UVC or ionizing radiation of mammalian cells elicits a complex genetic response that allows recovery and cell survival. Kin17 gene, which is highly conserved among mammals, is upregulated during this response. Kin17 gene encodes a 45 kDa protein which binds to DNA and presents a limited similarity with a functional domain of the bacterial RecA protein. Kin17 protein is accumulated in the nucleus of proliferating fibroblasts and forms intranuclear foci. Using expression vectors, we show that overexpression of kin17 protein inhibits cell-cycle progression into S phase. Our results indicate that growth inhibition correlates with disruption of the nuclear morphology which seems to modify the intranuclear network required during the early steps of DNA replication. We report that a mutant encoding a protein deleted from the central domain of kin17 protein enhanced these effects whereas the deletion of the C-terminal domain considerably reduced them. These mutants will be used to elucidate the molecular mechanism by which kin17 protein alters cell growth and DNA replication.

In Ethiopia,there are 32.85 millions of sheep,more than 99 % of which are indigenous.However,the productivity of local sheep under traditional production system is low with high mortality of sheep.There are two ways of improving performance of sheep and goats,namely improving the enviroment of animals and/or improving there genetic potential.The aim of this study was to predict genetic gains of breedingobjective traits and select the best sheep selection scheme for Gumuz andWashera sheep. Body size(six month weight and yearling weight) and litter size were breeding objective traits identified by own flock animal ranking experiment and personal interview. Deterministic approach of ZPLAN computor program is used for modeling input parametres of Gumuz and Washera sheep and simulating breeding plans using gene flow method and selection index procedures. One-tier cooperative sheep breeding scheme were proposed whereby ram exchange between and within villages is the main means of genetic dissimination. Genetic gains predicted for six month weight of Gumuz and Washera sheep were 0.43 and 0.55 kg,respectively. Genetic gains predicted for yearling weight of Gumuz and Washera sheep were 0.55 and 0.60 kg,respectively. Genetic gains predicted for litter  size of Gumuz and Washera sheep were 0.08 and 0.09 lambs,respectively. The lower rate of inbreeding, the higher monetary genetic gain for aggregate genotype,higher return to investmnet and higher profit/ewe/year were quality measures of breeding program considered to prefer scheme 4 for both Gumuz and Washera sheep.Hence,for both Gumuz and Washera sheep populations a sheep selection scheme designed with 15 % selection proportion and one year ram use for breeding was recommended. Special emphasis need to be given to yearling weight with higher predicted genetic response and higher percentage return to investment.

ABSTRACT In Pseudomonas aeruginosa, the narK 1 K 2 GHJI operon encodes two nitrate/nitrite transporters and the dissimilatory nitrate reductase. The narK 1 promoter is anaerobically induced in the presence of nitrate by the dual activity of the oxygen regulator Anr and the N-oxide regulator Dnr in cooperation with the nitrate-responsive two-component regulatory system NarXL. The DNA bending protein IHF is essential for this process. Similarly, narXL gene transcription is enhanced under anaerobic conditions by Anr and Dnr. Furthermore, Anr and NarXL induce expression of the N-oxide regulator gene dnr. Finally, NarXL in cooperation with Dnr is required for anaerobic nitrite reductase regulatory gene nirQ transcription. A cascade regulatory model for the fine-tuned genetic response of P. aeruginosa to anaerobic growth conditions in the presence of nitrate was deduced.

We describe the isolation and characterization of more than 60 novel cDNA clones that constitute part of the immediate genetic response to resting human peripheral blood T cells after mitogen activation. This primary response was highly complex, both in the absolute number of inducible genes and in the diversity of regulation. Although most of the genes expressed in activated T cells were shared with the activation response of normal human fibroblasts, a significant number were more restricted in tissue specificity and thus likely encode or effect the differentiated functions of activated T cells. The activatable genes could be further differentiated on the basis of kinetics of induction, response to cycloheximide, and sensitivity to the immunosuppressive drug cyclosporin A. It is of note that cyclosporin A inhibited the expression of more than 10 inducible genes, which suggests that this drug has a broad genetic mechanism of action.

Ratios of the phenotypic values of two traits may be used as selection criteria in animal and plant breeding to improve the ratio traits themselves or to effect changes in their two component (numerator and denominator) traits. Prediction of genetic responses to ratio-based selection would facilitate quantitative analysis and evaluation of selection based on ratios. Methods for predicting such responses are derived and presented here. They employ expressions for the truncation value of a ratio and for the phenotypic selection differentials of the numerator and denominator traits. The derivation of these expressions is based upon the assumption that the phenotypic values of each of these traits are normally distributed. Worked examples relating to livestock and crop improvement are included to demonstrate how responses to selection for ratios may be predicted.Key words: ratio selection criterion, selection differential, genetic response.
