The advancement and quality of science rely on research that is robust and unbiased in its experimental design, execution, analysis, and reproducibility. In preclinical research, a better understanding of animal emotions and refinement of their husbandry, housing, and handling are important goals in providing good animal welfare in a laboratory setting which underpins rigorous research quality. Induction of positive emotional state in animals is a key component of their well-being, and one approach is to increase their environmental complexity using, for example, ball pits or playpens in rats. In this study, we recorded 50 kHz ultrasonic vocalisations (USVs) during animals’ exposure to the ball pit and playpen. We have previously shown that 50 kHz USVs provide a graded and quantifiable measure of an animal’s emotional state, and here find that access to the ball pit and playpen increases 50 kHz USVs, indicative of a more positive affective state. Using our affective bias test (ABT) we next quantified the animals’ emotional response to an aversive intervention and whether this could be attenuated by access to a playpen. The playpen exposure completely mitigated the negative affective state induced by an anxiogenic drug when compared with animals who experienced the drug in the home cage. Together, these findings suggest ball pits and playpens provide a simple and effective method to improve the welfare of laboratory rats and reduce the cumulative suffering they experience from their housing conditions and minor, aversive procedures.

Background. Osteoporosis is a progressive systemic bone disease that results in decreased bone mineral density and, as a result, increases the risk of bone fractures. Changes that occur in the subchondral bone in osteoporosis or because of hormones can cause degenerative changes in the articular cartilage that underlie osteoarthritis. Objective. The aim of the study was to identify and compare morphological changes that occur in the subchondral bone in experimental simulations of osteoporosis and in adult rats that were born from females that were exposed to glucocorticoid solution administration in the 3rd trimester of pregnancy. Methods. Tibias of 26 white mature laboratory rats were studied. In serial sections, the relative areas occupied by bone trabeculae and lacunae were calculated. Results. The relative area involving the bone trabeculae (23,2 ± 3,70%) statistically significantly decreased at 21 day in the group of animals that undergone a simulation of osteoporosis in comparison with the control group. Similar changes are observed in the group of experimental animals that were born from females that were exposed to glucocorticoid solution administration in the 3rd trimester of pregnancy. Conclusion. Thus, the results of the study demonstrate the similarity of morphological changes occurring in the subchondral bone in rats that undergone a simulation of osteoporosis and rats that were born from females that were exposed to glucocorticoid solution administration in the 3rd trimester of pregnancy.

Background. In Ukraine, there has recently been an increase in the number of limb bone fractures among the population, in particular, due to increase in the number of elderly people, which is associated with the development of age-related osteoporosis and fragility of bones. Therefore, the use of artificial implants in traumatology is becoming increasingly important. The search for new bioinert and biodegradable materials, that are capable of providing rapid fracture consolidation and do not require repeated surgical intervention, continues. Objective. To elaborate a model for the research of using carbon-carbon composite nail for intramedullary osteosynthesis in laboratory rats with fractures of femur and tibia in norm and with experimental osteoporosis. Methods. The work investigated the lower limbs of 6 groups of Wistar rats. The first part of the study involved 4 groups of rats with modeled tibial fracture with subsequent osteosynthesis in norm and with experimental osteoporosis. The second part of the research included two groups of laboratory rats with modeled femur fracture. In both parts we used metal injection needle, as well as carbon-carbon composite nail for osteosynthesis. Results. All animals underwent surgery well, stepped on the injured limb. X-ray examination after the osteosynthesis demonstrated satisfactory reposition of the fragments. One month after modeled fracture in rats with experimental osteoporosis a violation of the normal consolidation of the fracture was observed, especially in the group where carbon-carbon composite nails were used for osteosynthesis. Further, until the 180th day after the surgery there were no peculiarities, function of the limbs was restored, mainly without expressed angular deformities. Conclusion. The above mentioned model was successfully elaborated for the research of using carbon-carbon composite nail for intramedullary osteosynthesis in laboratory rats with fractures of femur and tibia in norm and with experimental osteoporosis.

