Mortality from acute selective portal vein occlusion (SPVO) is a matter of concern for surgeons during the management of traumatic portal vein injury. However, mortality rates related to different periods of SPVO remains undetermined. PURPOSE: To determine the mortality rates resulting from different periods of acute SPVO in rats. METHODS: Wistar male rats were randomized into 8 experimental, and 8 control groups. Experimental animals underwent SPVO during 15 to 75 minutes, and control groups underwent sham procedures. All surviving animals were followed up to 14 days for assessment of mortality rate. RESULTS: Death rates varied from 0% in the 15 min SPVO group, to 100% with 65 and 75 minutes of SPVO. A strongly positive correlation was observed between mortality rates and SPVO periods (p<0.001) with either linear or quadratic regression analysis tests. All deaths in the 20min and 25min SPVO groups occurred after 75 min from the moment of clamping (or after 60 min from unclamping); death from 30 or more min SPVO occurred predominantly within 75 min from clamping moment (or within 60 min from unclamping). (Exact Fisher test, p=0.009). CONCLUSIONS: The mortality from SPVO in rats increases with longer duration of SPVO; with deaths occurring later for short periods (< 25 min) of SPVO and earlier for longer periods (> 30 min) of SPVO.

Intravenous perfluorocarbons (PFC) have reduced the effects of decompression sickness (DCS) and improved mortality rates in animal models. However, concerns for the physiological effects of DCS combined with PFC therapy have not been examined in a balanced mixed-sex population. Thirty-two (16 male, 16 female) instrumented and sedated juvenile Yorkshire swine were exposed to 200 feet of seawater (fsw) for 31 min of hyperbaric air. Pulmonary artery pressure (PAP), cardiac output (CO), and systemic arterial pressure (SAP) were monitored before (control) and after exposure. Animals were randomized to treatment with Oxycyte (5 ml/kg; Oxygen Biotherapeutics, Inc., Morrisville, NC) vs. saline (control) with 100% oxygen administered upon DCS onset; animals were observed for 90 min. Parameters recorded and analyzed included PAP, CO, and SAP. In all animals PAP began to rise prior to cutis marmorata (CM) onset, the first sign of clinical DCS, generally peaking after CM onset. Female swine, compared with castrated males, had a more rapid onset of CM (7.30 vs. 11.46 min postsurfacing) and earlier onset to maximal PAP (6.41 vs. 9.69 min post-CM onset). Oxycyte therapy was associated with a sustained PAP elevation above controls in both sexes (33.41 vs. 25.78 mmHg). Significant pattern differences in PAP, CO, and SAP were noted between sexes and between therapeutic groups. There were no statistically significant differences in survival or paralysis between the PFC and control groups during the 48-h observation period. In conclusion, Oxycyte therapy for DCS is associated with a prolonged PAP increase in swine. These species and sex differences warrant further exploration.

Prizidilol (SK&F 92657-A2), an anti-hypertensive agent, has undergone a range of prescribed toxicity studies required for the investigation of possible adverse drug effects. During the second year of the 2-year rat oral study, a variety of symptoms were exhibited by males receiving 1600 mg of the compound day–1kg–1 by gavage. These animals became lethargic, slouched and developed dyspnoea which became progressively more severe during the course of the study. Necropsy of the affected rats revealed severely haemorrhagic lungs, cardiac hypertrophy and lordosis of the spine into the thoracic cavity. At the 2-year terminal kill, a proportion of the male rats receiving 100 and 400 mg of prizidilol day–1kg–1 were identified with similar but less severe spinal deformation. No female was found with spinal changes but all the rats receiving prizidilol showed haemorrhagic lungs and enlarged hearts. The lordosis of the affected males was always confined to the thoracic spine and this, along with the cardiac hypertrophy, presumably led to marked reduction in the volume of the thoracic cavity, inducing the dyspnoea. Thoracic vertebral body damage, possibly a precursor to the spinal deformities, was found in male rats from both drug-treated and control groups. The nature of the spinal lesion is at present under investigation.

Insecticide-treated cotton seeds can pose risks to Apis mellifera Linnaeus, 1758 populations during crop establishment if chemical residues reach these insects near agricultural fields via dust drift produced during planting. However, the treatment of seeds with insecticides is essential to protect cotton plants from damage caused by pests, including thrips and aphids. Thus, the aim of this study was to evaluate the toxic effects (acute toxicity) of soil dust from cotton fields planted with insecticide-treated seeds on A. mellifera adults using a toxicity assay developed in the laboratory. Forager honeybees were maintained in 700-ml plastic cages with 7 g of surface soil where insecticide-treated cotton seeds were sown (270 g a.i. clothianidin, 270 g a.i. imidacloprid, 210 g a.i. thiamethoxam, and 75 g a.i. fipronil/100 kg of seeds, and an untreated group). Ten bees were placed in each cage. The experimental design was randomized, with five treatments and twelve replicates. The mortality rate was evaluated during the entire assay. Data were transformed to x + 0.5 &nbsp;and compared with a regression analysis and contrast test. The linear regression model revealed a significant relationship between bee mortality and exposure time. The mortality rate gradually increased as time progressed in all treatments and the control group. The contrast test did not reveal significant differences between the insecticide and control groups. Thus, residues of the products tested, thiamethoxam, clothianidin, imidacloprid, and fipronil, did not influence the mortality of A. mellifera adults as a result of exposure to soil contaminated with insecticide-treated seeds.
