Diabetes mellitus and its complications are one of the most important causes of death. As a consequence of hyperglycemia in patients with diabetes there is an increased risk of concomitant diseases, and one of the important consequences of diabetes is impaired ability to repair. Healing disorders in diabetes are the result of complex pathophysiology involving vascular, neuropathic, immune and biochemical componentsAim of research: was to analyze changes in inflammation markers, microbial contamination and nonspecific immunity in skin homogenate of rats with diabetes after using sutures and skin glue.Material and research methods. Experimental studies were performed on 130 white outbred adult male rats, weight from 240 to 320 g. Diabetes mellitus was induced by using streptozotocin (Sigma, USA) (intraperitoneally - 65 mg / kg) with previous (15 minutes) injection of nicotinamide (intraperitoneally - 230 mg / kg). On the background of obesity, which was caused by 4 weeks of keeping animals on a high-fat diet. The development of diabetes mellitus 2 was confirmed by determining the concentration of glucose in the blood using a glucometer BAYER Contour Next (Germany). Animals of all groups (I-IV) under thiopental anesthesia (40 mg / kg body weight of rats) were made full-layer rectilinear incisions, 2 cm long, in the anterior-lateral region of the abdomen. All animals were divided into 4 experimental groups: І group (30 rats) – healthy rats, wound edges were sutured with Vicryl 5/0 thread (ETHICON, Inc. and Johnson & Johnson company (USA); ІI group (30 rats) – healthy rats, fixing of the wound edges was performed by applying skin glue Dermabond (ETHICON, Inc. and Johnson & Johnson company (USA)); ІII group (30 rats) – rats with diabetes mellitus, wound edges were sutured with Vicryl 5/0 thread; IV group (30 rats) – Rats with diabetes mellitus, fixing of the wound edges was performed by applying skin glue Dermabond. For control, all the results were compared with those of intact animals (10 rats). Animals were removed from the experiment on 3, 7, 28 days after surgery under thiopental anesthesia (90 mg / kg body weight of rats).The degree of contamination of the wound with opportunistic and pathogenic microflora determined by the level of urease activity using Nessler's reagent. Lysozyme activity was determined by bacteriolytic method, using as a substrate a suspension of bacteria Micrococcus lysodeicticus.Results. The obtained results prove that the wound process under conditions of streptozotocin diabetes mellitus in rats leads to a significantly greater contamination of the wound with pathogenic and opportunistic microflora and a decrease in nonspecific immunity, compared with healthy animals. Also, different effects of suture materials on the indicators of microbial contamination and immune protection were found: significantly larger deviations from the norm were found in groups of rats, where we used surgical sutures to fix the wound edges. When comparing the intensity of microbial contamination and nonspecific immunity in the long term (28 days) in the homogenate of the skin of rats with diabetes mellitus and the use of skin glue, the level of urease was 27.3% lower and the level of lysozyme 7.0% lower than in III group where we used surgical sutures.Changes in the activity of the antimicrobial enzyme lysozyme allow to assess the state of nonspecific immunity in the studied tissues. Many authors note a decrease in lysozyme activity in various bioliquids and tissues in diabetes mellitus. The results of our studies prove a significant decrease in the activity of lysozyme in the homogenate of postoperative skin wounds of rats with diabetes.Conclusion. Wound process in rats with experimental diabetes mellitus leads to reliable contamination of the wound with pathogenic and opportunistic microflora, as evidenced by increased urease activity, and reduced nonspecific immunity, characterized by reduced lysozyme activity in the homogenate of postoperative wound/scar tissue in relation to intact animals during all the terms of observation.In the remote period (28 days) in the skin homogenate of rats with diabetes where skin glue was used for wound closure, the level of urease was 27.3% lower and the level of lysozyme was 7.0% lower than that of rats with diabetes, where surgical sutures were used.

