Evaluation of Inorganic Phosphate Solubilizing Efficiency and Multiple Plant Growth Promoting Properties of Endophytic Bacteria Isolated From Root Nodules Erythrina Brucei

Abstract Background: The majority of phosphorous in the soil is fixed and unavailable to plant nutrition, hence in scarcity. Phosphate solubilizing bacteria, the ecological engineers, are considered as the best, sustainable and eco-friendly options. The objectives of this study were to screen and evaluate inorganic phosphate solubilizing efficiency and assess multiple plant growth promoting traits of E. brucei root nodule bacterial endophytes.Results: A total of 304 nodule bacterial endophytes were screened for phosphate solubilization potential on solid PA medium among which 119 (39%) were potential tricalcium phosphate solubilizers. None of these isolates were able to form clearly visible halos on aluminum phosphate (AlPO4), Al-P or iron phosphate (FePO4), Fe-P supplemented PA medium. Out of 119 inorganic phosphate solubilizing endophytes, 40.3% were IAA producers. Based on phosphate solubilization index, the potential bacterial endophytes were identified to Gluconobacter cerinus, Acinetobacter soli, Achromobacter xylosoxidans and Bacillus thuringiensis using the 16S rRNA gene sequences analysis. All the selected isolates were potential solubilizers of the three inorganic phosphates (Al-P, Fe-P and tricalcium phosphate, TCP) included in liquid NBRIP medium. The highest values of solubilized TCP were recorded by isolates AU4 and RG6 (A. soli), 108.96 mg L-1 and 107.48 mg L-1, respectively at sampling day3 and 120.36 mg L-1 and 112.82 mg L-1, respectively at day 6. The highest values of solubilized Al-P and Fe-P were recorded by isolate RG6, 102.14 mg L-1 and 96.07 mg L-1, respectively at sampling days 3 and 6, respectively. The highest IAA, 313.61µg mL-1 was recorded by isolate DM17 (B. thuringiensis). These selected potential isolates were also HCN, NH3, and hydrolytic enzymes producers. The isolates were also varied in tolerance to eco-physiological stressors and exhibited versatility to carbon and nitrogen substrate utilization. Conclusions: The genera and species Gluconobacter cerinus, Acinetobacter soli, Achromobacter xylosoxidans and Bacillus thuringiensis are the first reports from E .brucei root nodules and Gluconobacter is also the first report to the science as phosphate solubilizer. Isolates AU4 and RG (A. soli) could be potential bio-inoculant candidates for the growth enhancement of the host plant for better agro-forestry practices in acidic and alkaline soils in Ethiopia.

Pseudomonas argentinensis strain SA190 is a plant endophytic-inhabiting bacterium that was isolated from root nodules of the desert plant Indigofera argentea collected from the Jizan region of Saudi Arabia. Here, we report the genome sequence of SA190, highlighting several functional genes related to plant growth–promoting activity, environment adaption, and antifungal activity.

Background: Phosphorus has been considered as the key element as it is directly involved in most of the life processes including in plants. Soil contains both organic and inorganic forms of phosphorus. Phosphate solubilization is the important property of soil bacteria to develop them as plant growth promoting bacteria in the agricultural field. The present study report the phosphate solubilizing and plant growth promoting properties of selected actinobacteria isolated from rhizosphere soil.Methods: Actinobacterial strains were isolated from rhizosphere soil and screened for in vitro phosphate solubilizing properties using agar plate method. The efficiency of phosphate solubilization and phosphatase activity of isolated actinobacterial strains were tested using Pikovskaya broth. Result: In the present study phosphate solubilization and phosphatase activity of isolated actinobacteria, Streptomyces sp. CTD-2 was comparatively higher in lab conditions. In pot trial experiment strain CTD-2 showed higher growth when compared to the control plant. The chlorophyll content of leaves in the experimental plants were found maximum with actinobacteria strain CTD-2 treated plant. Plant growth measurements such as root length, shoot length, leaf length, total plant growth measurements, quick yield production were also observed.

Azotobacter is a plant growth promoting bacteria which solubilize phosphorous and make it available for plants. In the present study the efforts have been taken to fulfill the aim to isolate, characterize and screen Azotobacter for phosphate solubilization from the rice rhizosphere of Durg district. Total 8 strains of Azotobacter were isolated from different rice fields. It was observed that all the strains were found to be phosphate solubilizing but in different quantity. RAZB6 is highest phosphate solubilizer (234.4±1mg/L) and RAZB4 shows minimum (82.09±1mg/L) quantity of phosphate solubilization. Since phosphorous is necessary for the plants proper development and it increases the growth of rice plant and make soil more fertile for other plants growth also so they must be commercialized and used more and more in the crop fields other than the agrochemicals.

AbstractPrevious studies have shown the sugarcane microbiome harbors diverse plant growth promoting microorganisms, including nitrogen-fixing bacteria (diazotrophs), which can serve as biofertilizers. The genomes of 22 diazotrophs from Colombian sugarcane fields were sequenced to investigate potential biofertilizers. A genome-enabled computational phenotyping approach was developed to prioritize sugarcane associated diazotrophs according to their potential as biofertilizers. This method selects isolates that have potential for nitrogen fixation and other plant growth promoting (PGP) phenotypes while showing low risk for virulence and antibiotic resistance. Intact nitrogenase (nif) genes and operons were found in 18 of the isolates. Isolates also encode phosphate solubilization and siderophore production operons, and other PGP genes. The majority of sugarcane isolates showed uniformly low predicted virulence and antibiotic resistance compared to clinical isolates. Six strains with the highest overall genotype scores were experimentally evaluated for nitrogen fixation, phosphate solubilization, and the production of siderophores, gibberellic acid, and indole acetic acid. Results from the biochemical assays were consistent and validated computational phenotype predictions. A genotypic and phenotypic threshold was observed that separated strains by their potential for PGP versus predicted pathogenicity. Our results indicate that computational phenotyping is a promising tool for the assessment of bacteria detected in agricultural ecosystems.

Microbial treatment has recently been attracting attention as a sustainable agricultural strategy addressing the current problems caused by unreasonable agricultural practices. However, the mechanism through which microbial inoculants promote plant growth is not well understood. In this study, two phosphate-solubilizing bacteria (PSB) were screened, and their growth-promoting abilities were explored. At day 7 (D7), the lengths of the root and sprout with three microbial treatments, M16, M44, and the combination of M16 and M44 (Com), were significantly greater than those with the non-microbial control, with mean values of 9.08 and 4.73, 7.15 and 4.83, and 13.98 and 5.68 cm, respectively. At day 14 (D14), M16, M44, and Com significantly increased not only the length of the root and sprout but also the underground and aboveground biomass. Differential metabolites were identified, and various amino acids, amino acid derivatives, and other plant growth-regulating molecules were significantly enhanced by the three microbial treatments. The profiling of key metabolites associated with plant growth in different microbial treatments showed consistent results with their performances in the germination experiment, which revealed the metabolic mechanism of plant growth-promoting processes mediated by screened PSB. This study provides a theoretical basis for the application of PSB in sustainable agriculture.
