The aim was to develop a timed artificial insemination (TAI) system in suckled beef cows. Cows (n=227), 60-80 days postpartum, received estradiol benzoate (5mg) and a vaginal device containing 250µg of medroxyprogesterone acetate (MPA; day 0). On day six, cloprostenol (125µg) and eCG (400IU) were administrated and calves were weaned for 88h. The devices were removed on day seven (BioRep group) or on day eight (TAI group). All cows of TAI group and cows of BioRep group that did not exhibit standing estrus received GnRH (100µg) on day 9. In experiment I, the follicular growth was monitored daily by transrectal ultrasound exams, from day 6 to day 9. The average size of the dominant follicle on day nine was 11.1±0.99mm (BioRep, n=7) and 11.5±0.65mm (TAI, n=7) and all animals ovulated. In experiment II, the BioRep group cows (n=106) were observed for estrous behavior after withdrawal of the device, twice a day for 48h, and inseminated 12h after detection. In the TAI group (n=107), the devices were withdrawn on day eight and after 24h these cows and those from the BioRep group, which were not stand in estrus, received 100µg of GnRH and TAI 16h later. The pregnancy rates were 57.6% (BioRep) and 52.3% (TAI). In conclusion, an increase on MPA exposure time did not affect the follicular dynamics and pregnancy rates and allow TAI without estrous observation. Furthermore, the treatment for eight days provides an efficient TAI system in suckled beef cows.

Abstract Advancements in reproductive technologies have improved pregnancy rates in postpartum cows; however, undesirable consequences leading to reduced fertility still occur. Thus, the objectives were to determine if presence of a corpus luteum (CL) at the start of estrous synchronization (AIM1) or if the length of proestrus (interval from progesterone removal to a GnRH-induced LH surge; AIM2) alters fixed timed artificial insemination (FTAI) pregnancy rates. Cycling postpartum cows (n = 285) at two locations were synchronized using the CO-Synch+CIDR® FTAI protocol. Ultrasonographic measurements of luteal area were recorded at CIDR insertion. To determine the impact of proestrus length, cows were randomly assigned to either undergo CIDR removal at the time of PGF2a injection (long proestrus, n = 145), or CIDR removal 24 hours post PGF2a injection (short proestrus, n = 140). Blood samples were collected at CIDR insertion and at insemination for progesterone (P4) analysis. Pregnancy status to FTAI was assessed using ultrasonographic diagnosis at 30 d post insemination. Data were analyzed using a MIXED procedure of SAS and examined for effects of length of proestrus, P4 concentration, luteal area, sire, location, and age. Statistical significance was determined at P0.05) pregnancy rates. Pregnancy rates were greater in cows with >1 ng/mL P4 compared to cows with 0.05). Cows > 4 y old had higher AI pregnancy rates compared to 2–3 y cows (70.2±7.4 vs. 51±5.8%, respectively; P > 0.05). Based on these data, incorporating a presynchronization program to ensure the presence of a functional CL at the start of a FTAI protocol may improve pregnancy success in postpartum beef cows.

Two synchronization protocols for fixed-timed AI (FTAI) in White-tail deer were evaluated over a 2-year period. InYear 1, White-tail does (n = 38), with a mean body weight of 55 kg and mean age of 2.4 years, were stratified by weight, age, and last fawning date randomly across 2 estrous cycle synchronization treatment groups. Does received either a CIDR-G device for 14 days (CIDR 14) and underwent AI 60 h post-CIDR removal, or received a CIDR-G device for 7 days (CIDR 7) with 1 mg of estradiol benzoate (i.m.) at CIDR insertion (Day 0), 25 mg of prostaglandin F2α (PGF2α; Dinoprost) on Day 6 and 1 mg of estradiol benzoate (i.m.) on Day 7 with AI occurring 52 h post-CIDR removal. In Year 2, White-tail does (n = 36), with a mean body weight of 60 kg and mean age of 3.9 years, were stratified as described for Year 1 and allotted to 2 treatments. Does received a CIDR-G device for 14 days and were inseminated 60 h post-CIDR removal (Treatment A) or were synchronized in the same way as does inTreatment A but also received 200 IU (i.m.) of eCG (Sigma, St. Louis, MO) at CIDR removal (Treatment B). All does received 1 mg of Domosedan (i.v.) before initiating the AI procedure. In both years, electroejaculated semen was collected from a single buck and frozen for AI. In both years, clean-up bucks were introduced into the pen with does for natural cover at no less than 14 days following AI. A gestation length of 195 � 7 days was used to determine whether the fawn(s) resulted from AI or from natural mating. In Year 1, FTAI pregnancy rates were not different between the CIDR 14 (56%) and CIDR 7 (24%) treatment groups. Likewise, pregnancy rates were not different between CIDR 14 FTAI (56%) and clean-up bucks (75%). However, the FTAI pregnancy rate was lower (P < 0.001) for the CIDR 7 treatment group (24%) compared with the clean-up bucks (100%). Treatments did not affect fecundity rates; however, those females pregnant from FTAI had lower (P < 0.001) fecundity rates compared with those females pregnant from natural cover (1.2 v. 1.9 fawns/doe). In Year 2, treatment did not affect FTAI pregnancy rates across treatments (33% for Treatment A and 55% Treatment B); however, fewer (P = 0.024) does in Treatment A were pregnant following FTAI (33%) compared with those pregnant from clean-up bucks (75%). There was no difference in FTAI fecundity rates across treatment groups (1.3 v. 1.7 for Treatments A and B, respectively) or between FTAI treatments and natural cover (1.3, 1.7, and 1.5 for Treatments A, B, and clean-up bucks, respectively). These results indicate that the use of a 14-day CIDR synchronization protocol with FTAI occurring 60 h after CIDR removal yields acceptable pregnancy rates of White-tail does following transcervical AI.