The presence of laboratory rats that are maintained and bred for laboratory purposes or laboratory observations is very necessary. The presence of endoparasite in laboratory rats will have an impact on the result of the research or laboratory observations. This study aims to detect helminth endoparasites in the liver of laboratory rats (Rattus norvegicus) from animal breeders in Banyumas and Purbalingga Districts. This research was an observational study with a cross-sectional design. A total of 52 laboratory rats were used in the study. Rats are killed with chloroform, liver surgery then identify the presence of worm larvae. Out of the 52 rats obtained, 7 (29.17%) from 24 laboratory rats in Banyumas District and 5 (17.86%) from 28 laboratory rats in Purbalingga District were infected with Taenia taeniaeformis. It is necessary to control helminth infections in laboratory rats, such as laboratory animal quarantine, health monitoring, and antihelmintic treatment. It is important to handle carefully during travel to assure the results of research or laboratory observations using the animals.

In laboratory animal science (LAS) education and training, five simulators are available for exercises on handling and routine procedures on the rat, which is—beside mice—the most commonly used species in LAS. Since these simulators may have high potential in protecting laboratory rats, the aim of this study is to investigate the simulators’ impact on the 3R (replace, reduce, refine) principle in LAS education and training. Therefore, the simulators were evaluated by 332 course participants in 27 different LAS courses via a practical simulator training workshop and a paper-based two-part questionnaire—both integrated in the official LAS course schedule. The results showed a high positive resonance for simulator training and it was considered especially useful for the inexperienced. However, the current simulators may not completely replace exercises on live animals and improvements regarding more realistic simulators are demanded. In accordance with literature data on simulator-use also in other fields of education, more research on simulators and new developments are needed, particularly with the aim for a broad implementation in LAS education and training benefiting all 3Rs.

 A method for revealing genetic affinity between laboratory rats has been developed, which is based on the transplantation of ear skin-chondrocyte allografts onto the surface of a full-thickness skin defect. On the example of Wistar rats (laboratory rats were taken from three research institutes), it was shown that the rejection of allografts occurs in the interval between 8 and 21 days with the same genetic differences that existed between the animals. However, at the same time, only those structures of ear allografts such as the dermis, epidermis and its derivatives die and are rejected, but the layers of chondrocytes are submerged under the granulation tissue and remain in a viable position for at least 210 days. The latter phenomenon is probably due to the membranes that cover the chondrocytes and act as an immunological barrier.

AbstractRepeated methamphetamine use leads to long lasting brain and behavioral changes in humans and laboratory rats. These changes have high energy requirements, implicating a role for mitochondria. We explored whether mitochondrial function underpins behaviors that occur in rats months after stopping methamphetamine self-administration. Accordingly, rats self-administered intravenous methamphetamine for 3 h/day for 14 days. The mitochondrial toxin rotenone was administered as (1 mg/kg/day for 6 days) via an osmotic minipump starting at 0, 14 or 28 days of abstinence abstinence. On abstinence day 61, expression of methamphetamine-induced behavioral sensitization was obtained with an acute methamphetamine challenge in rotenone-free rats. Rotenone impeded the expression of sensitization, with the most robust effects obtained with later abstinence exposure. These findings verified that self-titration of moderate methamphetamine doses results in behavioral (and thus brain) changes that can be revealed months after exposure termination, and that the meth-initiated processes progressed during abstinence so that longer abstinence periods were more susceptible to the consequences of exposure to a mitochondrial toxin.

This present work is devoted to the study of cumulative properties of the complex means Z-88. The studies were carried out on intact mongrel laboratory rats that had previously passed quarantine measures for 12 days. Two equivalent groups were created. The animals of the experimental group were injected intramuscularly in the thigh area with Z-88, and the control group was similarly injected with 0.9% sterile sodium chloride solution. On the first day of the experiment, the rats were injected with test drugs at a dose of 0.5 ml per animal, which was 1/10 of the maximum volume used in experiments to determine the parameters of acute toxicity. Both Z-88 and sterile saline solution were administered once a day throughout the experiment. Z-88 was administered once a day throughout the experiment. Starting from the fifth day, in every subsequent four days, the dose of the drug was increased by 1.5 times. The duration of the experiment was 24 days. During the experimental period of visible changes on the part of the studied indicators.
