Phytoplankton blooms are known to influence barnacle recruitment and in boreal regions spring blooms work as an important trigger. Close to the west coast of the sub-continent of India, blooms tend to be triggered by breaks in the monsoon and the recurrence of the monsoon after a short break can stress the new recruits. The recruitment of Balanus amphitrite, an acorn barnacle, at Dona Paula Bay at the mouth of Zuari estuary, Goa, India was studied. Observations included variations in recruitment, larval abundance, development and reproduction. Adult conditioning and inter-brood variations were important factors in the larval ecology of this organism. The results indicate that the impulsive release of larvae during breaks between monsoons could be a short-sighted luxury for Balanus amphitrite in these waters. Temporal variations or recruitment failure in such environments can be attributed to inappropriate cue synchronization.

Transcriptomic information provides fundamental insights into biological processes and can be used to determine gene expression in cell, tissue, or organism under specific physiological conditions, or in response to any environmental perturbation. Extraction of high quality RNA is a challenging step mainly in non-traditional organisms, and protocols for preservation and isolation need to be adjusted in many cases. In the present work, we aimed to develop a protocol for preservation and isolation of high-quality and quantity of RNA from blood and liver tissues of Caiman latirostris. Three preservation methods were tested: 1) flash freezing (LN2) and storage at –80°C; 2) RNAlater® conservation with progressive cooling up to –80°C); 3) preservation in TRIzol® reagent, flash freezing in LN2 and storage at –80°C. Methods 1 and 2 were tested for liver, while 2 and 3 for blood. Our results showed that both preservation methods resulted in excellent outcomes for liver samples. For blood samples however, TRIzol® preservation was an efficient procedure for adequate RNA quality, quantity, and integrity, while conservation in RNAlater® solution was inadequate in both quality and quantity for an optimal RNA extraction. Appropriate protocols were established for each tissue and are being used now for transcriptomic studies in this sentinel organism.

Green building design has emerged as a global force, with one green building standard reporting more than 3.5 billion square feet certified worldwide. Green buildings focus on reducing environmental impact through improved water storage, reducing environmental perturbation, and reducing energy usage. Although the environmental benefits of green building design are now well established, it is only more recently that the field has come to appreciate the health benefits of green building design. This chapter discusses the green building movement and the challenges and opportunities it represents, with lessons that can be learned and are generalizable to urban health scholarship and action worldwide.
