The temporal structure and dynamics of biological rhythms were formed in the course of evolution under the influence of environmental factors. Circadian rhythm as a central one in biological objects developed in response to daily luminosity and temperature rhythms related to rotation of the Earth. New causative factors that could be involved in this process and affect the entire spectrum of biological rhythms emerged with the advent of space research. The discovery of solar wind, interplanetary magnetic field, and Earth’s magnetosphere revealed similar periodicity of biological rhythms and magnetic factors which suggests possible participation of the latter in the formation of the former. Disturbances in magnetic rhythms may lead to desynchronization of biological processes by the adaptive stress mechanism as exemplified by circadian rhythm disorders in response to jetlag. This hypothesis forwarded by the authors in the 1990s was confirmed by further investigations including those reported by foreign researchers.

MicroRNAs (miRNAs) interact with 3′untranslated region (UTR) elements of target genes to regulate mRNA stability or translation, and play a crucial role in regulating many different biological processes. bantam, a conserved miRNA, is involved in several functions, such as regulating Drosophila growth and circadian rhythm. Recently, it has been discovered that bantam plays a crucial role in the core circadian pacemaker. In this paper, based on experimental observations, a detailed dynamical model of bantam-regulated circadian clock system is developed to show the post-transcriptional behaviors in the modulation of Drosophila circadian rhythm, in which the regulation of bantam is incorporated into a classical model. The dynamical behaviors of the model are consistent with the experimental observations, which shows that bantam is an important regulator of Drosophila circadian rhythm. The sensitivity analysis of parameters demonstrates that with the regulation of bantam the system is more sensitive to perturbations, indicating that bantam regulation makes it easier for the organism to modulate its period against the environmental perturbations. The effectiveness in rescuing locomotor activity rhythms of mutated flies shows that bantam is necessary for strong and sustained rhythms. In addition, the biological mechanisms of bantam regulation are analyzed, which may help us more clearly understand Drosophila circadian rhythm regulated by other miRNAs.

The article presents data from the Russian and foreign literature of the properties of melatonin, its role in the vital activity of the body and the regulation of circadian rhythms. The features of using the drug in desynchronosis, the optimal dosage and time of administration, as well as the possibility of its use in combination with non-pharmacological treatment methods are discussed. These methods include dosed light exposure and exercise. Additionally, new studies are considered, indicating the beneficial effect of melatonin and its agonists on the immune, cardiovascular, endocrine and nervous systems. Based on the collected data, it is asserted that melatonin can significantly accelerate the process of adaptation to a new time zone without the development of side effects. Therefore, this drug is the most effective and safe treatment for desynchronosis. It is also important to note that the study of the multifaceted effects of melatonin on body functions opens up great prospects in the treatment of many diseases.

A novel group of medications, dual orexin receptor antagonists, emerged as a competent group that challenges current first-line hypnotics. They have relatively infrequent and mostly well-tolerated side effects, primarily in the form of residual somnolence, fatigue and nightmares/disturbing dreams. The advantage over conventional hypnotics stems from the specifics of their target receptors, which translates into lack of tolerance after long term use and good safety profile. They are particularly favoured in some specific populations, including the elderly. Ongoing and future studies are set to explore their effect on selected conditions, such as addiction and psychiatric disorders, dementias, perimenopausal condition and circadian rhythm disorders, to name a few.

Hepatocellular carcinoma (HCC) is among the most common and lethal form of cancer worldwide. However, its diagnosis and treatment are still dissatisfactory, due to limitations in the understanding of its pathogenic mechanism. Therefore, it is important to elucidate the molecular mechanisms and identify novel therapeutic targets for HCC. Circadian rhythm-related genes control a variety of biological processes. These genes play pivotal roles in the initiation and progression of HCC and are potential diagnostic markers and therapeutic targets. This review gives an update on the research progress of circadian rhythms, their effects on the initiation, progression, and prognosis of HCC, in a bid to provide new insights for the research and treatment of HCC.
