The efficacy of Baird-Parker agar, Baird-Parker agar with bacitracin (0.8 μg/ml), Baird-Parker agar supplemented with acriflavin (7 μg/ml), polymyxin (20 units/ml) and sulphonamides (55 μg/ml), KRANEP agar, mannitol salt agar, and Staphylococcus medium 110 agar for the isolation and enumeration of Staphylococcus species was investigated. Bovine blood agar was used as the control medium. Thirty-seven staphylococci strains representing 23 species and 19 non-Staphylococcus species were tested. None of the six selective media supported the growth of all 37 Staphylococcus strains. The number of Staphylococcus species that grew on a specific medium ranged from only the S. aureus strains on Baird-Parker agar supplemented with acriflavin, polymyxin, and sulphonamides, to all but S. warneri BG 647 on Staphylococcus medium 110 agar. Strains of Bacillus, Corynebacterium, and Micrococcus grew on all six selective media.

Corneal epithelium maintains visual acuity and is regenerated by the proliferation and differentiation of limbal progenitor cells. Transplantation of human limbal progenitor cells could restore the integrity and functionality of the corneal surface in patients with limbal stem cell deficiency. However, multiple protocols are employed to differentiate human induced pluripotent stem (iPS) cells into corneal epithelium or limbal progenitor cells. The aim of this study was to optimize a protocol that uses bone morphogenetic protein 4 (BMP4) and limbal cell-specific medium. Human dermal fibroblast-derived iPS cells were differentiated into limbal progenitor cells using limbal cell-specific (PI) medium and varying doses (1, 10, and 50 ng/mL) and durations (1, 3, and 10 days) of BMP4 treatment. Differentiated human iPS cells were analyzed by real-time polymerase chain reaction (RT-PCR), Western blotting, and immunocytochemical studies at 2 or 4 weeks after BMP4 treatment. Culturing human dermal fibroblast-derived iPS cells in limbal cell-specific medium and BMP4 gave rise to limbal progenitor and corneal epithelial-like cells. The optimal protocol of 10 ng/mL and three days of BMP4 treatment elicited significantly higher limbal progenitor marker (ABCG2, ∆Np63α) expression and less corneal epithelial cell marker (CK3, CK12) expression than the other combinations of BMP4 dose and duration. In conclusion, this study identified a successful reprogramming strategy to induce limbal progenitor cells from human iPS cells using limbal cell-specific medium and BMP4. Additionally, our experiments indicate that the optimal BMP4 dose and duration favor limbal progenitor cell differentiation over corneal epithelial cells and maintain the phenotype of limbal stem cells. These findings contribute to the development of therapies for limbal stem cell deficiency disorders.

An abbreviated procedure for the biochemical identification of Yersinia enterocolitica isolated from food enrichments on CIN (cefsulodin–irgasan–novobiocin) agar was investigated. A total of 170 colonies resembling Y. enterocolitica in colonial morphology and appearance on CIN agar were selected for identification using API strips. Ninety-three of these isolates were examined with the PathoTec ornithine decarboxylase, Voges–Proskauer, and urease test strips. The PathoTec urease strip alone was adequate for identification of all isolates of Y. enterocolitica. Christensen's urea agar was applied to the remaining 77 isolates and found less specific in that 1 isolate of Enterobacter agglomerons was urease positive along with 10 isolates of Y. enterocolitica. CIN agar is a highly specific medium for isolation of Y. enterocolitica, requiring only Kligler iron agar and urea slants for confirmation of presumptive colonies.

AIM: A previous study confirmed the contamination of water sources with this parasite in Arak, Markazi Province, Iran. The current study investigated soil sources and determined the predominant genotype of Acanthamoeba in this region of Iran. MATERIAL AND METHODS: Forty-eight soil samples, collected from different regions of Arak, Markazi province, Iran, were evaluated in this study. The samples were processed and identified by culturing on a specific medium, performing PCR assay, and sequencing the PCR products. Finally, using the NCBI database, the genotypes were determined. RESULTS: Of 48 soil samples, 33.3% and 31.25% were contaminated with Acanthamoeba according to the culture and molecular assays, respectively. The majority of these isolates belonged to the T4, T5 and T6 genotypes of Acanthamoeba. CONCLUSION: The genotypes of most isolates from soil samples in Arak similar to other regions of Iran belong to T4 genotype of this parasite. New sequence accession numbers include MG066681 and MG298785-MG298794.

Since stories increasingly take on pictorial and mixed-medial forms, narratology needs to investigate to what extent narrative devices exceed the boundaries of a specific medium. One way to examine this issue is to focus on film adaptations of narratologically complex novels or stories. This article presents a detailed comparison of the narration in McEwan’s (1982) [1981] The Comfort of Strangers and Schrader’s (1990) film based on a scenario by Harold Pinter. It is shown how the novel creates deliberate confusion (via free indirect speech and thought) about the agency responsible for the conveyance of crucial information, and how the film finds non-verbal means to achieve the same effect.

A pregnant heifer with an advanced clinical stage of paratuberculosis was reported in a herd in Argentina. Thus, the animal was euthanized and samples of organs of the cow and its fetus was taken and cultured for bacteriology in specific medium. Tissues were analyzed by histopathology (hematoxylin-eosin and Ziehl-Neelsen staining). Histopathological analysis of the cow’s samples revealed the presence of lesions consistent with paratuberculosis, and Ziehl-Neelsen staining revealed the presence of acid-fast bacilli, whereas the fetal tissues showed absence of lesions but the presence of acid-fast bacilli by Ziehl-Neelsen staining. After growing in specific medium, colonies in tissues from both cow and fetus were positive for IS900-PCR, confirming the presence of Mycobacterium avium subsp. paratuberculosis (MAP). Finally, the isolates were typed by Multiple-Locus Variable-number tandem-repeat Analysis (MLVA), which confirmedthe epidemiological link between them. This study is the first in Argentina to report the detection of MAP that shares an identical MLVA type in a pregnant cow and its fetus. The results of this study are consistent with previous reports and highlight the intra-uterine transmission of MAP as an important source of infection within herds.

This chapter focuses on aspects of the technological and interface dimensions of Badrul Khan's model, arguing that a correlation exists between the medium of instruction, students' performance, and the instructional content. Media-based learning is not necessarily more effective, simply because it uses a medium. Several variables exist that influence its success: the medium itself, its properties, production and consumption restraints; the content, and the way it can be presented in the context of a specific medium, and learners' cognitive styles. All these variables and more have to be taken into consideration, alone and interacting, in order to decide whether and where media-based learning is to be used, and where it might be counterproductive.
