Summary.Changes in the state of gut-associated lymphoid tissue (GALT) and the composition of the intestinal microbiome, both in experimental STZ-induced diabetes and in development of type 1 diabetes in humans as well as chronic inflammation due to stimulation of innate immunity are crucially important in the development of type 1 diabetes mellitus. One of the most important mediators for interactions between the intestinal microbiome and GALT are specialized M cells of the follicle-associated epithelium, providing transcytotic delivery of antigens to the underlying lymphoid structures. TNFα-signaling also plays a supporting role in the formation of M cells. Therefore, the aim of our work was to study some features of TLRs expression and transcriptional activity of the Gp2, Spi-B, Nf-kB1, c-Rel, TNFα and TNFr genes in GALT in experimental diabetes mellitus (EDM), and after pentoxifylline administration. To identify TLR2+ cells and TLR4+ cells, an immunofluorescence method was used with monoclonal antibodies to corresponding pattern-recognizing receptors. To study the transcriptional activity of genes, the method of real-time reverse transcription polymerase chain reaction (RT-PCR) was used. In the course of developing experimental pathology, at the terms of 2 and 4 weeks, a decrease in the total density of TLR2+ and TLR4+ lymphocytes was observed in lamina propria of villus (villus) and subepithelial zone isolated lymphoid follicles (ILF) of rat ileum. At the same time, the density of TLR2 on the membrane of immunopositive cells was increased for small lymphocytes, and TLR4 density has became higher in medium and small lymphocytes. The pentoxifylline administration to diabetic rats resulted in a decrease in the total density of TLR2+ cells at the 2nd week of development of the pathology, and an increase in this index at the 4th week. The total density of TLR4+ cells showed changing growth rates only in villus at the 2nd week of EDM development in the presence of pentoxifylline. Changes in the density of TLR2 and TLR4 on the surface of lymphocytes were multidirectional. The development of diabetes is also reflected in the transcriptional induction of genes of the key transcription factors NF-kB1 and c-Rel in GALT cells at both the 2nd and 4th week of the development of EDM. Meanwhile, administration of pentoxifylline resulted in a significantly reduced level of normalized expression of NF-kB1 mRNA during the entire observation period and increased this indicator for c-Rel mRNA at the 2nd week. The growth of normalized expression of markers of M cells Gp2 and Spi-B was observed both on the 2nd and on the 4th week of the development of experimental pathology. Administration of pentoxifylline to diabetic animals was largely reflected in the change in the intensity of mRNA expression of the mature M cell Gp2 marker. This parameter was increased during the 2nd week of developing pathology, and on the 4th week, a downward trend was shown. The development of EDM led to a significantly increased level of near-normalized expression of proinflammatory TNFα cytokine and its receptor TNFr, and demonstrated a trend towards their decrease following pentoxifylline administration in diabetic animals.

Abstract Background and aims: Diabetes mellitus (DM) is a multifactorial metabolic disorder characterized by hyperglycaemia caused by insulin deficiency or insulin resistance. It is a global public health problem. This study aimed to determine specific pharmacological effect of quercetin in water soluble and liposomal preparations in experimental diabetes mellitus. Material and methods: We examined the effect of Corvitin and Lipoflavone (at the dose of 10 mg / kg body weight) in a comparative study in white rats with type 1 diabetes and type 2 diabetes coupled with obesity. To simulate the forms of diabetes mellitus most analogous to those in humans we used Streptozotocin at the doses of 30 mg / kg and 50 mg / kg. We tested the levels of glucose, glycosylated hemoglobin, C-reactive protein, and interleukins 6 and 4 in the blood. Results: In animals with type 1 and type 2 diabetes Lipoflavone significantly reduces glucose and glycosylated hemoglobin levels compared to the rats treated with Corvitin. When administered to animals with diabetes, the effect of quercetin in liposomal form on the concentrations of IL-6, IL-4 and Creactive protein is also larger compared to the water-soluble form. Conclusions: Water soluble quercetin preparation Corvitin and to a larger extent liposomal preparation of this flavonoid, Lipoflavone, show anti-inflammatory effect and restore key parameters of carbohydrate metabolism in experimental type 1 diabetes mellitus and type 2 diabetes coupled with obesity, reducing blood glucose and glycosylated hemoglobin levels.

The aim is to study morphofunctional structure of NALT (nasal associated lymphoid tissue) and visceral lymph nodes of rats with experimental type 1 diabetes mellitus (DM-1) and to define the effectiveness of the treatment with herbal drug “Imupret”. Materials and methods: The experiment involved 20 animals, divided into 4 groups: the 1st group was the control group of healthy rats, the 2nd group was the control group subject to prophylactic treatment with “Imupret, the 3rd group included rats with experimental diabetes, and the 4th group included rats with experimental diabetes subject to treatment with “Imupret. Functional changes in immune organs were evaluated by the results of morphometric analysis; morphological pattern was evaluated by histostructural changes. Results. The research revealed that under conditions of diabetes mellitus type 1, the volume, area and density of the lymphoid tissue decreased, and only its "fine" cell was detected. The paper demonstrates the development of relative immune deficiency in immunocompetent organs in rats with diabetes mellitus type 1. The use of drug "Imupret" demonstrated its immunomodulatory function, which is especially important in terms of immunosuppression in patient with DM-1. Conclusion. The received results are of a great clinical significance, and show the necessity of early prevention and treatment of immunity disorders under conditions of diabetes mellitus type 1.

Aims:analysis of the sperm viability of mature offspring of female rats with experimental type 1 diabetes.Materials and methods:The studies were conducted on white laboratory rats (Wistar), in which type 1 diabetes was simulated, and their mature offspring.A comparative analysis of the total sperm count in the epididymal suspension, as well as their subpopulation composition by viability, with the subsequent determination of the index of motor activity, the content of atypical forms of sperm is estimated.Results:It was found that the offspring of experimental animals has a decrease in the total sperm count in 1 ml of epididymal suspension.In this case, an increase in the number of atypical forms and inhibition of the viability of spermatozoa occurs, as evidenced by a decrease in their motor activity.Conclusions: The results obtained convincingly indicate that offspring of mothers with experimental diabetes mellitus are born with a violation of the morphofunctional formation of the male reproductive system.