Abstract: The objective of this work was to compare two types of ovulation inducers - estradiol benzoate (EB group) or prostaglandin F2α (PG group) -, in postpartum dairy cows subjected to timed artificial insemination (TAI). Lactating Holstein-Gir crossbred cows (n=118) with 60 to 120 days of milk production were used in this study. All cows were treated with an intravaginal progesterone-releasing device (CIDR) plus 2 mg of estradiol benzoate (EB) on day 0. The CIDR inserts were removed on day 9, and all cows received 500 µg sodium cloprostenol. Twenty-four hours after CIDR removal, cows were randomly assigned into two treatment groups and were given either 1 mg of EB i.m. (EB group) or 500 µg sodium cloprostenol i.m. (PG group). TAI were performed at 52 to 54 hours, in both EB and PG groups, after CIDR removal. No differences were observed in the ovulation (85.2% vs 72.7%) and pregnancy rates (40.7% vs 38.2%) between the EB and PG groups, respectively. Fertility is similar in the TAI protocols that use prostaglandin F2α or EB to induce ovulation in lactating dairy cows.

The objective of the experiment was to evaluate the effects of recombinant bovine somatropin (rbST), and the reuse of the progesterone (P4) releasing devices in resynchronization, on the pregnancy rates of Nellore cows submitted to fixed-time artificial insemination. A group of 123 multiparae Nellore cows, was submitted to a resynchronization protocol: on day 0 a Bovine Intravaginal Device (DIB® ) with 1,0g of P4 was implanted, associated with intramuscular administration of 2,0mg of estradiol benzoate (IM); on day 8 DIB was removed; and 1,0mg of estradiol cypionate, 0,15mg of prostaglandin F2? and 300 UI of equine chorionic gonadotropin were administered; on day 10, fixed-time artificial insemination was conducted (FTAI). The cows were randomized into G1 (n=12) – without rbST / with used Bovine Intravaginal Device, G2 (n=50) – without rbST / with new DIB, G3 (n=11) - with rbST / with used DIB and G4 (n=50) – with rbST/ with new DIB. rbST was introduced on the eighth day of the protocol. Sixty days after TAI, pregnancy diagnoses were conducted, via rectal palpation. Blood samples were taken on day 0, 8 and 10 of the protocol, in order to assess P4 plasma concentrations. Pregnancy rates were statistically evaluated through Generalized Linear Models Theory and their significance was tested with Analysis of Deviance. Pregnancy rates were 58%, 40%, 81% and 48% for G1, G2, G3 and G4, respectively, with significant statistical difference for G3. Plasma concentrations of P4 were not statistically different among groups, or collections. In view of the results obtained, we concluded that the administration of rbST in association with P4 DIB, used for the third time, improves pregnancy rates. Estrus resynchronization and re-insemination positively impacted pregnancy rates.

AbstractThe objective was to evaluate different oestrous synchronization schemes in beef cattle under range conditions. In experiment 1, 202 heifers averaging 26 months of age were assigned randomly to three treatments: (a) GnRH-PG. (no. = 44) day 0 (D0): injection of GnRH; D7: injection of PGF2α; D0 to D25 oestrous detection (OD) and artificial insemination (AI); (b) OD-PG. (no. = 45) D0 to D4; OD + AI; D5: injection of PGF2α; D5 to D25 OD + AI; and (c)Ovsynch. (no. = 113) D0: injection of GnRH; D7: injection of PGF2α; D9: injection of GnRH and 16 h later AI at fixed time. In experiment 2, 318 non-suckling cows were assigned randomly to three treatments: (a) GnRH-PG. (no. = 106) D0: injection of GnRH; D7: injection of PGF2α; D0 to D25 OD + AI; (b) GnRH-MAP-PG. (no. = 106) D0: injection of a GnRH analogue and insertion of an intravaginal sponge impregnated with 250 mg of medroxyprogesterone acetate; D7: injection of PGF2α and sponge withdrawal; D0 to D25 OD + AI; (c) controls. (no. = 106) OD and AI. The experimental period lasted 25 days and, with exception of the Ovsynch treatment, oestrous detection was carried out twice a day (a.m./p.m.) and inseminations performed 12 h later. The pregnancy rates assessed by ultrasonography 30 days after AI were: experiment 1: (a) 30·0%; (b) 28·6% and (c) 62·3% (P < 0·05); experiment 2: (a) 60·2%; (b) 57·8% and (c) 45·5% (P < 0·05). It is concluded that in heifers and non-suckling cows, oestrous synchronization treatments result in higher pregnancy rates. In non-suckling cows, the addition of a progestagen did not improve the response.
