systematic execution and summarization of over 3,300 configurable data quality checks. We describe the implementation of check types across a data quality framework of conformance, completeness, plausibility, with both verification and validation. We illustrate how data quality checks, paired with decision thresholds, can be configured to customize data quality reporting across a range of observational health data sources. We discuss how data quality reporting can become part of the overall real-world evidence generation and dissemination process to promote transparency and build confidence in the resulting output. Transparently communicating how well CDM standardized databases adhere to a set of quality measures adds a crucial piece that is currently missing from observational research. Assessing and improving the quality of our data will inherently improve the quality of the evidence we generate.