Diabetic animals induced by alloxan show severe hyperglycemia and intense catabolism characterized by the absence of insulin. Therefore, the objective of this study is to assess whether the alloxan 6CH, is able to reverse or mitigate the changes promoted by diabetes mellitus, as well as assess the effects of thymulin. In biological tests male Wistar rats were used induced to experimental diabetes by the administration of alloxan (iv 42 mg / kg). The sample comprised four groups (n = 4): G1 – control without the induction of diabetes, G2 - diabetic without treatment, G3 - diabetic treated with thymulin 12CH and G4 - treated with alloxan 6CH. The data were statistically analyzed by ANOVA followed by Tukey-Kramer test (p < 0.05). After treatment for 40 days slight decrease of glucose in animals treated with alloxan (502 ± 28) mg/dl and thymulin (500 ± 10) mg/dl was observed compared with untreated animals (563 ± 23)mg/dl. Remained unchanged feed intake and water, however, significant decrease of body weight in diabetic group (96 ± 21)g was observed compared to animals treated with alloxan (27 ± 23)g and thymulin (20 ± 16)g, fact not observed when the last two groups are compared with the control (5.1 ± 3.9)g. Significant reduction in the percentage of lymphocytes in diabetic animals (44.8 ± 2.4)% and increase in the group treated with thymulin (12CH) (83.3 ± 4.5)% was checked, when compared to the others. Animals treated with alloxan and thymulin showed clinical improvement. Based on these findings it is concluded that alloxan and thymulin improve the general state of the animal, and suggest inhibition of strong catabolism observed in diabetic animals without treatment.

Цель - исследование влияния куркумы длинной и галеги восточной на осмо- и ионорегулирующую функции почек крыс при аллоксан-индуцированном сахарном диабете и острой почечной недостаточности в эксперименте. Методика. Эксперименты выполнены на самцах крыс Wistar (n=70) с моделью сахарного диабета (1-я серия) и острой почечной недостаточности (2-я серия). В обеих сериях животные были поделены на 3 группы: крыс 1-й группы содержали на стандартном корме, крысам остальных групп в корм добавляли куркуму (2-я группа) или галегу (3-я группа) (2% от массы корма). На 7-е сут эксперимента проводили исследование диуретической и ионоуретической функций почек натощак и после 5% водной нагрузки. Концентрацию ионов в моче и плазме определяли методом пламенной фотометрии; осмотическую концентрацию биологических жидкостей - методом криоскопии; биохимические показатели крови - колориметрическим методом. Результаты. У животных с сахарным диабетом фоновый диурез, а также экскреция натрия и калия были статистически значимо выше, чем у контрольных животных. При острой почечной недостаточности наблюдался более низкий уровень диуреза и ионоуреза, особенно после водной нагрузки. Прием куркумы и галеги вызывал улучшение осмо- и ионорегулирующей функции почек у крыс с сахарным диабетом, и практически не влиял на эти функции почек при острой почечной недостаточности. Заключение. При сахарном диабете оба фитопрепарата вызывали понижение концентрации глюкозы, креатинина, мочевины и улучшение ионно-осмотических показателей плазмы крови, при этом эффект куркумы был выражен отчетливее. При острой почечной недостаточности эти фитопрепараты не давали описанного эффекта. Aim. To study effects of the phytomedicines, Curcuma longa and Galega orientalis, on osmosis- and ion-regulating renal functions in rats with experimental diabetes mellitus (DM) and acute renal failure (ARF). Methods. Experiments were performed in two series on Wistar male rats (n=70) with modeled diabetes mellitus (series 1) and acute renal failure (series 2). In each series, the animals were divided into 3 groups, 1) rats of group 1 receiving a standard diet; 2) rats of groups 2 and 3 receiving a standard diet supplemented with turmeric or galega (2% of food weight), respectively. On the 7th day of the experiment, the diuretic and ionuretic renal function was studied in fasting state and after 5% water loading. Concentrations of ions in urine and plasma were determined by flame photometry; osmotic concentrations of biological fluids were measured by cryoscopy; blood biochemical parameters were measured by colorimetry. Results. In diabetic rats, background diuresis and sodium and potassium excretion were significantly higher than in the control animals. In rats with acute renal failure, diuresis and ionuresis were significantly lower, particularly after the water loading. Turmeric and galega supplementation improved the osmotic and ion-regulating renal function in diabetic rats and left practically unchanged these functions in rats with acute renal failure. Conclusion. In rats with diabetes mellitus, both herbal remedies reduced concentrations of glucose, creatinine, and urea and improved ion-osmotic parameters of blood plasma with a more pronounced effect of turmeric. In acute renal failure, these phytomedicines did not produce the described effects.
